本発明は、真皮コラーゲン線維束再構築剤及びそれを含有する、化粧料などの皮膚外用剤に関する。

【従来の技術】
人間は年齢とともにその形態が変化し、老化の兆候を見せる。皮膚に於けるこの様な兆候の一つにしわの形成が挙げられ、この様な兆候の形成の抑制が化粧料の大きなテーマとなっている。この様なしわの形成について、その発生メカニズムとしては1)エラスチンなどの皮膚弾力線維の断裂、断片化などに伴う弾力損失や2)真皮コラーゲン線維束構造の崩壊乃至はその構造の乱れ等があるといわれている。このうち、1)エラスチンなどの皮膚弾力線維の断裂、断片化などに伴う弾力損失については、エラスチンの補給や大豆蛋白などによるエラスチンの断裂の抑制等の対応策が挙げられているが、2)真皮コラーゲン線維束構造の崩壊乃至はその構造の乱れに対しては、僅かにウルソール酸及びその誘導体に真皮コラーゲン線維束の再構築作用が知られているにすぎず、真皮コラーゲン線維束構造を再構築する、新たな真皮コラーゲン線維束再構築剤の開発が望まれていた。

本発明は、この様な状況下為されたものであり、真皮コラーゲン線維束構造を再構築する、新たな真皮コラーゲン線維束再構築剤を提供することを課題とする。

この様な状況に鑑みて、本発明者らは、真皮コラーゲン線維束構造を再構築する、新たな真皮コラーゲン線維束再構築剤を求めて鋭意研究努力を重ねた結果、ハイショウソウのエッセンスにそのような作用を見いだし発明を完成させた。即ち、本発明は次に示す技術に関するものである。
(1) オミナエシ科(Valerianaceae)オトコエシ(Patrinia villosa(THUNB.)JUSS.)のエッセンスからなる真皮コラーゲン線維束再構築剤を含有する、しわの改善或いは予防用の皮膚外用剤。
(2) 前記オトコエシのエッセンスが、全草或いは、葉部、茎部、根部、果実、種子、花から選ばれる1種乃至は2種以上からなる極性溶媒抽出物乃至はその溶媒除去物であることを特徴とする、(1)に記載の皮膚外用剤。

