Claims (1)
- 【特許請求の範囲】 1.物質の微生物による製造方法であって、これらの物質を生産している微生物 においてトランスアルドラーゼの活性が増加され、この微生物におけるホスホエ ノールピルベート(PEP)に依存する糖摂取系の活性が存在するか、減らされ るかまたは不在であるところの方法。 2.物質の製造が、トランスアルドラーゼ活性の増加に先立ち、予めホスホトラ ンスフェラーゼ系を有する株(pts+株)から、スウィッチを切られたホスホトラ ンスフェラーゼ系を有する株(pts-株)として選択されなかった微生物において なされる請求項1記載の方法。 3.その合成においてペントースホスフェート経路の少なくとも1の中間体が含 まれるところの物質が製造される請求項1または2記載の方法。 4.中間体がエリトロース 4-ホスフェート(Ery4P)である請求項3記載の方法。 5.大腸菌(Escherichia coli)トランスアルドラーゼの活性が増加される請求 項1または4記載の方法。 6.大腸菌(Escherichia coli)トランスアルドラーゼB(TalB)の活性が増加さ れる請求項1〜5のいずれか1項記載の方法。 7.トランスケトラーゼの活性が追加的に増加される請求項1〜6のいずれか1 項記載の方法。 8.大腸菌(Escherichia coli)トランスケトラーゼの活性が増加される請求項 7記載の方法。 9.大腸菌(Escherichia coli)トランスケトラーゼA(TktA) の活性が増加される請求項7または8記載の方法。 10.PEPに依存しない糖の摂取のための輸送タンパク質の活性および/または 関連する糖をホスホリル化するキナーゼの活性が、追加的に増加される請求項1 〜9のいずれか1項記載の方法。 11.輸送タンパク質が促進剤である請求項10記載の方法。 12.促進剤が、ザイモモナス モビリス(Zymomonas mobilis)グルコース促進剤 タンパク質(Glf)である請求項11記載の方法。 13.キナーゼが、ヘキソースをホスホリル化するキナーゼである請求項10〜12の いずれか1項記載の方法。 14.キナーゼが、ザイモモナス モビリス(Zymomonas mobilis)から誘導される請 求項13記載の方法。 15.キナーゼが、ザイモモナス モビリス(Zymomonas mobilis)グルコキナーゼ(G lk)である請求項14記載の方法。 16.PEPに依存する糖摂取系(もし、この系があるならば)の活性が追加的に 減少され、またはスウィッチを切られる請求項10〜15のいずれか1項記載の方法 。 17.トランスアルドラーゼ、トランスケトラーゼ、輸送タンパク質およびキナー ゼのうち少なくとも1の成分の活性が、 a)遺伝子の導入によって、 b)および/または遺伝子コピー数を増加することによって、 c)および/または遺伝子発現を増加することによって、 d)および/または上記酵素の内在性の活性を増加することによって。 e)および/または酵素の構造を変えることによって、 f)および/または調節解除された酵素を使用することによって、 g)および/または調節解除された酵素をコードする遺伝子を挿入することによ って 増加される請求項1〜16のいずれか1項記載の方法。 18.1または複数の遺伝子を、1またはいくつかの遺伝子構造物中に組込むこと によって活性の増加が達成され、ここで1または複数の遺伝子は、単一のコピー としてまたは増加されたコピー数で、遺伝子構造物中へ導入される請求項17記載 の方法。 19.物質の合成に追加的に関与する1以上の酵素が調節解除されおよび/または 増加された活性を示すところの微生物が使用される請求項1〜18のいずれか1項 記載の方法。 20.製造される物質が芳香族アミノ酸である請求項1〜19のいずれか1項記載の 方法。 21.芳香族アミノ酸がL-フェニルアラニンである請求項20記載の方法。 22.使用される微生物が、エシェリヒア(Escherichia)属、セラティア(Serratia )属、バチルス(Bacillus)属、コリネバクテリウム(Corynebacterium)属またはブ レビバクテリウム(Brevibacterium)属に属する請求項1〜21のいずれかl1項記 載の方法。 23.微生物が大腸菌(Escherichia coli)である請求項22記載の方法。 24.組換え形で、 a)トランスアルドラーゼをコードする遺伝子、またはトランスアルドラーゼを コードする遺伝子およびトランスケトラーゼをコードする遺伝子、および b)PEPに依存しない糖の摂取のための輸送タンパク質をコードする、または 糖をホスホリル化するキナーゼをコードする、少なくとも1の遺伝子 を含む遺伝子構造物。 25.輸送タンパク質のための遺伝子が促進剤をコードし、キナーゼのための遺伝 子がヘキソースをホスホリル化するキナーゼをコードする請求項24記載の遺伝子 構造物。 26.トランスアルドラーゼおよびトランスケトラーゼのための遺伝子が大腸菌( Escherichia coli)から誘導され、かつ輸送タンパク質およびキナーゼのための 遺伝子がザイモモナスモビリス(Zymomonas mobilis)から誘導される請求項24ま たは25記載の遺伝子構造物。 27.大腸菌(Escherlchia coli)トランスアルドラーゼのための遺伝子がtalBで あり、かつ大腸菌(Escherichia coli)トランスケトラーゼのための遺伝子がtk tAであり、およびザイモモナス モビリス(Zymomonas mobilis)輸送タンパク質 のための遺伝子がglfであり、かつザイモモナス モビリス(Zymomonas mobilis) キナーゼのための遺伝子がglkである請求項24〜26のいずれか1項記載の遺伝子 構造物。 28.遺伝子構造物が、遺伝子の1つに割り当てられた少なくとも1の調節遺伝子 配列を含む請求項24〜27のいずれか1項記載の遺伝子構造物。 29.遺伝子構造物中の遺伝子の少なくとも1が、それが、誘導可能なプロモータ ーの制御下にあるように組込まれている請求項28記載の遺伝子構造物。 30.複製可能な形で請求項24〜29のいずれか1項記載の遺伝子構造物を有する形 質転換された細胞。 31.細胞において、物質の合成に追加的に関与する1以上の酵素が調節解除され ているおよび/または増加された活性を示す請求項30記載の形質転換された細胞 。 32.細胞が、大腸菌(Escherichia coli)細胞である請求項30または31記載の形 質転換された細胞。 33.PEPに依存する糖摂取系(もし、この系があるならば)の活性が減少され 、またはスウィッチを切られる請求項30〜32のいずれか1項記載の形質転換され た細胞。 34.芳香族アミノ酸を生産することができる請求項30〜33のいずれか1項記載の 形質転換された細胞。 35.芳香族アミノ酸がL-フェニルアラニンである請求項34記載の形質転換された 細胞。 36.請求項29記載の遺伝子構造物が存在する、請求項30〜35のいずれか1項記載 の形質転換された細胞を培養し、最も初期の2細胞分裂後に誘導がなされる請求 項1〜23のいずれか1項記載の物質の微生物による製造方法。 37.ペントースホスフェート経路の中間体の他に、形質転換された細胞が中心の 代謝の他の代謝産物をまた、増大された入手可能性で含む請求項36記載の方法 。
