Claims (4)
- 파라핀포매된 생물학적 조직의 샘플을 슬라이스하여 만든 절편을 탈파라핀용액으로 탈파라핀한 후, 용해 완충액(lysis buffer)과 단백질분해제를 순차적으로 투입하여 분해된 단백질이 함유된 용해 완충액을 DNA를 추출하기 위한 DNA 추출용액으로 수득하는 과정을 포함하여 이루어지며,
상기 용해 완충액의 투입시에 용해 완충액을 염색하기 위한 염색시약을 투입하여 상기 용해 완충액이 염색되도록 하는 것을 특징으로 하는 파라핀포매된 샘플로부터 DNA를 추출하는 방법.
- 파라핀포매된 생물학적 조직의 샘플을 슬라이스하여 절편을 만드는 과정;
상기 절편을 원심분리용 튜브에 투입하고 탈파라핀용액을 첨가하는 과정:
상기 원심분리용 튜브에 용해 완충액(lysis buffer)과 상기 용해 완충액을 염색시키는 염색시약을 첨가하는 과정;
상기 절편에 용해 완충액과 염색시약이 첨가된 샘플을 원심분리하는 과정;
상기 원심분리된 샘플의 하층액에 단백질분해제를 첨가하는 과정;
상기 단백질분해제가 첨가된 샘플을 원심분리하여 분해된 단백질이 함유되며 상기 염색시약에 의해 염색된 용해 완충액과 탈파라핀용액을 분리하는 과정;
상기 용해 완충액을 DNA를 추출하기 위한 DNA 추출용액으로 수득하는 과정; 및
상기 DNA 추출용액에서 DNA를 추출하는 과정;을 포함하여 이루어진 것을 특징으로 하는 파라핀포매된 샘플로부터 DNA를 추출하는 방법.
- 제1항에 있어서,
상기 용해 완충액은 물을 포함하며,
상기 염색시약은 용해 완충액의 물에 용해되어 발색되는 것을 특징으로 하는 파라핀 포매된 샘플로부터 DNA를 추출하는 방법.
- 제3항에 있어서,
상기 염색시약은 자일렌시아놀(Xylene cyanol), 브롬페놀블루(Bromphenol blue), 폰소 S(Ponceau S) 중에서 선택되는 것을 특징으로 하는 파라핀 포매된 샘플로부터 DNA를 추출하는 방법.
