Claims (38)
- 런어웨이 레플리콘(runaway replicon)과 생활성 소 성장 호르몬 유도체를 코드화하는 하기 유전자 (1), (2), (3), (4), (5) 또는 (6)의 판독대 중에 존재하는 전사 및 해독 활성화 서열을 연결시킴을 특징으로 하여, 선별가능하며 자발적으로 복제하는 제조합체 DNA 발현 벡터를 제조하는 방법.상기식에서 A는 데옥시아데닐이고, G는 데옥시구아닐이며, C는 데옥시시토실이고, T는 티미딜이며, R은 소 성장 호르몬의 아미노산 9(로이신) 내지 191(페닐알라닌)을 코드화하는 DNA 서열이고, R1은 하기의 해독정지 시그널을 코드화하는 DNA 서열이다.
- 제1항에 있어서, 전사 활성화 서열이 이.콜라이 락토오즈, 박테리오파아지 λPLOL, 박테이오파아지 λPROR, tac, 이.콜라이 리포프로테인 또는 이.콜라이 트립토판 전사 활성화 서열인 방법.
- 제1 또는 2항에 있어서, 벡터가 플라스미드인 방법.
- 제3항에 있어서, 해독 활성화 서열 이.콜라이 리포프로테인 서열인 방법.
- 제3항에 있어서, 해독 활성화 서열이 이.콜라이 트립토판 서열인 방법.
- 플라스미드 pCZ101을 BstEII 제한효소로 처리하고, 생성된 DNA를 정제한 후, 정제된 DNA를 연결시켜 플라스미드 pCZ013을 생성시킴을 특징으로 하여, 제8 및 17 내지 23항중 어느 한항에 따른 방법에 사용되기에 적합한 플라스미드를 제조하는 방법.
- a) 런어웨이 레플리콘과 b) 작용성 폴리펩타이드의 카복시 말단을 코드화하는 뉴클레오티드 트리플리트에 인접하여 하부에 위치한 해독 정지 시그널을 함유하는 작용성 폴리펩타이드를 코드화하는 뉴클레오티드 서열에 판독대 중에 존재하는 전사 및 해독활성화 서열로 이루어진 선별가능하고 자발적으로 복제하는 재조합체 DNA 발현 벡터로 캄피턴트 세포를 형질 전환시킨 후, 형질 전환된 세포를 유전자 발현 및 런어웨이 레플리콘의 활성화 또는 유도에 적절한 증식 조건하에서 배양시킴을 특징으로 하여, 세포내 균질성이 높은 단백성 과립을 제조하는 방법.
- 제25항에 있어서, 재조합체 DNA 발현 벡터가 플라스미드인 방법.
- 제25 또는 제26항에 있어서, 전사 활성화 서열이 이.콜라이 트립토판 전사 활성화 서열, 이.콜라이 리포프로테인 전사 활성화 서열, 이.콜라이 락토오즈 전사 활성화 서열, 박테이오파아제 λPLOL전사 활성화 서열 또는 박테리아파아지 λPROR전사 활성화 서열인 방법.
- 제25 내지 27항중 어느 한항에 있어서, 전사 활성화 서열이 하나 이상의 이.콜라이 전사 활성화 서열을 직렬로 함유하는 방법.
- 제27항에 있어서, 전사 활성화 서열이 이.콜라이 트립토판 전사 활성화 서열 또는 이.콜라이 리포프로테인 전사 활성화 서열인 방법.
- 제25항 내지 29항중 어느 하나에 있어서, 작용성 폴리펩타이드 코드화 서열이 소 성장 호르몬, 인체 성장 호르몬, 인체 전-성장 호르몬, 돼지의 성장 호르몬, 포유동물 성장 호르몬, 조류 성장 호르몬, 인체 인슐린 A쇄, 인체 인슐린 B쇄, 인체 프로인슐린, 인체 프리-프로인슐린(pre-proinsulin), 인터페론, 유로키나아제, 인체조직 플라스미노겐 활성화제, 성장 호르몬 유리인자 또는 인터루킨(interleukin) II를 코드화하는 서열인 방법.
- 제26 내지 30항중 어느 한항에 있어서, 플라스미드가 플라스미드 pCZ1920, pJR1, pCZ112, pAT1, pAT2, pASP1, pASP2, pCZ201, pJR1.3, pAT1.3, pAT2.3, pASP1.3, pASP2.3, pCZ154.3, pCZ155.3 또는 pCZ156.3인 방법.
- 제25 내지 31항중 어느 한항에 있어서 형질전환된 숙주 세포가 원핵 세포인 방법.
- 제32항에 있어서, 형질전환된 숙주 세포가 이.콜라이인 방법.
- 제33항에 있어서, 형질전환된 숙주 세포가 이.콜라이 K12 RV308인 방법.
- 제1 내지 23항중 어느 한항에 따른 방법에 의해 생성되는 벡터내에 포함되어 있는 유전자에 의해 코드화되는 소의 성장 호르몬 유도체(단, 메티오닐 소 성장 호르몬은 제외)를 우유 생산을 증가시키는데 유효한 양으로 젖소에게 투여함을 특징으로 하여, 젖소에서의 우유 생산을 증가시키는 방법.
- 제35항에 있어서, 유효량이 동물당 1일 약 0.05 내지 100,000μg인 방법.
- 제36항에 있어서, 유효량이 동물당 1일 약 5 내지 25,000μg인 방법.
