Abstract
一种将式Ⅰ化合物在化妆品或药剂中用作UV过滤物质,尤其是作为UV-A过滤物质,用来保护人体皮肤或人体毛发免受太阳辐射的方法,它可以被单独使用或与化妆品和药物制剂的现有技术中已知的化合物结合使用,它能够吸收UV范围的辐射,其中C=C双键以E和/或Z构型存在。
Description
本发明涉及烯胺衍生物作为光稳定紫外线(UV)过滤物质在化妆品和药物制剂中的应用,采用这种化妆品和药物制剂可保护人体表皮或毛发免受UV辐射侵害,特别是320-400nm波长的辐射。
在化妆品和药物制剂中使用遮光剂(screening agent)的目的是防止阳光对人体皮肤构成伤害,或至少可以降低这种伤害的影响。另一方面,这些遮光剂还可以保护其它成分不被UV辐射破坏或不因UV辐射降解。在护发剂中需要用它来减少UV辐射对角蛋白纤维的损害。
照到地球表面的阳光包括UV-B辐射(280-320nm)和UV-A辐射(>320nm),它们直接跟在可见光范围之后。晒斑就足以说明它对人体皮肤的作用,尤其是UV-B辐射的作用。因此,市场上提供了相当多数量的吸收UV-B辐射的物质,它们可用来防止晒斑。
皮肤学研究表明UV-A辐射也会造成皮肤损害和例如,因角蛋白或弹性蛋白受伤而引起的过敏。这意味着皮肤的弹性和持水能力降低,即皮肤缺乏柔软性以致将形成皱纹。在强日晒地区皮肤癌发病率明显高的事实清楚地表明细胞中遗传因子的损害是由阳光引起的,尤其是UV-A辐射。知道这些后也就清楚开发有效UV-A辐射过滤物质的必要了。
因此,人们对主要作为UV-A过滤物质的化妆品和药物制剂中的遮光剂的需求有所增长,而且其最大吸收波长应该在约320-380nm范围。为了用最少量的原料达到所要求的作用,遮光剂还应具有高度的比吸收。
而且,遮光剂要用于制备化妆品还必须满足许多其它要求,例如,在化妆油中具有好的溶解生,由其制备的乳液具有高稳定性,能够让人接受的毒性,以及本身没有什么气味和颜色。
遮光剂所要满足的另一个要求是适当的光稳定性。但是,迄今为止,还没有哪种吸收UV-A辐射的遮光剂能够或仅仅不充分地满足这一点。
在法国专利申请2,440,933中4-(1,1-二甲基乙基)-4-甲氧基-二苯甲酰基甲烷被描述为UV-A过滤物质。该文中建议这种由GIVAUDAN以“PARSOL 1789商标名出售的具体UV-A过滤物质可与UV-B过滤物质结合使用以吸收所有波长在280-380nm范围内的UV辐射。
但是,当单独使用这种UV-A过滤物质或与UV-B过滤物质结合使用时,它对长时间日光浴下保证对皮肤恒定保护不具备充分的光化学稳定性,因此,如果要求它对整个范围的UV辐射达到有效的皮肤防护就必须相隔一定的短时间间隔后重新将其用于皮肤。
因此,EP 0,514,491建议添加本身就对UV-B范围辐射起过滤作用的2-氰基-3,3-二苯基丙烯酸酯来使不够光稳定的UV-A过滤物质稳定。
而且,EP 251,398也建议将具有吸收UV-A和UV-B辐射能力的发色团通过连接链与单个分子结合。但这样做的缺点是在化妆品中将UV-A过滤物质与UV-B过滤物质自由结合是不可能的,而且在化学连接发色团时遇到的困难只允许用于某些结合。
因此,本发明一个目的是为化妆品和药物制备提供能够高度吸收UV-A辐射并且光稳定的遮光剂,而且本身没有什么颜色,即表现出已经确定的键结构并根据所用替代物能溶解于油或水。
