MTT 분석은 Carmichael의 방법에 따라 실험을 진행하였다. 96 웰 플레이트에 1X105 cells/well인 Raw264.7을 180 μl씩 분주하였다. 분주된 세포에 엉겅퀴 및 모링가 온천수 발효 추출물 20 μl를 첨가한 후 37℃, 5% CO2 배양기에서 24시간 동안 배양하였다. 여기에 2.5 mg/ml 농도로 제조한 MTT 용액을 40 μl 첨가하여 4시간 동안 배양하였다. 배양 후 배양액을 제거하고 각 웰당 DMSO 100 μl를 가하여 실온에서 10분 동안 반응시킨 뒤 ELISA 리더로 540 nm에서 흡광도를 측정하였다. 세포 독성 측정은 엉겅퀴 및 모링가 온천수 발효 추출물 첨가군과 무첨가군의 흡광도 감소율로 나타내었으며, 하기 수학식 3과 같이 계산하였다. 엉겅퀴 및 모링가 온천수 발효 추출물에 대한 Raw264.7의 MTT 분석 결과는 도 4에 나타내었다.MTT analysis was conducted according to Carmichael's method. The results of MTT analysis of Raw264.7 on fermented extracts of milk thistle and Moringa hot spring water are shown in FIG. 4 .
