Claims (1)
- 【特許請求の範囲】 1.複数のDNA配列のライブラリであって、その各配列がウイルス粒子上での 提示のための少なくとも1つの亜鉛フィンガー結合モチーフをコードし、該配列 は、位置−1、+2、+3、+6ならびに位置+1、+5および+8のうち少な くとも1つにおいてアミノ酸がランダムに割当てられた亜鉛フィンガー結合モチ ーフをコードする、DNA配列のライブラリ。 2.複数のDNA配列のライブラリであって、その各配列がウイルス粒子上での 提示のための亜鉛フィンガー結合ポリペプチドの少なくとも中央フィンガーにお ける亜鉛フィンガー結合モチーフをコードし、該配列は、位置−1、+2、+3 および+6においてアミノ酸がランダムに割当てられた結合モチーフをコードす る、DNA配列のライブラリ。 3.該結合モチーフをコードする配列は、位置+1、+5および+8のうちの1 つまたはそれ以上においてさらにアミノ酸がランダムに割当てられている、請求 項2に記載の配列のライブラリ。 4.該結合モチーフをコードする配列は、位置+1、+5および+8においてア ミノ酸がランダムに割当てられている、請求項1、2または3のいずれかに記載 の配列のライブラリ。 5.コードされた該配列は、複数の亜鉛フィンガーを含み、 隣接するフィンガーが介在するリンカーペプチドによって連結される、亜鉛フィ ンガーポリペプチドを備える、前述の請求項のいずれかに記載の配列のライブラ リ。 6.コードされた該配列は、Zif268ポリペプチドの亜鉛フィンガーを含む 、前述の請求項のいずれかに記載の配列のライブラリ。 7.コードされた該配列は、規定されたアミノ酸配列を有する2つまたはそれ以 上の亜鉛フィンガー間に位置付けられ、アミノ酸がランダムに割当てられている 亜鉛フィンガーを含む、前述の請求項のいずれかに記載の配列のライブラリ。 8.バクテリオファージfdの小コートタンパク質との融合物としてクローニン グするのに適する形である、前述の請求項のいずれかに記載の配列のライブラリ 。 9.特定の標的DNA配列に結合するための亜鉛フィンガーポリペプチドを設計 する方法であって、標的DNA配列の少なくとも有効な部分に対して複数の亜鉛 フィンガー結合モチーフの各々をスクリーニングするステップと、標的DNA配 列に結合するモチーフを選択するステップとを含む、方法。 10.2ラウンドまたはそれ以上のスクリーニングが行なわれる、請求項9に記 載の方法。 11.特定の標的DNA配列に結合するための亜鉛フィンガーポリペプチドを設 計する方法であって、複数の亜鉛フ ィンガー結合モチーフの各々の1つまたはそれ以上のDNAトリプレットに対す る結合を比較するステップと、好ましい結合特性の見られるモチーフを選択する ステップとを含む、方法。 12.請求項9または10に記載のスクリーニングステップをさらに最初に含む 、請求項11に記載の方法。 13.標的DNA配列に結合するための亜鉛フィンガーポリペプチドを設計する 方法であって、複数の亜鉛フィンガー結合モチーフを単一の亜鉛フィンガーポリ ペプチド内で組合せるステップを含み、モチーフの各々は請求項9または10に 記載の方法によってスクリーニングされており、かつ/または請求項11または 12に記載の方法によって選択されている、方法。 14.隣接する亜鉛フィンガー結合モチーフ間に介在するリンカーペプチドは、 天然に存在する亜鉛フィンガー結合ポリペプチドにあるもの、または人工のペプ チド配列か、または人工の非アミノ酸リンカーである、請求項13に記載の方法 。 15.請求項9から14のいずれかに記載の方法に従い設計される、標的DNA 配列に結合するための亜鉛フィンガーポリペプチド。 16.64の配列からなるDNAライブラリであって、各配列は請求項11また は12に記載の選択方法で用いるのに適した形態で3つのDNA塩基の64の可 能な順列のう ちどれか1つを含む、ライブラリ。 17.配列は、分離手段に組合わされるか、または組合わせ得るものである、請 求項16に記載のライブラリ。 18.該分離手段は、ミクロタイタープレート、磁気もしくは非磁気ビーズまた は沈降分離可能な粒子、およびアフィニィティクロマトグラフィカラムの1つか ら選択される、請求項17に記載のライブラリ。 19.該配列はビオチニル化されている、請求項16、17または18のいずれ かに記載のライブラリ。 20.該配列は12のミニライブラリ内に含まれる、請求項16から19のいず れかに記載のライブラリ。 21.対象となる核酸配列に結合するための亜鉛フィンガーポリペプチドを作る ためのキットであって、ベクター中にクローニングするのに適した形態の、結合 特性のわかった亜鉛フィンガー結合モチーフをコードするDNA配列のライブラ リと、該ライブラリからの1つまたはそれ以上の配列を受入れるのに適したベク ター分子と、使用のための説明書とを含む、キット。 22.該ベクターは、単一の亜鉛フィンガーポリペプチドとしてクローニングさ れた配列の発現を導くことができる、請求項21に記載のキット。 23.該ベクターはウイルス粒子の表面上に提示される単一の亜鉛フィンガーポ リペプチドとしてクローニングされた配列の発現を導くことができる、請求項2 1または22 に記載のキット。 