Description
Fremgangsmåte for fremstilling av nye 5-nitro-2-furan med antibakteriell virkning.
Oppfinnelsen vedrører en fremgangsmåte for fremstilling
av nye 5_nitro-2-furaner med formel I
hvor R betegner hydrogen eller en lavere alkylgruppe og R-^ betyr metyl, acetyl eller metylkarbamoyl.
Disse nye forbindelser har sterk antibakteriell virk-
ning og angriper i meget små mengder både gram-positive og gram-negative bakterier, som Staphylococcus aureus, Streptococcus pyogenes,
Escherichia coli, Proteus vulgaris og Erysipelothrix insidiosa. De
I
kan således kombineres med vanlige bærere for dannelse av desinfek-sjonsmiddel. For slike formål tilberedes de vanligvis i form av pucider, besprøytningsmiddel, suspensjoner, oppløsninger, tabletter, salver og liknende under anvendelse av velkjente drøye og tilsetnings-midler for slike formål.
Forbindelsene med formel I har ikke bare baktericid virkning in vitro, idet de etter oral administrering på dyr infisert med patogene organismer som Staphylococcus auréus, Salmonelle Typhosa og Escherichis coli, beskytter dyrene mot å dø. Hvis således forbindelsene administreres peroralt på mus, som er letalt infisert med nevnte organismer, i doser på 40 - 150 mg/kg, kan de forhindre at dyrene dør som følge av den kraftige infeksjon. En slik administrering medfører også baktericid innhold av slike forbindelser i serum. De er således verdifulle som kemoterapeutika for behandling av systeminfek-sjoner.
De nye forbindelser fremstilles således at en forbindelse med formel II
hvor A betegner en gruppering av formelen
der R har den ovenfor angitte betydning, for det tilfelle at R-^ er metyl eller acetyl, omsettes med et alkyleringsmiddel inneholdende gruppen R-^, og for det tilfelle at R^ er metylkarbamoyl, med metylisocyanat.
j Utgangsmaterialet for syntesen av stoffene med formel II kan fremstilles ifølge norsk utlegningsskrift nr. 116.416 Eksempel 1.
lTmetylkarbamoyl- 3- ( 5- nitro- 2- furyl )-lH-l. 2. fl- triazol. ( NF- 1223)
En oppløsning av 50 g (0,278 mol) 3-(5-nitro-2-furyl)-1H-1,2,4-triazol i 300 ml dimetylformamid inneholdende 50 ml metylisocyanat oppvarmes på et dampbad under omrøring i 45 minutter. Den varme oppløsning behandles med trekull, filtreres, avkjøles, fortynnes med 300 ml vann og avkjøles grundig. Produktet filtreres ogivaskes godt med vann. Omkrystallisering tilveiebringes hurtig ved behandling av deler av produktet med kokende etanol, eh hurtig filtrering av blandingen ved frasugning og en så hurtig avkjøling av filtratet som mulig. Det i overskriften nevnte produkt fraskilles i form av lange gule nåler, som smelter ved l88-190°C (korrigert) med fornyet smelte-punkt og spaltning ved 257-259°c (korrigert).
Utbytte er 31,5 g (47.7%).
Analyee beregnet for CgH^O^: C 40,51, H 2,97, N 29,53.
Funnet: C 40,75, H 3,l8,. N 29,45.
Eksempel 2.
1- acetyl- 3-( S- nitro- 2- furyl)-lH-l. 2, 4- triazol ( NF- 1226 ).
En oppløsning av 45 g (0,25 mol) 3-(5-nitro-2-furyl)-1H-1,2,4-triazol i 300 ml eddiksyreanhydrid oppvarmes på et dampbad i 2 timer. Oppløsningsmidlet fjernes under vakuum, hvorpå resten om-krystalliseres fra etanol. Produktet fraskilles som blekgule nåler, som smelter ved l67-l68°C (korrigert) i et utbytte på 38,6 g (69,5%) av det teoretiske.
Analyse beregnet for CgHgN^: C 43.25, H 2,72, N 25,22.
Funnet: C 43.40, H 2,72, N 25,28.
Eksempel 3.
1. 5- dimetyl- 3-( S- nitro- 2- furyl)- l. 2. 4- triazol ( NF- 1103).
Til en omrørt suspensjon av 40 g (0,21 mol) 3_(5-ni*?r°-2- furyl)-lH-l,2,4-triazol i 1 liter metanol settes 11,3 g (0,21 mol) natriummetylat. Etter tilbakeløpskokning av blandingen i 20 minutter tilsettes 40 ml metyljodid dråpevis i løpet av 10 minutter. Blandingen tilbakeløpskokes i ytterligere 1,5 timer. Oppløsningsmidlet fjernes under vakuum på dampbad, resten oppslemmes med koldt vann og filtreres. Omkrystallisering fra metanol gir det i tittelen angitte produkt i form av gule nåler som smelter ved 178-l80°C med et utbytte på 15 g (34,456). Omkrystallisasjonen øker smeltepunktet til 184,5 - l85°C (korrigert).
Analyse beregnet for CgHgN^: C 46,15, H 3,87, N 26,92.
Funnet: C 46,25, H 4,06, N 26,76.
Ved sammenlikning mellom forbindelsene i henhold til eksempel 1, l-metyl-karbamoyl-3-(5-nitro-2-furylJ-1H-1,2,4-triazol, med forbindelsen i henhold til eksempel V i det franske patent nr. 3092M, nemlig 5-acetylamino-3-(5-nitro-2-furyl)-^<2>-l,2,4-tri-azolin, fremkommer følgende resultater ved peroral behandling av dyr
som er infisert med Staphylococcus aureus eller Salmonelle typhosa:
i På liknende måte fåes ved sammenlikning av forbindelsen i henhold til eksempel 2 ifølge oppfinnelsen, nemlig l-acetyl-3-(5~ nitro-2-furyl)-lH-l,2,4-triazol, med forbindelsen i henhold til éksempel IV i det franske patent nr. 3092M, nemlig l-acetyl-5-imino-3-(5-nitro-2-furyl)-A<2->1.2,4-triazolin, følgende resultat:
På samme måte fåes ved sammenlikning med forbindelsene
i henhold til eksempel 3 ifølge oppfinnelsen, nemlig l,5-dimetyl-3-(5-nitro-2-furyl)-lH-l,2,4-triazol, med forbindelsen i henhold til éksempel III ifølge fransk patent nr. 3092M> nemlig l-metyl-5-okso-3-(5-nitro-2-furyl)-A -1,2,4-triazolin, følgende resultater:

- Fremgangsmåte for fremstilling av nye antibakterielt virksomme 5-nitro-2-furaner med formel ihvor R betegner hydrogen eller en lavere alkylgruppe og R-^ betegner metyl, acetyl eller metylkarbamoyl, karakterisert ved at en forbindelse med formelen i hvor A betegner en gruppering av formelen for det tilfelle at R^ er metyl eller acetyl, omsettes med et alkyleringsmiddel inneholdende R^ eller for det tilfelle at R-^ er metylkarbamoyl, med metylisocyanat.