本发明这个目的可以通过使用式I化合物实现,其中C=C双键以E和/或Z构型存在,其它变量具有下列含义:R1指COOR5,COR5,CONR5R6,CN,O=S(-R5)=O,O=S(-OR5)=O,R7O-P(-OR8)=O;R2指COOR6,COR6,CONR5R6,CN,O=S(-R6)=O,O=S(-OR6)=O,R7O-P(-OR8)=O;R3指氢,至多有18个碳原子的任意取代的脂肪族,环脂肪族,芳脂肪族或芳香族基团;R4指含有5-12个环原子的任意取代的芳香族或杂芳香族基团;R5-R8独立地指氢,至多有18个碳原子的开链或支链脂肪族,芳脂肪族,环脂肪族或任意取代的芳香族基团,其中变量R3-R8也可以与它们相连的碳原子一起形成一个5-或6-元环,如果需要,它们可以是稠合的,即在化妆品或药物制剂中用式I化合物作为UV过滤物质,尤其是UV-A过滤物质,以保护人体皮肤或人体毛发免受太阳辐射,它可以被单独使用或与化妆品和药物制剂的现有技术中已知的化合物结合使用,它能够吸收UV范围的辐射。
优选的式I化合物是其中R3代表氢,R1代表CN,COOR5和COR5以及R2代表CN,COOR6和COR6的化合物,其中R5和R6独立地指至多有8个碳原子的开链或支链脂肪族或任意取代的芳香族基团。
特别优选的式I化合物是其中R3代表氢,R1代表CN,COOR5和COR5,R2代表CN,COOR6和COR6的化合物,其中R5和R6独立地指至多有8个碳原子的开链或支链脂肪族或任意取代的芳香族基团,以及R4代表含有至多10个环碳原子的任意取代的芳香族或杂芳香族基团,尤其是取代的苯基,噻吩基,呋喃基,吡啶基,吲哚基或亚萘基,更优选任意取代的苯基或噻吩基。
合适的取代基包括含有,例如,多至20个碳原子的亲油和亲水取代基。亲油基团,即改善式I化合物油溶解性的基团,是,例如,脂肪族或环脂肪族基团,特别是含有1-18个碳原子的烷基,烷氧基,一烷基氨基和二烷基氨基,烷氧羰基,一烷基-和二烷基-氨基羰基,一烷基-和二烷基-氨基磺酰基,氰基(cyanogeno),硝基,溴,氯,碘或氟取代基。
亲水基团,即使式I化合物溶于水的基团,例如有羧基和次硫酸基,特别是它们与生理上可接受的阳离子结合形成的盐,例如碱金属盐或三烷基铵盐如三(羟基烷基)铵盐或2-甲基-丙-1-醇-2-铵盐。其它合适的基团有含有任何生理上可接受的阴离子的铵,特别是烷基铵基团。
合适的烷氧基是含有1-12个碳原子,尤其是含有1-8个碳原子的烷氧基,其实例包括:甲氧基,乙氧基,异丙氧基,正丙氧基,1-甲基丙氧基,正丁氧基,正戊氧基,2-甲基丙氧基,3-甲基丁氧基,1,1-二甲基丙氧基,2,2-二甲基丙氧基,己氧基,1-甲基-1-乙基丙氧基,庚氧基,辛氧基,2-乙基己氧基。
合适的一烷基氨基或二烷基氨基,例如,为所含烷基具有1-8个碳原子的基团,其中烷基例如是甲基,正丙基,正丁基,2-甲基丙基,1,1-二甲基丙基,己基,庚基,2-乙基己基,异丙基,1-甲基丙基,正戊基,3-甲基丁基,2,2-二甲基丙基,1-甲基-1-乙基丙基和辛基。这些基团也可以一烷基-和二烷基-氨基羰基和相应的磺酰基形式存在。
烷氧基羰基例如是含有上述烷氧基或高级醇,例如,含有多至20个碳原子的醇如异-C15-醇的酯。

