인삼의 재배 후 갓 수확되는 형태의 말리지 않은 인삼을 수삼이라 하는데 수삼은 수분을 약 60~70%를 포함 하고 있다. 이는 수삼의 저장성을 약화시키는 원인이 되는데 백삼은 수삼을 그대로 또는 껍질을 살짝 벗겨내고 말려 건조한 삼이다. 수분 함량이 15% 이하가 되도록 건조한 인삼으로 형태에 따라 직삼(直蔘), 반곡삼(半曲蔘), 곡삼(曲蔘)으로 구분한다. 홍삼은 수삼을 수증기로 찐 다음 건조시킨 담홍갈색의 인삼이다. 태극삼은 홍삼과 백삼의 중간 상태의 제품이다. 수삼을 뜨거운 물속에 일정 시간 담가 익힌 다음 건조해 제조한다. 상기 실시예 1에서 공비증류에 의해 분리된 인삼 자유수의 항산화 효과를 확인하기 위하여 ABTS (2,2-Azino-bis(3-ethylbensothiazoline-6-sulfonic acid) assay를 수행하였다. 증류수를 용매로 하여 7.4 mM ABTS 용액과 2.45 mM potassium persulfate용액을 준비한 뒤, 시약을 질량비 1:1로 혼합하고 암소에서 24시간 보관하여 ABTS radical을 형성시켰다. L-ascorbic aicd는 농도가 25, 50, 100, 200 ppm이 되도록 준비하고 시험할 시료는 50배로 희석하여 준비하였다. 실험하기 전, 암소에서 24시간 방치한 ABTS radical 용액 5 mL을 증류수 95 mL로 희석하고 시험할 시료 및 양성대조군을 10 μL씩 96-well plate에 넣었다. 희석한 ABTS radical 용액을 190 μL씩 넣어주고 control 군에는 동일한 양의 증류수를 넣어주었다. 해당 96-well plate는 실온, 암소에서 10분간 반응시키고 microplate reader를 이용하여 UV 730 nm애서 흡광도를 측정하였다. ABTS 자유 라디칼 소거 활성은 하기의 화학식 1에 따라 계산하고, 그 결과를 도 3에 나타내었다. ABTS (2,2-Azino-bis(3-ethylbensothiazoline-6-sulfonic acid) assay was performed to confirm the antioxidant effect of free ginseng water separated by azeotropic distillation in Example 1.