24.対象となる核酸配列に結合するための亜鉛フィンガーポリペプチドを作る ためのキットであって、請求項9または10に記載の方法に従うスクリーニング 、および/または請求項11または12に記載の方法に従う選択を行なうのに適 した形態の、亜鉛フィンガー結合モチーフを各々がコードするDNA配列のライ ブラリと、使用のための説明書とを含む、キット。 25.DNA配列の該ライブラリは、請求項1から8のいずれかに記載のもので ある、請求項24に記載のキット。 26.請求項16から20のいずれかに従うライブラリをさらに含む、請求項2 4または25に記載のキット。 27.適切な緩衝液および/または結合された亜鉛フィンガーモチーフを検出す るための試薬をさらに含む、請求項24、25または26のいずれかに記載のキ ット。 28.亜鉛フィンガー結合モチーフをコードするDNA配列のライブラリから選 択される1つまたはそれ以上の配列を受入れるのに適したベクターをさらに含む 、請求項24から27のいずれかに記載のキット。 29.標的細胞内で対象となる遺伝子の発現を改変する方法であって、必要に応 じて対象となる遺伝子の構造領域および/または調節領域のDNA配列の少なく とも部分を決定するステップと、決定された配列のDNAに結合するよう亜鉛フ ィンガーポリペプチドを設計するステップと、標 的細胞内に前記亜鉛フィンガーポリペプチドが存在するようにするステップとを 含む、方法。 30.該亜鉛フィンガーポリペプチドは、請求項9から14のいずれかに従い設 計される、請求項29に記載の方法。 31.該亜鉛フィンガーポリペプチドは、1つまたはそれ以上のさらなる機能的 ドメインを含む、請求項29または30に記載の方法。 32.該亜鉛フィンガーポリペプチドは、標的細胞の核へ該亜鉛フィンガーポリ ペプチドを送達するような核位置決定シグナルを含む、請求項29、30または 31のいずれかに記載の方法。 33.該亜鉛フィンガーポリペプチドは、SV40の大型T抗原からの核位置決 定シグナルを含む、請求項29から32のいずれかに記載の方法。 34.該亜鉛フィンガーポリペプチドは、ポリペプチドの細胞内発現を導くDN Aの細胞へのデリバリーによって、該標的細胞内に存在するようにされる、請求 項29から33のいずれかに記載の方法。 35.請求項29から34のいずれかに記載の方法に従い遺伝子の発現を改変す ることによって細胞分裂を阻害する方法であって、該遺伝子が細胞分裂を調節す ることにかかわるものである、細胞分裂を抑制する方法。 36.癌細胞の分裂を可能にする遺伝子の発現を阻害する亜鉛フィンガーポリペ プチドを、患者へ送達するかまたは それが患者の中に存在するようにするステップを含む、癌を治療する方法。 37.亜鉛フィンガーポリペプチドを結合することによって、サンプル混合物に 存在する、対象となる核酸配列を修飾する方法であって、サンプル混合物を対象 となる配列の少なくとも一部分に対して親和性を有する亜鉛フィンガーポリペプ チドと接触させ、該亜鉛フィンガーポリペプチドが対象となる該配列に特異的に 結合できるようにするステップを含む、方法。 38.該亜鉛フィンガーポリペプチドは、請求項9から14のいずれかに記載の 方法に従い設計される、請求項37に記載の方法。 39.サンプルの残部から該亜鉛フィンガーポリペプチド(およびそれに特異的 に結合された核酸配列)を分離するステップをさらに含む、請求項37または3 8に記載の方法。 40.該亜鉛フィンガーポリペプチドは固相支持体に結合される、請求項37、 38または39のいずれかに記載の方法。 41.対象となる該配列に結合される該亜鉛フィンガーポリペプチドの存在は、 1つまたはそれ以上の検出試薬を加えることにより検出される、請求項37から 40のいずれかに記載の方法。 42.対象となる該DNA配列は、アクリルアミドまたは アガロースゲルマトリックスに存在するか、または膜の表面に存在する、請求項 37から41のいずれかに記載の方法。 43.疾病関連遺伝子の発現を阻害することのできる亜鉛フィンガーポリペプチ ド。 44.該ポリペプチドは、天然に存在するものではなく、かつ疾病関連遺伝子の 発現を阻害するべく特異的に設計されている、請求項43に記載の亜鉛フィンガ ーポリペプチド。 45.請求項9から14のいずれかに記載の方法によって設計される、請求項4 3または44に記載の亜鉛フィンガーポリペプチド。 46.癌細胞の発現を阻害することかできる、請求項43、44、または45の いずれかに記載の亜鉛フィンガーポリペプチド。 47.BCR−ABL融合癌遺伝子の発現を阻害することができる、請求項43 から46のいずれかに記載の亜鉛フィンガーポリペプチド。 48.DNA配列GCAGAAGCCに結合するよう設計された、請求項43か ら47のいずれかに記載の亜鉛フィンガーポリペプチド。 49.ras癌遺伝子の発現を阻害することができる、請求項43から46のい ずれかに記載の亜鉛フィンガーポリペプチド。 50.DNA配列GACGGCGCCに結合するよう設計された、請求項49に 記載の亜鉛フィンガーポリペプチド。
