Claims (6)
- 파스퇴르 피치아 파스토리스(Pichia pastoris)를 균종으로 사용하여, 먼저 1차 종자 배양을 진행하고, 균농도가 20 ± 2 g/L 일 때 2차 종자 배양으로 전환하고, 균농도가 120 ± 10 g/L일 때 2차 종자 배양을 글리세린 배양 단계로 전환하며, 글리세린 배양 단계에서 글리세린의 첨가량은 60 내지 70 g/L이고, 용존 산소가 급격히 상승하여 상대적으로 안정적인 상태에 도달할 때, 메탄올 유도 단계에 진입하고, 120 ± 8 h 동안 유도한 후 발효를 종료하는 것을 특징으로 하는, 피치아 파스토리스 발현 재조합 단백질의 발효 공법.
- 제1항에 있어서,
상기 파스퇴르 피치아 파스토리스의 기탁 번호가 CGMCC 제5021호인 것을 특징으로 하는, 피치아 파스토리스 발현 재조합 단백질의 발효 공법. - 제1 항에 있어서,
상기 1차 종자 배양은, 균종 파스퇴르 피치아 파스토리스를 종자 배지에 접종하여, 30 ℃에서 균체 습중량이 20 ± 2 g/L에 도달할 때까지 24 내지 36 h 동안 진탕 배양하는 것을 특징으로 하는 피치아 파스토리스 발현 재조합 단백질의 발효 공법. - 제1 항에 있어서,
상기 2차 종자 배양은, 1차 종자액을 모두 발효 배지에 옮긴 후, 30 ℃에서 배양하며, 암모니아수로 pH 값을 5.0으로 조절 및 제어하고, 용존 산소는 20 내지 30 %로 제어하며, 균체 습중량이 120 ± 10 g/L에 도달할 때까지 배양하는 것을 특징으로 하는, 피치아 파스토리스 발현 재조합 단백질의 발효 공법. - 제1 항에 있어서,
상기 글리세린 배양 단계는, 2차 종자액을 발효 배지에 옮긴 후, 60 내지 70 g/L의 글리세린을 첨가하여, 30 ℃에서 배양하며, 암모니아수로 pH값을 5.0으로 조절 및 제어하고, 통기량은 30 m3/h이며, 교반 속도는 300 내지 500 rpm이고, 용존 산소가 급격이 상승할 때, 1 h 동안 기아 상태로 하여 글리세린을 소진시키는 것을 특징으로 하는, 피치아 파스토리스 발현 재조합 단백질의 발효 공법. - 제1 항에 있어서,
상기 메탄올 유도 단계는, 유도 단계 온도를 30 ℃, pH 값을 5.0으로 제어하고, 메탄올 보충재료와 용존 산소의 연동을 설정하며, 용존 산소가 20 %보다 높을 때 메탄올 유가(fed-batch)를 시작하고, 용존 산소가 20 %보다 낮을 때 메탄올 유가를 정지시키며, 120 ± 8 h 동안 유도한 후, 발효를 종료하고, 발효액을 원심 분리하며, 원심 분리 후의 상청액을 수집하여 농축, 한외 여과 및 나노 여과 탈색 탈염을 차례로 진행함으로써 재조합 인간 콜라겐을 얻는 것을 특징으로 하는, 피치아 파스토리스 발현 재조합 단백질의 발효 공법.