含有遮光剂的化妆品和药剂通常以一个至少含有一种油相物质的载体为基础。或者,当使用含有亲水取代基的化合物时制剂只可能用水作为基础。因此,油,水包油型乳液,油包水型乳液,乳膏和油膏,保护性唇膏组合物或无动物脂凝胶都是合适的剂型。
因此,这些防晒制剂都可以液体,糊状或固体形式出现,如油包水型乳膏,水包油型乳膏和洗液,溶胶发泡乳膏,凝胶,油,动物脂粘胶,粉末,喷雾液或醇的水洗液。

其它能够吸收UV-A范围辐射的已知物质也可以添加使用,条件是它们在将根据本发明结合使用UV-B和UV-A过滤物质的整个系统中是稳定的。
本发明还涉及含有占化妆品和药物制剂总重量0.1-10wt%,优选1-7wt%的一种或多种式I化合物和已知适于在化妆品和药物制剂中作为UV遮光剂的化合物的化妆品和药物制剂,式I化合物的用量通常比吸收UV-B的化合物的用量少。
用来保护人体表皮的化妆品和药物制剂中的遮光剂主要由吸收UV-B范围的UV光,即280-320nm范围辐射的化合物构成。例如,用于本发明的UV-A吸收剂含量占整个UV-B和UV-A过滤物质的10-90wt%,优选20-50wt%。

最后,微粒体积(5微米以下)颜料如二氧化钛和氧化锌也可以使用。
为了保护人体毛发免受UV辐射本发明式I的这些遮光剂可以被混入香波,洗液,凝胶或乳液中,浓度为0.1-10wt%,优选1-7wt%。所得各种制剂,特别是可用于洗涤,染色和成形头发。
本发明化合物通常对UV-A辐射都有特别高的吸收能力,并表现出清晰的带结构。另外,它们很容易溶解于化妆油中,并且很容易与化妆品制剂共同使用。用化合物I形成的乳液的显著特征是它们具有很高的稳定性,而且化合物I较高的光稳定性尤为突出,用化合物I制成的制剂在皮肤上的感觉也很好。
本发明还涉及用作医疗和药物制剂的式I化合物,用来预防皮肤感染和过敏,以及用来防止某些皮肤癌的发病,所说制剂至少含有一种有效量的作为活性物质的式I化合物。
本发明药物制剂可以口服或局部给药。对于口服给药,这些药剂可以是,尤其是锭剂,明胶胶囊,糖锭剂,糖浆,溶液,乳液或悬浮液形式。对于这些药剂的局部给药是采用它们的油膏,乳膏,凝胶,喷雾剂,溶液或洗液形式。
实施例I.制备实施例1一般性说明(针对表2的化合物1)
将0.1mol对氨基苯甲酸2-乙基己酯,0.1mol新戊酰乙腈和0.1mol原甲酸三乙酯在100mL二甘醇中于120℃加热2h,经过蒸馏除去乙醇。冷却到80℃后加水,并滤除沉淀。母液从石油醚重结晶,得到表2中的化合物1,产率为80%。实施例2
将0.1mol氨茴酸2-乙基己酯,0.1mol 2,4-戊二酮和0.1mol原甲酸三乙酯在100mL二甘醇中于120℃加热2h,经过蒸馏除去乙醇。冷却到80℃后加水,并滤除沉淀。母液从石油醚重结晶,得到表2中的化合物24,产率为70%。实施例3
将0.1mol间甲苯胺,0.1mol新戊酰基乙腈,0.1mol原甲酸三乙酯和1g氯化锌在100mL二甘醇中于120℃加热2h,经过蒸馏除去乙醇。冷却到80℃后加水,并滤除沉淀。母液从石油醚重结晶,得到表2中的化合物2,产率为70%。

Claims (13)
