Abstract
NEW MATERIAL:5,6-Trans-N-Desacetyl derivative of antibiotic PS-5 of formula I and a carboxyl-protected derivative of formula II (R1 is carboxyl-protecting group).
USE: Antibacterials and an intermediate for their synthesis, effective against a wide variety of Gram-positive and Gtram-negative bacteria including β-lactam antibiotic- resistant bacteria, particularly staphylococci.
PROCESS: A carboxyl-protecting group, e.g. a lower alkyl group, is introduced into a 5,6-trans antibiotic of formula III to give a compound of formula II, which is then reacted with an iminohalogenating agent, e.g. PCl5, to form a compound of formula V (X is halogen). The resulting compound is reacted with an alcohol to yield a compound of formula VI (R2 is hydrocarbon group), which is treated with a large amount of water. The protecting group is eliminated to give the compound of formula I.
