Description
本願は、米国特許法第119条(e)項に基づき、2012年9月26日出願の米国仮出願第61/705,731号の出願日の利益を主張し、その内容全体が参照により本明細書に組み込まれる。
下に与えるプロセスパラメーターは、当業者の特定の必要性に合うように当業者が適応させ採用することができる。
UV検出器又は同等機器を備えた水(Water)HPLCシステム
カラム:YMC−Pack ODS−AQ 250×4.6mm;5μm(品番=AQ12S05−2546WT)

移動相:A=水1リッター中85%H3PO41ml;B=アセトニトリル/テトラヒドロフラン 30:70(v/v)(注:LC/MS分析で移動相A中に、H3PO4の代わりに0.1%TFAを用いた)

グラジエント
所与の混合物のアリコート(20μL)をオートサンプラーバイアル中アセトニトリル(1.5mL)に加えた。バイアルにキャップをし、振盪し、次いで15℃のオートサンプラー中に置いた。各HPLCランごとに注入量(15μL)を分析した。
マイタンシノール(50.1mg、0.0888mmol)、(4S)−3,4−ジメチル−2,5−オキサゾリジンジオン(30.2mg、0.233mmol、2.6当量)、亜鉛トリフレート(133mg、0.366mol)及び、真空下250℃で予備乾燥し、次いで室温で冷却した4Aモレキュラーシーブ(0.50g)を、10mlフラスコに加えた。内容物を無水ジメチルホルムアミド(0.75mL)中に取り、ジイソプロピルエチルアミン(62μL、0.357mmol)を加えた。混合物を室温で24時間撹拌した。粗混合物の資料をHPLCによって分析したところ、N2’−デアセチル−マイタンシン生成物が総HPLC面積の80%を占めた。反応混合物を、1:1の飽和NaHCO3:飽和NaCl(1.2mL)と酢酸エチル(3mL)で希釈し、混合し、次いでセライトでろ過し、次いでリン酸カリウム緩衝液(1mL、400mM、pH7.5)で洗った。有機層を無水硫酸マグネシウムで乾燥し、ろ過し、次いで蒸発させると、黄色い固体が形成された。この固体に、3−メチルジチオプロパン酸(25mg、0.16mmol)、塩酸N−(3−ジメチルアミノプロピル)−N’−エチルカルボジイミド(30mg、0.16mmol)、及びジクロロメタン(3mL)を加えた。2時間撹拌した後、混合物を酢酸エチル(8mL)で希釈し、1.0M pH6.5リン酸カリウム緩衝液(2mL)で洗い、水溶液を酢酸エチル(2×8mL)で抽出した。有機層を合わせ、無水硫酸マグネシウム上で乾燥し、濃縮し、シリカクロマトグラフィー、95:5のジクロロメタン:メタノールで精製すると、51mg(70%)のDM1−SMeが得られた。
実施例1の反応を10倍大きい規模で行い、490mg(68%)のDM1−SMeを得た。
25mLフラスコの無水ジメチルホルムアミド(15mL)中にマイタンシノール(1.0g、1.77mmol)を溶解させ、氷/水浴中冷却した。2分後、ジイソプロピルエチルアミン(DIPEA、0.92g、7.07mmol)及び亜鉛トリフレート(3.8g、10.6mmol)を磁気撹拌しながら加え、次いで(4S)−3,4−ジメチル−2,5−オキサゾリジンジオン(0.913g、7.07mmol)を手早く加え、混合物を24時間撹拌した。粗混合物の試料をHPLCによって分析したところ、N2’−デアセチル−マイタンシン生成物が総HPLC面積の65%を占めた。反応混合物を、1:1の飽和NaHCO3:飽和NaCl(25mL)と酢酸エチル(40mL)で希釈し、混合し、次いでセライトでろ過し、飽和NaClで洗った。有機層を無水物硫酸ナトリウムで乾燥し、ろ過し、次いで蒸発させた。残渣をジクロロメタン(30mL)中に取り、3−メチルジチオプロパン酸(1.1g、7.0mmol)及び塩酸N−(3−ジメチルアミノプロピル)−N’−エチルカルボジイミド(1.34g、7.0mmol)を手早く加え、反応物をアルゴン下室温で2時間撹拌した。混合物を酢酸エチル(30mL)で希釈し、1.0Mリン酸カリウム緩衝液(30mL)、pH6.5、で洗い、水溶液を酢酸エチル(2×40mL)で抽出した。有機層を合わせ、無水硫酸ナトリウム上で乾燥し、濃縮し、シリカクロマトグラフィー、95:5のジクロロメタン:メタノールで精製すると、698mg(50%)のDM1−SMeが得られた。
実施例3の反応を同じ規模で繰り返し、735mg(53%)のDM1−SMeを得た。
25mLフラスコの無水ジメチルホルムアミド(7mL)中にマイタンシノール(0.5g、0.89mmol)を溶解させ、氷/水浴中冷却した。2分後、ジイソプロピルエチルアミン(0.5g、3.5mmol)及び亜鉛トリフレート(1.9g、5.3mmol)を磁気撹拌しながら加え、次いで(4S)−3,4−ジメチル−2,5−オキサゾリジンジオン(4.52g、3.5mmol)を手早く加え、混合物を24時間撹拌した。この保存液のアリコート(各0.5mL)を次の実験で用いるため、各アリコートを約0.13mmolのマイタンシノールから作製した。
17:16で、HPLCパーセント面積の比率は3.0:70.9であった。
N2’−デアセチル−マイタンシン保存液(0.50mL)を6mL容量バイアルに加え、それにメタノール(75μL、1.8mmol)を加え、バイアルにキャップをし、内容物を1時間磁気撹拌した。次いで酢酸エチル(1.5mL)及び1:1の飽和NaCl:NaHCO3(0.75mL)を加え、バイアルにキャップをし、混合した。有機層を保持し、無水Na2SO4(120mg)上で乾燥した。有機層(1.0mL)を取り、プロピオン酸(20.0μL、0.27mmol)を加えた。次いで溶液を、塩酸N−(3−ジメチルアミノプロピル)−N’−エチルカルボジイミド(40mg、0.209mmol)を含有するバイアルに移した。反応を2.5時間進行させ、その後それをHPLCによって分析した。17に対するHPLCのピークはほとんど検出できず、積分はできなかった。反応を繰り返しが、再度17はほとんど検出できず、積分はできなかった。したがって、予備クエンチング法は、望ましさがより低くない化合物15及び17を生成させる。

