Abstract
POSTUPAK ZA DOBIJANJE NOVIH GRF ANALOGA definisanih formulom (I): H-Tyr-Ala-Asp-Ala-Ile-Phe-Thr-Asn-Ser-Tyr-Arg-Lys-R13-Leu-Gly-Gln-Leu-Ser-Ala-Arg-Lys-Leu-Leu-Gln-Asp-Ile-Met-R28-Arg-Gln-Gln-Gly-Glu-Arg-Asn-Gln-Glu-Gln-Gly-Ala-R41-(I) Val-Arg-Leu-Y Gde Y je OH ili NH2, R13 je Val ili Ile R28 je Ser ili Asn i R14 je Arg ili Lys, naznačen time, što obuhvata: -reakciju BOC-Leu u odnosu od oko 1 mmol na gram MBHA ili hlormetilovane polistirenske smole u odgovarajućem rastvaraču, kao što je metilen hlorid ili dimetilformamid ili njihova smeša; -deprotektiranje Leu uklanjanjem BOC grupe i vezivanje zaostalih amino kiselina u nizu tako da grade jedinjenje koje ima formulu X1-Tyr (X2)-Ala-Asp- (X3)-Ala-Ile-Phe-Thr(X4)-Asn-Ser(X5)- (II) Tyr(X2)-Arg(X6)-Lys(x7)-R13-Leu-Gly-Gln-Leu-Ser(X5)-Ala-Arg(X6)-Lys(X7)-Leu-Leu-Gln-Asp(X3)-Ile-Met-R28(X5)-Arg(X6)-Gln-Gln-Gly-Glu-(X3)-Arg(X6)-Asn-Gln-Glu(X3) Gln-Gly-Ala-R14(X6 ili X7)-Val-Arg(X6)-Leu-X8 gde X1 je BOC, X2 je 2,6-dihlorbenzil, X3 je Bzl estar, X4 je Bzl, X5 je Bzl, X6 je Tes, X7 je 2-hlorbenziloksikarbonil, i X8 je -O-CH2-smolasti nosač ili -NH-smolasti nosač, i - cepanje zaštitnih grupa i odvajanje od pomenutog smolastog nosača pomoću TFA u odgovarajućem rastvaraču, kao što je CH2C12, i zatim pomoću HF i hvatača radikala, kao što je anizol, metil-etilsulfid ili njihova smeša, na niskoj temperaturi, poželjno oko 0 C do -20 C, i tada rastvaranje u odgovarajućem rastvaraču, poželjno sirćetnoj kiselini i dalje prečišćavanje.
