Abstract

PURPOSE: To obtain the titled compound in high yield industrially and advantageously, by reacting a hydroxycholest-5-one having a hydroxyl group protected with a specific protecting group and then with a debrominating agent, and further irradiating the reaction product with ultraviolet light, etc.
CONSTITUTION: A hydroxycholest-5-one, e.g. 1α,3β-diethoxycarbonyloxycholest- 5-ene, of formula I (R1 and R2 are lower lower alkoxycarbonyl; R3 and R4 are H or alkoxycarbonyloxy) is reacted with preferably 1W2 equivalent amounts of an alkylic brominating agent, e.g. N-bromosuccinimide. The reaction product is then reacted with a dehydrobrominating agent, e.g. trimethyl phosphite, in a molar amount of preferably two or more times that of the bromination product to give the corresponding hydroxycholesta-5,7-diene, which is then irradiated with ultraviolet light and isomerized by heat. The protecting groups are further removed to afford the aimed compound, e.g. 1α-hydroxycholecalciferol, of formula II.
