Abstract
PURPOSE: To obtain the titled compound in high yield industrially and advantageously with eacy separation of the unreacted substance, by reacting a specific hydroxycholest-5-ene with a brominating agent, and then a dehydrobrominating agent, irradiating the reaction product with ultraviolet light, and isomerizing the irradiated previtamin D3 under heating.
CONSTITUTION: A hydroxycholest-5-ene of formula I (R1 and R2 are lower alkoxycarbonyl; R3 and R4 are H or lower alkoxycarbonyloxy), having hydroxyl groups protected by lower alkoxycarbonyl groups is reacted with an allylic brominating agent, preferably in an inert organic solvent at room temperature to 140°C. The reaction product is then reacted with a dehydrobrominating agent preferably at 100W180°C to give a hydroxycholesta-5,7-diene of formula II, which is irradiated with ultraviloet light to form a previtamin D3 of formula III. The unreacted compound of formula II is separated by an organic solvent, etc. and circulated for reuse. The compound of formula III is isomerized by heat, and the protecting groups are removed to afford the aimed compound of formula IV.
