Description
i)配列番号:2で表されるアミノ酸配列を有するタンパク質の活性が不活性化され、プトレシン生産能が向上したコリネバクテリウム属微生物を培地で培養する段階;及び
ii)前記段階で得られる微生物または培地からプトレシンを分離する段階を含む、プトレシンの生産方法を提供する。
1)上記タンパク質をコードするポリヌクレオチドの一部または全体の欠失、
2)上記ポリヌクレオチドの発現が減少するように発現調節配列の変形、
3)上記タンパク質の活性が弱化するように染色体上の上記ポリヌクレオチド配列の変形、または
4)その組み合わせなどを使用して行うことができるが、特にこれに限定されない。
1)上記酵素を暗号化するポリヌクレオチドのコピー数の増加、
2)上記ポリヌクレオチドの発現が増加するように発現調節配列の変形、
3)上記酵素の活性が強化されるように、染色体上のポリヌクレオチド配列の変形、または
4)その組み合わせにより強化されるように変形する方法などにより行うことができるが、これに限定されない。

i)配列番号:2で表されるアミノ酸配列を有するタンパク質の活性が不活性化され、プトレシン生産能が向上したコリネバクテリウム属微生物を培地で培養する段階;
及び
ii)前記段階で得られる微生物または培地からプトレシンを分離する段階を含む、プトレシンの生産方法を提供する。
<1−1> ATCC13032ベースのプトレシン生産菌株からNCgl2523欠損菌株の製作
コリネバクテリウム・グルタミクムATCC13032由来のNCgl2523の欠損がプトレシン生産能に関与するかどうかを確認するために、上記NCgl2523をコードする遺伝子を欠損するためのベクターを作製した。
コリネバクテリウム・グルタミクムATCC13869ベースのプトレシン生産菌株であるDAB12−a(大韓民国公開特許第2013−0082478号)及びDAB12−b(大韓民国公開特許第2013−0082478号; DAB12−a△NCgl1469)を対象にNCgl2523欠損菌株を製作した。
プトレシン生産菌株においてNCgl2523欠損がプトレシン生産に及ぼす効果を確認するために、前記実施例<1−1>及び<1−2>で製作したコリネバクテリウム・グルタミクム変異株を対象にプトレシン生産能を比較した。
NCgl2523活性が不活性化されたコリネバクテリウム・グルタミクム変異株において細胞内プトレシンの濃度変化を確認するために、コリネバクテリウム・グルタミクム変異株KCCM11240P△NCgl2523及び親菌株KCCM11240Pを対象に、有機溶媒を用いた抽出方法で細胞内プトレシン濃度を測定した。細胞内の代謝物質(intracellular metabolite)の分析は、文献(Nakamura J et al., Appl. Environ. Microbiol., 73(14): 4491-4498, 2007)で記載された方法により行った。
KEGG、MetaCyc DatabaseとNCBI blastPを通じてコリネバクテリウム・グルタミクムATCC13032由来のNCgl2523の相同性遺伝子の探索(Ortholog Search)を行い、遺伝子クラスターを通じて各微生物のゲノム上にNCgl2523とオペロンを構成するNCgl2522、 NCgl2524との組み合わせの形態を確認した。
Claims (8)
- プトレシン生産能を有するコリネバクテリウム(Corynebacterium)属微生物において配列番号:2で表されるアミノ酸配列を有するタンパク質の活性が不活性化された、プトレシン生産能が向上したコリネバクテリウム属微生物。
- さらにオルニチンデカルボキシラーゼ(ornithine decarboxylase、ODC)の活性が導入された、請求項1に記載のプトレシン生産能が向上したコリネバクテリウム属微生物。
- 前記ODCが配列番号:10のアミノ酸配列を有する、請求項2に記載のプトレシン生産能が向上したコリネバクテリウム属微生物。
- 前記微生物がさらにアセチルトランスフェラーゼの活性が不活性化された、請求項1に記載のプトレシン生産能が向上したコリネバクテリウム属微生物。
- 前記アセチルトランスフェラーゼが配列番号:15または16で表されるアミノ酸配列を含む、請求項4に記載のプトレシン生産能が向上したコリネバクテリウム属微生物。
- 前記微生物がコリネバクテリウム・グルタミクム(Corynebacterium glutamicum)である、請求項1に記載のプトレシン生産能が向上したコリネバクテリウム属微生物。
- 配列番号:2で表されるアミノ酸配列を有するタンパク質の活性が不活性化され、プトレシン生産能が向上したコリネバクテリウム属微生物を培地で培養する段階; 及び
前記段階で得られる微生物または培地からプトレシンを分離する段階を含む、プトレシン生産方法。 - 前記コリネバクテリウム属微生物がコリネバクテリウム・グルタミクムである、請求項7に記載のプトレシン生産方法。