【発明の実施の形態】
(1) 本発明の真皮コラーゲン線維束再構築剤
本発明の真皮コラーゲン線維束再構築剤は、ハイショウソウのエッセンスからなる。ここで、本発明でいうエッセンスとは、植物体それ自身、植物体を乾燥、細切、粉砕など加工した加工物、植物体乃至はその加工物に溶媒を加えて抽出した溶媒抽出物、抽出物から溶媒を除去した抽出物の溶媒除去物、それらをカラムクロマトグラフィーや液液抽出など精製した精製物などの総称を意味する。本発明のエッセンスに適用するハイショウソウとしては、漢方分類でハイショウソウに分類されるものであれば特段の限定無く使用でき、例えば、オミナエシ科(Valerianaceae)オトコエシ(Patrinia villosa (THUNB.)JUSS.)、オミナエシ科(Valerianaceae)オミナエシ(Patrinia scabriosaefolia FISCH.)アブラナ科(Cruciferae)グンバイナズナ(Thlaspi arvense L.),キク科(Compositae)ハチジョウナ(Sonchus brachyotus DC.)キク科(Compositae)タイワンハチジョウナ(Sonchus arvensis L.)、キク科(Compositae)ノゲシ(Sonchus oleraceus L.)、キク科(Compositae)レクセリス チネンシス(lxeris chinensis (THUB.) Versicolor KITAM.)、キク科(Compositae)レクセリス デンチキュラータ(lxeris denticulata STABB.)が好ましく例示でき、更に好ましくはオミナエシ科(Valerianaceae)オトコエシ(Patrinia villosa (THUNB.)JUSS.)又は、オミナエシ科(Valerianaceae)オミナエシ(Patrinia scabiosaefolia FISCH.)である。本発明のエッセンスの調製に用いる植物体の部位としては、植物体のいずれの部位も使用できるが、特に好ましいものは根を付けた全草である。これはこの部位に真皮コラーゲン線維束を再構築する成分が多く含まれているためである。又、本発明のエッセンスとしては、極性溶媒抽出物或いはその溶媒除去物が特に好ましく例示でき、極性溶媒としては、エタノールやメタノールなどのアルコール類、酢酸エチルや蟻酸メチルなどのエステル類、アセトンやメチルエチルケトン等のケトン類、ジエチルエーテルやテトラヒドロフランなどのエーテル類、クロロホルムや塩化メチレン等のハロゲン化炭化水素類、水等から選ばれる1種乃至は2種以上が好ましく例示できる。これらの内、特に好ましいものは、水及びアルコールから選ばれる1種乃至は2種以上である。この様な抽出物乃至はその溶媒除去物は、植物体乃至はその加工物に1〜10倍量の溶媒を加え、室温であれば数日間、沸点付近の温度であれば、数時間浸漬すればよい。溶媒除去は、蒸発乾固、減圧濃縮、凍結乾燥いずれも可能である。もっとも好ましいものは減圧濃縮と凍結乾燥である。本発明の皮膚外用剤に於いては、これら真皮コラーゲン線維束再構築剤は唯一種を含有させることもできるし、二種以上を含有させることもでき、好ましい含有量としては、0.01〜10重量%、更に好ましくは0.1〜5重量%が例示できる。これは、少なすぎると効果を発揮しない場合があり、多すぎても効果が頭打ちになり、処方の自由度が損なわれる場合があるからである。本発明の真皮コラーゲン線維束再構築剤は、老化や光などによって崩れた真皮コラーゲン線維束を再構築し、正常な構造に戻す作用及び真皮コラーゲン線維束構造が光や老化により崩れるのを防ぐ作用を有し、この様な作用によりしわを改善したり、しわが形成するのを抑制したりする作用を有する。本発明の真皮コラーゲン線維束再構築剤がその作用を発揮するしわは、真皮コラーゲン線維束の崩壊や構造の乱れに起因するものに適用するのが好ましい。

(2) 本発明の皮膚外用剤
本発明の皮膚外用剤は、上記真皮コラーゲン線維束再構築剤を含有することを特徴とする。本発明の皮膚外用剤としては、化粧料、皮膚外用医薬組成物などが好ましく例示でき、中でも化粧料に適用するのが特に好ましい。これは本発明の真皮コラーゲン線維束再構築剤の作用が緩和であり、加えて、このものの安全性が極めて高いため、長期連用に好適であるからである。本発明の皮膚外用剤は、含有する真皮コラーゲン線維束再構築剤の効果により、真皮コラーゲン線維束の崩壊又は構造の乱れに起因するしわの改善と予防に優れた作用を発揮する。又、しわに至らないまでも、同様のメカニズムによって生じるキメのあれやはりの低下についても改善或いは予防する作用を示す。言い換えれば、真皮コラーゲン線維束構造の崩壊や構造の乱れに起因して起こる様々な生体に好ましくない事象を予防或いは改善し、美肌を具現化する作用を示す。この様な用途に用いることが本発明の皮膚外用剤に於いては好ましい。特筆すべきは、真皮コラーゲン線維束の崩壊や構造の乱れによって生じるシワへの効果であって、この様な効果を発揮する皮膚外用剤は極めてまれである。従って、本発明の皮膚外用剤は、この様なメカニズムで起こるしわ用に適用することが好ましい。本発明の皮膚外用剤に於いては、前記本発明の真皮コラーゲン線維束再構築剤以外に、通常皮膚外用剤で使用される任意成分を含有することができる。かかる任意成分としては、例えば、スクワラン、ワセリン、マイクロクリスタリンワックス等の炭化水素類、ホホバ油、カルナウバワックス,オレイン酸オクチルドデシル等のエステル類、オリーブ油、牛脂、椰子油等のトリグリセライド類、ジメチコンやフェメチコン、アモジメチコン等のシリコーン類、ステアリン酸、オレイン酸、リチノレイン酸等の脂肪酸、オレイルアルコール、ステアリルアルコール、オクチルドデカノール等の高級アルコール、スルホコハク酸エステルやポリオキシエチレンアルキル硫酸ナトリウム等のアニオン界面活性剤類、アルキルベタイン塩等の両性界面活性剤類、ジアルキルアンモニウム塩等のカチオン界面活性剤類、ソルビタン脂肪酸エステル、脂肪酸モノグリセライド、これらのポリオキシエチレン付加物、ポリオキシエチレンアルキルエーテル、ポリオキシエチレン脂肪酸エステル等の非イオン界面活性剤類、ポリエチレングリコール、グリセリン、1,3−ブタンジオール等の多価アルコール類、キサンタンガム、グァーガム、カルボキシビニルポリマー、カチオン化セルロースエーテル、カチオン化グァーガムなどの増粘・ゲル化剤、酸化防止剤、紫外線吸収剤、香料、色材、防腐剤、粉体等が好ましく例示できる。本発明の皮膚外用剤は、これらの任意成分と必須の成分とを常法に従って処理することにより、本発明の皮膚外用剤は製造することができる。又、上記真皮コラーゲン線維束再構築剤を上記の真皮コラーゲン線維束再構築の目的で含有させなくとも、これらのエッセンスを含有すれば、前記のしわに対する作用が得られるので、この様なエッセンスを含有する真皮コラーゲン線維束の崩壊又は構造の乱れに起因するしわの予防又は改善のための皮膚外用剤も、本発明の皮膚外用剤の技術的範囲に属する。