1.一种将式I化合物在化妆品或药剂中用作UV过滤物质,尤其是作为UV-A过滤物质,用来保护人体皮肤或人体毛发免受太阳辐射的方法,它可以被单独使用或与化妆品和药物制剂的现有技术中已知的化合物结合使用,它能够吸收UV范围的辐射,其中C=C双键以E和/或Z构型存在,其它变量具有下列含义:R1指COOR5,COR5,CONR5R6,CN,O=S(-R5)=O,O=S(-OR5)=O,R7O-P(-OR8)=O;R2指COOR6,COR6,CONR5R6,CN,O=S(-R6)=O,O=S(-OR6)=O,R7O-P(-OR8)=O;R3指氢,至多有18个碳原子的任意取代的脂肪族,环脂肪族,芳脂肪族或芳香族基团;R4指含有5-12个环原子的任意取代的芳香族或杂芳香族基团;R5-R8独立地指氢,至多有18个碳原子的开链或支链脂肪族,芳脂肪族,环脂肪族或任意取代的芳香族基团,其中变量R3-R8也可以与它们相连的碳原子一起形成一个5-或6-元环,如果需要,它们可以是稠合的。
2.权利要求1所述式I化合物作为UV-A过滤物质的使用方法。
3.如权利要求1所述式I化合物的使用方法,其中R3代表氢,R1代表CN,COOR5和COR5以及R2代表CN,COOR6和COR6的化合物,其中R5和R6独立地指至多有8个碳原子的开链或支链脂肪族或任意取代的芳香族基团。
4.如权利要求1所述式I化合物的使用方法,其中R4代表任意被亲水或亲油取代基取代的苯基。
5.如权利要求1所述式I化合物的使用方法,其中R4代表烷氧基苯基或烷氧羰基苯基。
6.如权利要求1所述式I化合物的使用方法,其中R4代表载有选自羧酸盐,磺酸盐或铵基的水溶性取代基的苯基。
7.保护人体表皮或人体毛发免受280-400nm范围UV光线损害并含有遮光剂的化妆品或药物制剂,其中所说制剂含有有效量的式I化合物,作为光稳定UV过滤物质,在化妆品或药剂载体上单独或与能够吸收UV辐射并已知适于化妆品和药物制剂的化合物一起使用其中所有变量具有权利要求1中的定义。
8.权利要求7定义的遮光剂,含有作为UV-A过滤物质的式I化合物,其中R3代表氢,R1代表CN,COOR5和COR5以及R2代表CN,COOR6和COR6的化合物,其中R5和R6指至多有8个碳原子的任意取代的脂肪族或芳香族基团。
9.权利要求7定义的遮光剂,含有作为UV-A过滤物质的式I化合物,其中R4代表任意被亲水或亲油取代基取代的苯基。
10.权利要求7定义的遮光剂,含有作为UV-A过滤物质的式I化合物,其中R3代表氢,R1代表CN,COOR5和COR5以及R2代表CN,COOR6和COR6的化合物,以及R4代表可以被烷基,烷氧基,烷氨基羰基,烷氧羰基取代的苯基,上述各种情况至多含有20个碳原子,或者含有氰基或羧基,也可以含有下列水溶性取代基羧酸盐,磺酸盐或铵基。
11.式II所示的新化合物,其中C=C双键以E和/或Z构型存在;X指氰基或COCH3;R4指苯基,当X是CN时,它可以被一个或多个至多含有20个碳原子的烷基,烷氧基,烷基氨基羰基或烷氧羰基,或者氰基或羧基取代,也可以被下列水溶性取代基取代:羧酸盐,磺酸盐或铵基,例如碱金属羧酸盐或羰氧基三(羟乙基)铵基或磺酰氧基三(羟乙基)铵基,或者,当X是COCH3时,它可以被一个或多个至多含有20个碳原子的烷氧基,或含有4-20个碳原子的烷氧羰基取代,也可以被下列水溶性取代基取代:羧酸盐,磺酸盐或铵基;当X是CN时R5指C(CH3)3基团,或当X是COCH3时,R5指至多含有18个碳原子的开链或支链或环形烷基,烷氧基或烷氧基烷基,或R5指芳氧基。
12.用作药物的式I化合物。
13.含有有效量的至少一种权利要求1所述的式I化合物的药物制剂。
