Abstract
Один аспект настоящего изобретения относится к средству, представляющему собой двухнитевую РНК (dsRNA), способному подавлять экспрессию целевого гена. Смысловая нить средства, представляющего собой dsRNA, содержит по меньшей мере один нарушающий термостабильность нуклеотид, при этом по меньшей мере один указанный нарушающий термостабильность нуклеотид встречается в сайте, противоположном затравочному участку (положения 2-8) антисмысловой нити; а антисмысловая нить средства, представляющего собой dsRNA, содержит по меньшей мере два нуклеотида с модификацией, придающей нуклеотиду стерический объем, который меньше стерического объема 2'-ОМе-модификации или равен ему, где указанные нуклеотиды с модификацией разделены участком длиной 11 нуклеотидов. Другие аспекты настоящего изобретения относятся к фармацевтическим композициям, содержащим эти средства, представляющие собой dsRNA, подходящие для применения в терапевтических целях, и к способам подавления экспрессии целевого гена путем введения этих средств, представляющих собой dsRNA, например, для лечения различных патологических состояний.