Claims (27)
- 次の式:
a)マイタンシノールとN−カルボキシ無水物とを、塩基ならびに、亜鉛トリフレート、塩化亜鉛、臭化マグネシウム、マグネシウムトリフレート、銅トリフレート、臭化銅(II)、塩化銅(II)、及び塩化マグネシウムから選択されるルイス酸を追加して含ませた反応混合物において反応させ、それによって前記式(Ia)の化合物を形成するステップを含み、前記N−カルボキシ無水物は次の式:
b)ステップa)の反応混合物の未反応N−カルボキシ無水物を、メタノール、エタノール、n−プロパノール、イソプロパノール、及びtert−ブタノールから選択されるアルコールである求核試薬と反応させるステップを含む、
前記方法。 - 前記ルイス酸は亜鉛トリフレートである、請求項1に記載の方法。
- ステップa)の反応混合物は乾燥剤をさらに含む、ここで当該乾燥剤はモレキュラーシーブ、硫酸ナトリウム、硫酸カルシウム、塩化カルシウム、又は硫酸マグネシウムである、請求項1または2に記載の方法。

- Xはハロゲン化物、アルコキシ基、アリールオキシ基、イミダゾール、ニトロ若しくは塩化物で置換されていてもよい−S−フェニル、又は−OCORであり、式中、Rは直鎖C1〜C10アルキル基、分岐C1〜C10アルキル基、環状C3〜C10アルキル基、又はC1〜C10アルケニル基である、請求項6に記載の方法。
- COXは反応性エステルである、請求項6に記載の方法。
- R3は−Y−S−SR4であり、式中、YはC1〜C10アルキレンであり、R4はC1〜C10アルキル、アリール、又はヘテロアリール(ピリジル)である、請求項6に記載の方法。
