Abstract
Description
본 명세서에 포함된 도면들 중 일부는 상이한 시야각에서 관찰된 본 발명의 다양한 구체예들을 도시한다. 하기의 상세한 설명에서 평면도, 저면도, 또는 측면도와 같은 도면들이 언급되지만, 이러한 도면들은 단지 설명을 위한 것으로, 별도로 명시되지 않는 한, 본 발명이 특정한 공간상의 방향으로 구현되거나 이용되는 것을 암시하거나 요구하는 것은 아니다.
도 1은 예시적인 구체예로서 미세유체 분취(MA) 칩의 평면도를 도시한다.

상기 도면들은 본 발명을 기재된 형태로 정확하게 구현하거나 제한하기 위한 것이 아니다. 본 발명은 수정되거나 변경된 형태로 실시될 수 있으며 오직 청구항 및 이들의 균등물에 의해서만 제한됨을 이해해야 한다.
Claims (20)
- 반경, 중심, 상부면, 하부면, 외각 가장자리, 및 두께를 갖는 디스크 형태의 칩;
실질적으로 상기 칩의 중심에 배치되고, 상기 칩의 상부면으로 연장되며, 상기 칩의 상부면으로부터 접근 가능한 단일 중심웰(입구);
상기 칩의 외각의 환상 부분에 배치되고, 상기 칩의 상부면으로 연장되며, 상기 칩의 상부면으로부터 접근 가능한 복수 개의 측면 웰들(출구); 및
상기 칩의 하부면에서 연장되며, 상기 중심웰 및 상기 측면 웰들과 유체 소통하여(fluid communication) 상기 중심웰에서부터 상기 측면 웰들까지 연장되는 복수 개의 채널들;을 포함하는 것을 특징으로 하는 단일 세포 분리용 미세유체 분취(MA) 칩. - 제1항에 있어서,
상기 칩은 폴리디메틸실록산(PDMS), 폴리(메틸 메타크릴레이트)(PMMA), 폴리스티렌(PS), 및 폴리카보네이트(polycarbonate, PC)로 이루어진 군으로부터 선택되는 가요성 또는 반강성 재료로 제조된 얇은 시트를 포함하는 것을 특징으로 하는 단일 세포 분리용 미세유체 분취(MA) 칩. - 제1항에 있어서,
상기 칩은,
약 1 mm의 두께;
페트리 접시 내에 장착 가능한 크기 및 형태; 및
약 3 cm의 반경; 중 적어도 어느 하나의 특징을 갖는 것을 특징으로 하는 단일 세포 분리용 미세유체 분취(MA) 칩. - 제1항에 있어서,
상기 중심웰은 상기 칩을 완전히 통과하도록 연장되고; 그리고
상기 측면 웰들은 상기 칩을 완전히 통과하도록 연장되는; 것을 특징으로 하는 단일 세포 분리용 미세유체 분취(MA) 칩. - 제1항에 있어서,
상기 중심웰은,
약 3 mm의 직경; 및
약 4 ㎕의 부피; 중 적어도 하나의 특징을 갖는 것을 특징으로 하는 단일 세포 분리용 미세유체 분취(MA) 칩. - 제1항에 있어서,
상기 측면 웰들 중 적어도 일부의 각각은,
약 1.5 mm의 직경; 및
약 1 ㎕의 부피; 중 적어도 하나의 특징을 갖는 것을 특징으로 하는 단일 세포 분리용 미세유체 분취(MA) 칩. - 제1항에 있어서,
상기 복수개의 측면 웰들의 제1 세트는 상기 칩의 중심에서부터 제1 반경(r1)에 위치하고; 그리고 상기 복수개의 측면 웰들의 제2 세트는 상기 칩의 중심에서부터 제2 반경(r2)에 위치하며, 상기 제2 반경(r2)은 상기 제1 반경(r1) 보다 큰 것을 특징으로 하는 단일 세포 분리용 미세유체 분취(MA) 칩. - 제7항에 있어서,
상기 측면 웰들의 제1 세트 및 제2 세트는 상기 측면 웰들의 제2 세트가 60개의 측면 웰을 포함하는 상기 제1 세트 사이의 원주 위치에 위치하도록 상호배치(interleave)되는 것을 특징으로 하는 단일 세포 분리용 미세유체 분취(MA) 칩. - 제7항에 있어서,
상기 측면 웰들의 제1 세트 및 제2 세트는 상기 칩의 외각의 환상 부분 주위에 균등하게 분포되는 것을 특징으로 하는 단일 세포 분리용 미세유체 분취(MA) 칩. - 제1항에 있어서,
상기 칩의 외각의 환상 부분은 상기 칩의 반경의 적어도 70%에서부터 상기 칩의 외각 가장자리까지 연장하는 것을 특징으로 하는 단일 세포 분리용 미세유체 분취(MA) 칩. - 제1항에 있어서,
상기 채널들은,
