Abstract
Description
エアロゾルを発生するように構成された多孔質媒体と、多孔質媒体上に液体を分散させ、それにより多孔質媒体を濡らすように構成された移動可能な濡らし機構と、ガスチャネルとを含み、前記多孔質媒体が2つの側面を有し、第1の側面が移動可能な濡らし機構に面している、噴霧器を提供することと、
液体を供給することと、
移動可能な濡らし機構を作動させ、それにより多孔質媒体の前記第1の側面上に液体を分散させることと、
ガスチャネルを圧力源に連結して、圧力勾配を多孔質媒体に生じ、それによって多孔質媒体の第1の側面で、液体の液滴を含む、エアロゾルを発生することとを含む。
エアロゾルを発生するように構成された多孔質媒体と、液体を吸収するように構成された液体吸収材料と、液体吸収材料を多孔質媒体に押し付けるように構成された濡らし機構と、多孔質媒体に圧力勾配をもたらすように構成されたガスチャネルとを含み、多孔質媒体が2つの側面を有し、第1の側面が液体吸収材料に面している、噴霧器を提供することと、
液体を供給することと、
液体吸収材料を液体で濡らすことと、
液体吸収材料を多孔質媒体に押し付けることと、
圧力勾配を多孔質媒体に生じ、それによって多孔質媒体の第1の側面で、液体の液滴を含む、エアロゾルを発生することとを含む。
例1−水のエアロゾル液滴直径の測定
スポンジがある場合または無い場合において、いくつかの実施形態に従った噴霧器を使用して発生した水溶性染料のエアロゾル化された水溶液の累積的な液滴サイズ分布がテストされた。図6(正方形−スポンジ有り;三角形−スポンジ無し)に提示した結果は、液体吸収材料の存在下では、約100%の液滴が直径5ミクロン未満であり、液滴の80%は直径が1ミクロン未満であることを示している。しかし、液体吸収材料がない場合は、液滴の約70%だけが直径5ミクロン未満である。
スポンジがある場合または無い場合において、いくつかの実施形態に従った噴霧器を使用して発生したグリセリン(5%)を含有する水溶性染料のエアロゾル化された水溶液の累積的な液滴サイズ分布がテストされた。図7(正方形−スポンジ有り;三角形−スポンジ無し)に提示した結果は、液体吸収材料の存在下では、約95%の液滴が直径5ミクロン未満であり、液滴の85%は直径が約2ミクロン未満であることを示している。しかし、液体吸収材料がない場合は、液滴の約60%だけが直径5ミクロン未満である。
スポンジの存在下で、いくつかの実施形態に従った噴霧器を使用して発生した工業用ベントリン(登録商標)(5mg/mlアルブテロール)エアロゾルの累積的な液滴サイズ分布がテストされた。図8に提示した結果は、約90%の液滴が直径9ミクロン未満であり、液滴の80%は直径が5ミクロン未満であることを示している。
水溶性染料の異なる水性製剤(製剤1〜7)、ベントリン(商標)およびインスリンに対する累積的な液滴サイズ分布が測定された(図14)−液滴サイズ分布は、15リットル/分の流量で作動する冷却された次世代インパクタ(NGI)を使用して得られた。結果は、空気動力学的直径の質量中央値(MMAD)および幾何学的標準直径(GSD)の値が、それぞれ、約0.4〜7μmおよび約2〜5μmの範囲内で変わることを示している。
異なる生理化学的特性を有する可溶性染料トレーサーを含有する様々な水性製剤(2、5および6)に対する次世代インパクタ(NGI)板上の質量分布が図16に提示されている。製剤2、製剤5および製剤6は、以下の理由で選択された:製剤2は、全身送達に適した非常に小さい液滴を提供し、製剤6は、中央気道への送達に適したサイズで液滴を提供し、製剤5は、経鼻送達に適した大きな液滴を提供する。この結果は、本明細書で開示する噴霧器の利点を強調しており:噴霧器を使用して得られたエアロゾルは、医薬組成物を呼吸器系の様々な領域に投与するために使用され得る。
ベントリン(商標)およびインスリンの製剤に対する、累積的なサイズ分布プロットが、NGIを使用して測定された(図17)。図に示すように、ベントリン(商標)に対して得られたMMADは、約2.5ミクロンであって、気管支拡張薬の中央気道への送達に資する。他方、インスリンに対するMMADは、1ミクロンよりも低くて、これは、深肺への送達、従って、全身摂取に資する。
Claims (49)
- エアロゾルを発生するように構成された多孔質媒体と、液体を前記多孔質媒体上に分散させ、それにより前記多孔質媒体を濡らすように構成された移動可能な濡らし機構と、前記多孔質媒体に圧力勾配をもたらすように構成されたガスチャネルとを含む噴霧器。
- 前記移動可能な濡らし機構が、回転可能な細長部材を含む、請求項1に記載の噴霧器。
- 前記回転可能な細長部材が、前記多孔質媒体の表面を横切って移動し、それにより前記液体を前記表面上に均一的または半均一的に分散させるように構成される、請求項1に記載の噴霧器。
- 前記回転可能な細長部材が軸方向に移動可能である、請求項1に記載の噴霧器。
- 前記回転可能な細長部材が、前記多孔質媒体の前記表面のほぼ全体にわたって移動可能である、請求項1に記載の噴霧器。
- 前記移動可能な濡らし機構が、前記回転可能な細長部材を移動させるか、またはその前記移動を引き起こすように構成されたアクチュエータをさらに含む、請求項2に記載の噴霧器。
- 前記回転可能な細長部材が第1の磁石を含み、かつ前記アクチュエータが、前記第1の磁石と磁気的に関連付けられた、第2の磁石を含み、この特徴によって、前記第2の磁石を動かすことにより、前記回転可能な細長部材の移動が引き起こされる、請求項6に記載の噴霧器。
- 前記第1および/または第2の磁石が複数の磁石を含む、請求項7に記載の噴霧器。
- 前記複数の磁石のうちの1つ以上が電磁石を含む、請求項8に記載の噴霧器。
- 前記アクチュエータが、前記回転可能な細長部材を移動させるように構成されたモーターを含む、請求項6に記載の噴霧器。

- 前記多孔質媒体が複数の細孔を含み、かつ、前記回転可能な細長部材が、前記液体の少なくとも一部を、前記多孔質媒体の前記複数の細孔のうちの少なくとも一部に押し込むように構成される、請求項2に記載の噴霧器。
- 前記エアロゾルを被験者の呼吸器系に送達するように構成された開口部をさらに含む、請求項1に記載の噴霧器。
