Abstract

PURPOSE: To determine the amount of glucagon excellently by utilizing the immunological reactivity of an antibody obtained from glucagon and β-galactosidase-glucagon fragment conjugate obtained by using a specific cross-linking agent.
CONSTITUTION: A glucagon fragment which is a peptide shown by formula I (wherein R1 is shown by formula II) and β-galactosidase are made to react by a dithiocarboxylic acid cross-linking agent shown by formula III (wherein R2 is an organic group having S-S exchange reactivity, such as 2'-benzothiazolyl group, X is a spacer group such as an alkylene group) or a reactive derivative thereof. By using the β-galactosidase-glucagon fragment conjugate thus obtained and an antibody obtained by using the conjugate of a protein with the glucagon fragment of a peptide shown by formula IV (wherein R3 is shown by formula V), especially a peptide shown by formula VI, a very excellent immune reaction can be conducted, whereby the glucagon in the liquid specimen is determined enzymoimmunologically.