【実施例】
以下に、実施例を挙げて本発明について更に詳細に説明を加えるが、本発明がこれら実施例にのみ限定されないことはいうまでもない。

<実施例1>
オミナエシ科(Valerianaceae)オトコエシ(Patrinia villosa (THUNB.)JUSS.)の根を付けた全草乾を燥した部分500gに5lのメタノールを加え、室温で1週間浸漬し、濾過して、減圧濃縮し、本発明の真皮コラーゲン線維束再構築剤1を27g得た。このものについて、真皮コラーゲン線維束再構築作用をイン・ビトロで調べた。即ち、3回/週の最小紅斑量の紫外線照射を5週連続して行ったヘアレスマウス(Hr−/Kud,雌)の背部皮膚より、小片を切り出し、トリプシン処理をして皮膚由来の真皮線維芽細胞を採取し、5代継代して用いた。培養条件は5×104(5×10の4乗)個/mlの細胞濃度で、1.0mg/mlの酸可溶性コラーゲンを加え、10%FBS加イーグルの最少培地で10-4(10の−4乗)%の真皮コラーゲン線維束再構築剤1を添加して7日間培養した。図1に0%添加(非添加)の写真(図面代用写真)を図2に10-4(10の−4乗)%添加の写真を示す。図1ではコラーゲン線維が無秩序に分布しているのに対して、図2ではコラーゲン線維が2、3本ずつまとまり、構造を作り始めていることがわかる。これにより、本発明の真皮コラーゲン線維束再構築剤1が、真皮に於いて真皮コラーゲンの再構築を促していることがわかる。

上記実施例2の化粧水と、実施例2の化粧水の真皮コラーゲン線維束再構築剤1を水に置換した比較例1の化粧水に関して、1群5名のしわに悩むパネラーを用いて、長期連用試験(3ヶ月)を行い、しわの改善を著効、効果あり、効果なしの3段階で評価したところ、実施例2の使用テスト群では、2名に著効、2名に効果ありが認められたが、比較例1は2名が効果ありのみであった。これより、本発明の皮膚外用剤である化粧水が、しわの改善に有効であると判断された。又、実施例2の化粧水で効果なしであった1名は、しわの形成メカニズムが、真皮コラーゲン線維束の崩壊や構造の乱れによらないものであることが推測された。