상기 중심웰에서부터 상기 측면 웰들로 방사상으로 연장;
상기 칩 내로 단지 부분적으로 연장;
약 30 ㎛의 너비; 및
상기 중심웰에서부터 상기 측면 웰들까지 단일 세포들의 이동을 수용할 수 있는 크기 및 형상; 중 적어도 어느 하나의 특징을 갖는 것을 특징으로 하는 단일 세포 분리용 미세유체 분취(MA) 칩. - 제1항에 있어서,
상기 칩은,
현미경 관찰하에 상기 측면 웰들을 고유하게 식별할 수 있도록 상기 측면 웰들의 적어도 일부의 내측에 위치하는 비교적 작은 ㎛ 단위의 표식들; 및
육안에 의해 상기 측면 웰들을 고유하게 식별할 수 있도록 상기 측면 웰들의 적어도 일부의 외측에 위치하는 비교적 큰 mm 단위의 표식들;을 더 포함하는 것을 특징으로 하는 단일 세포 분리용 미세유체 분취(MA) 칩. - 제1항에 있어서,
상기 칩은 상기 중심웰 내로 세포 현탁액을 주입하기 위한 캡을 더 포함하고, 상기 캡은 관통 연장하는 구멍을 가지며, 상기 중심웰의 상부에 배치되는 것을 특징으로 하는 단일 세포 분리용 미세유체 분취(MA) 칩. - 제13항에 있어서,
상기 캡은 원통형이며, 상기 캡의 하부면으로부터 상기 중심웰 내로 연장하는 립 또는 숄더를 포함하며, 상기 립 또는 숄더는 세포 현택액의 주입 동안 상기 캡을 일시적으로 고정시키는 것을 특징으로 하는 단일 세포 분리용 미세유체 분취(MA) 칩. - 제13항에 있어서,
상기 캡은 상기 칩과 일체로 형성되는 것을 특징으로 하는 단일 세포 분리용 미세유체 분취(MA) 칩. - 단일 중심웰 및 복수개의 마이크로채널에 의해 상기 중심웰에 연결되는 복수개의 측면 웰들을 포함하는 미세유체 분취(MA) 칩 내로 소수개의 세포를 함유하는 세포 현탁액을 주입하는 단계; 및
상기 마이크로채널들을 통해 정압 구동 흐름에 의해 단일 세포들을 상기 측면 웰들로 이동시키는 단계;를 포함하는 것을 특징으로 하는 소수 개의 세포를 함유하는 세포 현탁액에서 단일 세포들을 분리하는 방법. - 제16항에 있어서,
상기 방법은,
혈액에서 하나의 또는 희소한 순환성 종양 세포(circulating tumor cells, CTCs)를 동정하고 분리하는 단계; 및
단일세포 및 대상을 복잡한 매트릭스에서 동정 및 분리하는 단계; 중 적어도 하나를 포함하고,
상기 CTC 세포들은 단일 또는 다단계의 MA 칩 공정 후에 포획되고, 상기 동정 및 분리하는 단계는 박테리아, 비드(bead), 입자, 바이러스를 포함하는 단일 대상을 매트릭스에서부터 분리하는 것, 결핵균을 분리하는 것, 및 특정 유전자형/표현형의 효모 세포를 분리하는 것을 포함하는 것을 특징으로 하는 소수 개의 세포를 함유하는 세포 현탁액에서 단일 세포들을 분리하는 방법. - 제16항에 있어서,
상기 방법은,
상기 측면 웰들 내의 세포를 식별하는 단계; 및
단일 세포 만을 함유하는 상기 측면 웰들로부터 세포들을 회수하는 단계;를 더 포함하는 것을 특징으로 하는 소수 개의 세포를 함유하는 세포 현탁액에서 단일 세포들을 분리하는 방법. - 중심 및 표면을 갖는 디스크 형태의 칩;
상기 칩의 실질적인 중심에 위치하는 단일 중심웰(입구);
상기 칩의 외각의 환상 부분에 위치하는 복수 개의 측면 웰(출구); 및
상기 중심웰에서부터 상기 측면 웰들로 연장하는 복수 개의 채널들;을 포함하는 미세유체 분취액 칩을 제공하는 단계를 포함하고,
단일 중심웰 및 복수개의 마이크로채널에 의해 상기 중심웰에 연결되는 복수개의 측면 웰들을 포함하는 미세유체 분취(MA) 칩의 입구로 세포 현탁액을 주입하는 단계; 및
상기 마이크로채널들을 통해 정압 구동 흐름에 의해 단일 세포들을 상기 측면 웰들로 이동시키는 단계;를 더 포함하는 것을 특징으로 하는 단일 세포 분석을 수행하는 방법, - 제19항에 있어서,
상기 방법은 상기 중심웰 내로 세포가 불포함된 배지를 첨가하여 상기 채널들에 잔여하는 단일 세포들이 상기 측면 웰들로 흘러가도록 양성 정수압을 생성하는 단계를 더 포함하는 것을 특징으로 하는 단일 세포 분석을 수행하는 방법.
