本発明は、概して、抗BCMAキメラ抗原受容体(CAR)を結合する抗体および抗原結合断片、ならびにそのコンジュゲート、組成物、ならびにそれらを使用する方法に関する。特に好適な実施形態では、本発明は、配列番号25に記載の抗BCMA CARを結合する抗体、断片、およびコンジュゲート、その組成物および使用に関する。
本開示をさらに詳細に記載する前に、その理解には、本明細書に使用される特定の用語の定義を提供することが役立ちうる。
具体的な実施形態では、抗BCMA CARを結合する抗体またはその抗原結合断片が提供される。
様々な実施形態では、抗体またはその抗原結合断片と標識とを含むコンジュゲートが提供される。好適な実施形態では、コンジュゲートは、抗BCMA CARを結合する抗体またはその抗原結合断片と、検出可能な標識または検出可能なシグナルを産生可能な標識とを含む。さらに好適な実施形態では、コンジュゲートは、検出可能な標識にカップリングされた抗BCMA CARを結合する抗体またはその抗原結合断片を含む。さらにいっそう好適な実施形態では、コンジュゲートは、検出可能な標識に共有結合により結合するかまたは化学的にカップリングされている、抗BCMA CARを結合する抗体またはその抗原結合断片を含む。
様々なポリペプチド、融合ポリペプチド、およびポリペプチドバリアントが本明細書で想定され、そのようなものとしては、以下に限定されないが、抗体およびその抗原結合断片が挙げられる。好適な実施形態では、ポリペプチドは、抗BCMA CARを結合する抗体またはその抗原結合断片を含む。
好適な実施形態では、抗BCMA CARを結合する抗体またはその抗原結合断片をコードするポリヌクレオチドが提供される。本明細書に使用される際に、用語「ポリヌクレオチド」または「核酸」は、メッセンジャーRNA(mRNA)、RNA、ゲノムDNA(gDNA)、相補的DNA(cDNA)、または組換えDNAを指す。ポリヌクレオチドとしては、一本鎖および二本鎖のポリヌクレオチドが挙げられる。具体的な実施形態では、ポリヌクレオチドとしては、本明細書で想定される任意の参照配列と少なくとも約50%、55%、60%、65%、66%、67%、68%、69%、70%、71%、72%、73%、74%、75%、76%、77%、78%、79%、80%、81%、82%、83%、84%、85%、86%、87%、88%、89%、90%、91%、92%、93%、94%、95%、86%、97%、98%、または99%の配列同一性を有するポリヌクレオチドまたはバリアントが挙げられる。様々な説明的な実施形態では、本明細書で想定されるポリペプチドをコードするポリヌクレオチドとしては、以下に限定されないが、配列番号1〜24に記載のポリペプチド配列が挙げられる。
本明細書で想定される組成物は、抗体または抗体断片およびその抗原結合断片、コンジュゲート、ポリペプチド、ポリヌクレオチドのうち1つまたは複数を含みうる。組成物としては医薬組成物が挙げられるがこれに限定されない。追加的な剤が組成物の活性成分の機能または活性に不利な影響を及ぼさない限り、組成物にやはり含まれうる他の構成成分には実質的に限定はない。
様々な実施形態では、抗BCMA CARを結合する1つまたは複数の抗体またはその抗原結合断片と、抗体を使用して1つまたは複数の免疫エフェクター細胞または抗BCMA CAR T細胞上の抗BCMA CARの発現を検出、決定、および/または測定するための指示書とを含むキットが提供される。
抗BCMA CAR T細胞活性は、免疫エフェクター細胞上の抗BCMA CARの発現に部分的に関連する。抗BCMA CARの発現をex vivoまたはin vitroとin vivoとの両方で正確に評価する能力が、薬剤製品の活性を評価するために、および患者内での抗BCMA CAR T細胞の生存および持続性を追跡するために必要である。

- 前記抗体またはその抗原結合断片または請求項1から35のいずれか1項と検出のための手段とを含むコンジュゲート。
- 前記抗体またはその抗原結合断片または請求項1から35のいずれか1項と検出手段とを含むコンジュゲート。
- 前記抗体またはその抗原結合断片または請求項1から35のいずれか1項と検出可能な標識とを含むコンジュゲート。

- 請求項1から35のいずれか1項に記載の抗体もしくはその抗原結合断片、請求項36から51のいずれか1項に記載のコンジュゲート、請求項52に記載のポリヌクレオチド、または請求項53に記載の宿主細胞を含む組成物。
- 請求項1から35のいずれか1項に記載の抗体またはその抗原結合断片を宿主細胞で発現することと、前記抗体を回収または単離することとを含む、抗体またはその抗原結合断片を生産する方法。
- 請求項1から35のいずれか1項に記載の抗体もしくはその抗原結合断片または請求項36から51のいずれか1項に記載のコンジュゲートと、使用のための指示書とを含むキット。
- 請求項1から35のいずれか1項に記載の抗体もしくはその抗原結合断片または請求項36から51のいずれか1項に記載のコンジュゲートにT細胞を接触させること;ならびに抗体:抗BCMA CAR複合体の形成を検出することを含む、T細胞上の抗BCMA CARの発現を検出する方法。
- 請求項1から35のいずれか1項に記載の抗体もしくはその抗原結合断片または請求項36から51のいずれか1項に記載のコンジュゲートにT細胞集団を接触させること;ならびに前記1つまたは複数のT細胞上の抗体:抗BCMA CAR複合体の形成を検出することを含む、T細胞集団での抗BCMA CARの発現を検出する方法。
- 請求項1から35のいずれか1項に記載の抗体もしくはその抗原結合断片または請求項36から51のいずれか1項に記載のコンジュゲートにT細胞を接触させること;抗体:抗BCMA CAR複合体の形成を検出すること;ならびに前記複合体の量を測定して前記T細胞上の前記抗BCMA CARの発現を決定することを含む、T細胞上の抗BCMA CARの発現を決定する方法。
- 請求項1から35のいずれか1項に記載の抗体またはその抗原結合断片または請求項36から51のいずれか1項に記載のコンジュゲートにT細胞集団を接触させること;1つまたは複数のT細胞上の抗体:抗BCMA CAR複合体の形成を検出すること;ならびに前記複合体の量を測定して前記1つまたは複数のT細胞上の前記抗BCMA CARの発現を決定することを含む、T細胞集団での抗BCMA CARの発現を決定する方法。
- 請求項1から35のいずれか1項に記載の抗体またはその抗原結合断片または請求項36から51のいずれか1項に記載のコンジュゲートにT細胞集団を接触させること;1つまたは複数のT細胞上の抗体:抗BCMA CAR複合体の形成を検出すること;ならびに前記抗BCMA CARを発現するT細胞を計数することを含む、T細胞集団中の抗BCMA CAR+T細胞の数を決定する方法。
- 免疫組織化学的検査、酵素結合免疫吸着アッセイ(ELISA)、免疫吸着アッセイ、化学発光アッセイ、電気化学発光アッセイ、表面プラズモン共鳴(SPR)ベースバイオセンサー(例えばBIAcore)、蛍光顕微鏡観察、またはフローサイトメトリー、蛍光活性化細胞分取法(FACS)を使用して、抗体:抗BCMA CAR複合体の形成を検出する、請求項57から61のいずれか1項に記載の方法。
