Claims (1)
- 【特許請求の範囲】 1.a)第1リガンド結合ドメインを含む第1受容体ポリペプチドをコードす る第1受容体発現カセット; b)第2リガンド結合ドメインを含む第2受容体ポリペプチドをコードす る第2受容体発現カセット; c)標的ポリペプチドをコードする標的発現カセット を含んでなる、トランスジェニック植物細胞または植物体およびその後代。 2.標的発現カセットが、植物稔性を妨害する標的ポリペプチドをコードする ものである、請求の範囲第1項に記載の植物細胞または植物体。 3.a)第1リガンド結合ドメインを含む第1受容体ポリペプチドをコードす る第1受容体発現カセット; b)第2リガンド結合ドメインを含む第2受容体ポリペプチドをコードす る第2受容体発現カセット; c)標的ポリペプチドをコードする標的発現カセット を有する植物細胞または植物体を形質転換することからなる、請求の範囲第1項 に記載の植物細胞または植物体の製法。 4.該発現カセットで形質転換させた植物細胞または植物体の後代を得ること を更に含んでなる、請求の範囲第3項に記載の方法。 5.該植物体がトウモロコシである、請求の範囲第1または2項に記載の植物 。 6.該植物体がコムギである、請求の範囲第1または2項に記載の植物。 7.a)該第1および第2受容体ポリペプチドを該植物体中で発現させ; b)該植物体を該第1または第2受容体ポリペプチドのリガンド結合ドメ インと相補的な1またはそれ以上の化学リガンドと接触させ、それによって、該 化学リガンドの存在下、該受容体ポリペプチドが該標的ポリペプチドの発現を活 性化するものである、 ことからなる、請求の範囲第1項に記載の植物体中での遺伝子発現を制御する方 法。 8.該第1および第2受容体ポリペプチドが核内受容体のクラスIIエステロイ ド およびチロイドホルモンスーパーファミリーである、請求の範囲第7項に記載の 方法。 9.該第1受容体ポリペプチドがエクジソン受容体である、請求の範囲第8項 に記載の方法。 10.該第1受容体ポリペプチドが更に異種トランス活性化ドメインを含む、請 求の範囲第9項に記載の方法。 11.該異種トランス活性化ドメインがトウモロコシのC1調節遺伝子由来のト ランス活性化ドメインである、請求の範囲第10項に記載の方法。 12.該第1受容体ポリペプチドが更に異種DNA結合ドメインを含む、請求の 範囲第9項に記載の方法。 13.該DNA結合ドメインが酵母のGAL4タンパク質由来のDNA結合ドメ インである、請求の範囲第12項に記載の方法。 14.該第2受容体ポリペプチドがウルトラスパイラクルである、請求の範囲第 8項に記載の方法。 15.該第2受容体ポリペプチドが更に異種トランス活性化ドメインを含む、請 求の範囲第14項に記載の方法。 16.該異種トランス活性化ドメインが単純ヘルペスのVP16タンパク質由来 のトランス活性化ドメインである、請求の範囲第15項に記載の方法。 17.該第1または第2受容体ポリペプチドがリガンド結合ドメインにおいて突 然変異している、請求の範囲第7項に記載の方法。 18.該化学リガンドが昆虫ホルモン、昆虫ホルモンアンタゴニストまたは昆虫 ホルモンアゴニストである、請求の範囲第7項に記載の方法。 19.該化学リガンドがフェノキシカルブ、CGA 59,205、テブフェナジンま たはRH5849である、請求の範囲第18項に記載の方法。 20.該標的発現カセットがさらに応答配列の1から11コピーを含む5'調節 領域を含んでなる、請求の範囲第7項に記載の方法。 21.a)該第1および第2受容体ポリペプチドを該形質転換植物体中で発現さ せ; b)該形質転換植物体を該第1または第2受容体ポリペプチドのリガンド 結合ドメインと相補的な1またはそれ以上の化学リガンドと接触させ、それによ って、該化学リガンドの存在下、該受容体ポリペプチドが該標的ポリペプチドの 発現を活性化するものである、 ことからなる、請求の範囲第2項に記載の植物の稔性を制御する方法。 22.該受容体発現カセットの少なくとも1つが該受容体ポリペプチドのコード 配列に作動可能に連結した葯特異的プロモーターを含む、請求の範囲第21項に 記載の方法。 23.該受容体発現カセットの少なくとも1つが該受容体ポリペプチドのコード 配列に作動可能に連結した雌蕊特異的プロモーターを含む、請求の範囲第21項 に記載の方法。 24.該標的ポリペプチドの発現が植物稔性を低減させる、請求の範囲第21項 に記載の方法。 25.該標的ポリペプチドがリボヌクレアーゼのバルナーゼである、請求の範囲 第24項に記載の方法。 26.該標的発現カセットが受精に重要なコード配列のアンチセンス版をコード するものである、請求の範囲第21項に記載の方法。 27.該標的ポリペプチドの発現が植物稔性を増大させる、請求の範囲第21項 に記載の方法。 28.該標的ポリペプチドがリボヌクレアーゼ阻害因子のバルスターである、請 求の範囲第27項に記載の方法。 29.a)植物細胞中での発現を促進する能力がある5'調節領域; b)リガンド結合ドメインを含む受容体ポリペプチドをコードする作動可 能に連結したコード配列 c)3'終結配列 を含んでなる、受容体発現カセット。 30.該受容体ポリペプチドが核内受容体のクラスIIステロイドおよびチロイド ホルモンスーパーファミリーである、請求の範囲第29項に記載の受容体発現カ セット。 31.該受容体ポリペプチドがエクジソン受容体である、請求の範囲第30項に 記載の受容体発現カセット。 32.該受容体ポリペプチドが更に異種トランス活性化ドメインを含む、請求の 範囲第30項に記載の受容体発現カセット。 33.該異種トランス活性化ドメインがトウモロコシのC1調節遺伝子由来のト ランス活性化ドメインである、請求の範囲第32項に記載の受容体発現カセット 。 34.該受容体ポリペプチドが更に異種DNA結合ドメインを含む、請求の範囲 第30項に記載の受容体発現カセット。 35.該異種DNA結合ドメインが酵母GAL4タンパク質のDNA結合ドメイ ンである、請求の範囲第34項に記載の受容体発現カセット。 36.該受容体ポリペプチドがウルトラスパイラクルである、請求の範囲第30 項に記載の受容体発現カセット。 37.該受容体ポリペプチドが更に異種トランス活性化ドメインを含む、請求の 範囲第36項に記載の受容体発現カセット。 38.該異種トランス活性化ドメインが単純ヘルペスVP16タンパク質のトラ ンス活性化ドメインである、請求の範囲第37項に記載の受容体発現カセット。 39.該受容体ポリペプチドがリガンド結合ドメインにおいて突然変異している 、請求の範囲第29項に記載の受容体発現カセット。 40.a)受容体ポリペプチドの相補的結合に必要な少なくとも1つの応答配列 を含む5'調節領域; b)標的ポリペプチドをコードする作動可能に連結したヌクレオチド配列 c)3'終結領域 を含んでなる、標的発現カセット。 41.昆虫ホルモン、昆虫ホルモンアンタゴニスト、または昆虫ホルモンアゴニ ストにより標的ポリペプチドの発現を誘導するための請求の範囲第1項に記載の 植物体の使用。 42.植物稔性を制御するための請求の範囲第2項に記載の植物体の使用。
