Description
一方で特許文献3に記載の方法では、アセトニトリル等と比べてより安価なトルエン等の芳香族炭化水素を用いているが、アミン塩を反応に必須としている。また、非特許文献1ではジクロロメタンと水との混合溶媒を用いているが、塩素系溶媒中でアジ化ナトリウムを反応させると爆発性の高いジアジドメタンが生成する危険性が高く、工業的に好ましくない。
[1] 下記一般式(II)(一般式(II)中、Mはアルカリ金属原子又はアルカリ土類金属原子を表し、mは1又は2を表す。)で表されるアジ化物と、下記一般式(III)(一般式(III)中、Yは、アルキル基、アリール基、アリールアルキル基、置換基を有するシリル基、又は置換基を有するシリルアルキル基を表し、Xはハロゲン原子を表し、pは0又は1を表し、qは0又は1を表し、R1は、qが0の場合にアルキル基を表し、qが1の場合にアルキレン基を表し、R2は、無置換の若しくは置換基を有するアリール基を表す。但し、pが0の場合、qは1である。)で表される化合物とを、水と芳香族炭化水素系溶媒と相間移動触媒とを含む混合溶媒中で反応させ、下記一般式(I)(一般式(I)中、Y、X、R1、R2、p、及びqは、一般式(II)と同様である。)で表される化合物を製造することを特徴とする、1H−テトラゾール誘導体の製造方法。

一般式(II)中、Mは、アルカリ金属原子又はアルカリ土類金属原子を表す。アルカリ金属原子としては、ナトリウム、カリウム、リチウム等が、アルカリ土類金属原子としては、マグネシウム、カルシウム等が挙げられる。これらの中でも、Mとしては、ナトリウム又はカリウムが好ましく、ナトリウムがより好ましい。
一般式(III)中、Yは、アルキル基、無置換の若しくは置換基を有するアリール基、無置換の若しくは置換基を有するアリールアルキル基、置換基を有するシリル基、又は置換基を有するシリルアルキル基を表し、Xは、ハロゲン原子を表し、pは0又は1を表し、qは0又は1を表し、R1は、qが0の場合にアルキル基を表し、qが1の場合にアルキレン基を表し、R2は、無置換の若しくは置換基を有するアリール基を表す。但し、pが0の場合、qは1である。
アルキル基としては、メチル基、エチル基、n−プロピル基、i−プロピル基、n−ブチル基、i−ブチル基、s−ブチル基、t−ブチル基、n−ペンチル基、n−ヘキシル基等が挙げられる。アルキル基を構成する炭素の数は1〜8が好ましい。

ハロアルコキシ基としては、2−クロロ−n−プロポキシ基、2,3−ジクロロブトキシ基、トリフルオロメトキシ基等が挙げられる。ハロアルコキシ基を構成する炭素の数は1〜8が好ましい。
アルキルスルホニル基としては、メチルスルホニル基、エチルスルホニル基、n−プロピルスルホニル基、i−プロピルスルホニル基、t-ブチルスルホニル基等が挙げられる。アルキルスルホニル基を構成する炭素の数は1〜8が好ましい。
無置換のアリール基として具体的には、フェニル基、1−ナフチル基、2−ナフチル基、アズレニル基、インダニル基、テトラリニル基等が挙げられる。

これら(1)〜(85)に例示された置換基は、その中にさらに(1)〜(85)に例示された置換基を化学的に許容される範囲で有することができる。
本発明の製造方法においては、一般式(II)で表されるアジ化物と一般式(III)で表される化合物とを、水と芳香族炭化水素系溶媒と相間移動触媒とを含む混合溶媒中で反応させる。水を含んでいるにも関わらず、アジ化物からのアジ化水素の発生や、一般式(III)で表されるイミドイルハライド誘導体の過度の加水分解が生じることなく、当該混合溶媒中では、安全に反応が進み、一般式(I)で表される化合物が製造される。また、反応溶媒として非極性有機溶媒である芳香族炭化水素系溶媒を用いているが、水も併用しているため、アミン塩が不要である。
反応溶液は、一般式(II)で表されるアジ化物、一般式(III)で表される化合物、水、芳香族炭化水素系溶媒、及び相間移動触媒を混合して調製する。例えば、一般式(II)で表されるアジ化物を水に溶解させた溶液と、一般式(III)で表される化合物を芳香族炭化水素系溶媒に溶解させた溶液と、相間移動触媒を水又は芳香族炭化水素系溶媒に溶解させた溶液とをそれぞれ調製し、これらの溶液を混合することにより、反応溶液を調製することができる。この場合、相間移動触媒は、水又は芳香族炭化水素系溶媒に溶解せずに、そのまま直接反応溶液に添加してもよい。
300mLの四つ口フラスコに、N−メチル−2−オキソ−2−アセタミド(KA)8.15g(0.05mol)、溶媒としてクロロホルム90g(KAに対して、1.2L/mol)、及び触媒としてDMF0.37g(KAに対して、10mol%(0.1倍モル量))を加えた。この溶液に、滴下ロートを用いて、塩化チオニル8.93g(KAに対して、150mol%)を滴下し、還流下6時間反応させた。反応終了確認のため、反応液の一部を分取し、過剰のアジ化ナトリウムとDMFに添加して分析用試料溶液を調製した。当該分析用試料溶液中のICから(1−メチル−1H−テトラゾール−5−イル)(フェニル)メタノン(TZ)を合成した後、当該分析用試料溶液中のKAとTZをHPLC分析し、HPLCチャートにおける面積に基づいて定量した。TZのHPLC面積に対するKAのHPLC面積の比([KAのHPLC面積]/[TZのHPLC面積])が1%以下になったことを確認した。
反応終了後、室温まで冷却し、減圧下、クロロホルムと過剰の塩化チオニルを留去した。さらに過剰の塩化チオニルを留去させるために、トルエン50mLを加え、塩化チオニルを完全に留去した。この結果、濃縮物としてN−メチル−2−オキソ−2−フェニルアセチミドイルクロライド(IC)9.7g(粗収率:107mol%)が得られた。

- 下記一般式(II)
- 前記相間移動触媒が、4級アンモニウム塩である、請求項1に記載の1H−テトラゾール誘導体の製造方法。
- 前記芳香族炭化水素系溶媒が、トルエンである、請求項1又は2に記載の1H−テトラゾール誘導体の製造方法。
- 前記相間移動触媒が、ベンジル−トリ−n−ブチルアンモニウムクロリドである、請求項1〜3のいずれか一項に記載の1H−テトラゾール誘導体の製造方法。
- 前記混合溶媒中、水と芳香族炭化水素系溶媒の含有量(容量)比が、1:3〜1:5である、請求項1〜4のいずれか一項に記載の1H−テトラゾール誘導体の製造方法。
