- 【特許請求の範囲】 1.一般式(I) [式中、 R1はS、N及び/又はOより成る群からの最高で3個の複素原子を有する飽 和もしくは芳香族5−もしくは6−員複素環を示し、それは窒素原子を介して結 合していることができそしてそれは場合によりアミノ、アジド、ホルミル、メル カプチル、カルボキシル、ヒドロキシル、それぞれ炭素数が最高で6の直鎖状も しくは分枝鎖状のアシル、アルコキシ、アルキルチオ又はアルコキシカルボニル 、ニトロ、シアノ、ハロゲン、フェニルあるいはそれ自身がヒドロキシル、アミ ノ、アジド、カルボキシル、それぞれ炭素数が最高で5の直鎖状もしくは分枝鎖 状のアシル、アルコキシ、アルコキシカルボニルもしくはアシルアミノにより又 は式−OR4の基により置換されていることができる炭素数が最高で6の直鎖状 もしくは分枝鎖状のアルキルにより最高で3回同一に又は異なって置換されてい ることができ、 ここで R4は炭素数が最高で5の直鎖状もしくは分枝鎖状アシル又は式−SiR5R6 R7の基を示し、 ここで R5、R6及びR7は同一もしくは異なり、炭素数が6〜10のアリール又は 炭素数が最高で6のアルキルを示し、 ならびに/あるいは式 又は−S(O)c−NR9R10の基により置換されており、 ここで a、b及びb’は同一もしくは異なり、0、1、2もしくは3の数を示し、 R8は水素又は炭素数が最高で4の直鎖状もしくは分枝鎖状アルキルを示し、 cは1もしくは2の数を示し、 R9及びR10は同一もしくは異なり、水素又は炭素数が最高で10の直鎖状も しくは分枝鎖状アルキルを示し、それは場合により炭素数が3〜8のシクロアル キルにより又はそれ自身がハロゲンにより置換されていることができる炭素数が 6〜10のアリールにより置換されていることができるか、あるいは 場合によりハロゲンにより置換されていることができる炭素数が6〜10のア リールを示すか、あるいは 炭素数が3〜7のシクロアルキルを示すか、あるいは R9及びR10は窒素原子と一緒になって5−〜7−員の飽和複素環を形成し、 それは場合によりさらなる酸素原子又は基−NR11を含有することができ、ここ で R11は水素、炭素数が最高で4の直鎖状もしくは分枝鎖状アルキル又は式 の基を示すかあるいはベンジル又はフェニルを示し、ここで環系は場合によりハ ロゲンにより置換されていることができ、 R2及びR3は二重結合を含んでN、S及び/又はOより成る群からの最高で3 個の複素原子を有する6−員の飽和もしくは芳香族複素環を形成し、それは場合 によりホルミル、カルボキシル、ヒドロキシル、メルカプチル、それぞれ炭素数 が最高で6の直鎖状もしくは分枝鎖状のアシル、アルキルチオもしくはアルコキ シカルボニル、ニトロ、シアノ、ハロゲン又はそれぞれ炭素数が最高で6であり 、それ自身がヒドロキシル、アミノ、カルボキシル、それぞれ炭素数が最高で5 の直鎖状もしくは分枝鎖状のアシル、アルコキシもしくはアルコキシカルボニル により置換されていることができる直鎖状もしくは分枝鎖状のアルキル又はアル コキシにより最高で3回同一にもしくは異なって置換されていることができなら びに/あるいは複素環は場合により式−NR12R13又は−S(O)c'NR9'R10' の基により置換されていることができ、ここで R12及びR13は同一又は異なり、水素又は炭素数が最高で6の直鎖状もしくは 分枝鎖状アルキルを示すか、あるいは R12は水素を示し、 R13はホルミルを示し、 c'、R9'及びR10'は上記のc、R9及びR10の意味を有し、これらと同一又 は異なっており、 ならびに/あるいは複素環は場合によりフェニルにより置換されていることがで き、それ自身はハロゲンにより又はそれぞれ炭素数が最高で6の直鎖状もしくは 分枝鎖状のアルキルもしくはアルコキシにより最高で 2回同一にもしくは異なって置換されていることができ、 ならびに/あるいは複素環は場合により式−N=CH−NR14R15の基により置 換されていることができ、ここで R14及びR15は同一又は異なり、水素、フェニル又は炭素数が最高で6の直鎖 状もしくは分枝鎖状アルキルを示し、 AはS、N及び/又はOより成る群からの最高で3個の複素原子を有する5− もしくは6−員の芳香族もしくは飽和複素環又はフェニルを示し、そのそれぞれ は場合によりアミノ、メルカプチル、ヒドロキシル、ホルミル、カルボキシル、 それぞれ炭素数が最高で6の直鎖状もしくは分枝鎖状のアシル、アルキルチオ、 アルキルオキシアシル、アルコキシもしくはアルコキシカルボニル、ニトロ、シ アノ、トリフルオロメチル、アジド、ハロゲン、フェニル又は炭素数が最高で6 であり、それ自身がヒドロキシル、カルボキシル、それぞれ炭素数が最高で5の 直鎖状もしくは分枝鎖状のアシル、アルコキシもしくはアルコキシカルボニルに より置換されていることができる直鎖状もしくは分枝鎖状アルキルにより最高で 3回同一にもしくは異なって置換されていることができ、 ならびに/あるいは式−(CO)d−NR16R17の基により置換されており、 ここで dは0又は1の数を示し、 R16及びR17は同一又は異なり、水素、フェニル、ベンジル又はそれぞれ炭素 数が最高で5の直鎖状もしくは分枝鎖状のアルキルもしくはアシルを示す] の置換ピラゾール誘導体ならびにその異性体及び塩。 2.R1がピリミジニル、ピリダジニル、ピリジル、ピラジニル、テトラヒドロ ピラニル、テトラヒドロフラニル、ピロリル、フリル、チエニル、イミダゾリル 、オキサゾリル、チアゾリル、1,2,3−トリアゾリル、ピラゾリル、オキサ ジアゾリル、チアジアゾリル、イソオキサゾリル、イソチアゾリル、ピラニル又 はモルホリニルを示し、それらのそれぞれは場合によりアミノ、ホルミル、メル カプチル、カルボキシル、ヒドロキシル、それぞれ炭素数が最高で5の直鎖状も しくは分枝鎖状のアシル、アルコキシ、アルキルチオもしくはアルコキシカルボ ニル、ニトロ、シアノ、アジド、フッ素、塩素、臭素、フェニル又はそれ自身が ヒドロキシル、アミノ、アジド、カルボキシル、それぞれ炭素数が最高で4の直 鎖状もしくは分枝鎖状のアシル、アルコキシ、アルコキシカルボニルもしくはア シルアミノにより又は式−OR4の基により置換されていることができる炭素数 が最高で5の直鎖状もしくは分枝鎖状アルキルにより最高で3回同一にもしくは 異なって置換されていることができ、 ここで R4は炭素数が最高で4の直鎖状もしくは分枝鎖状アシルを示し、 ならびに/あるいは式 又は−S(O)c−NR9R10の基により置換されており、 ここで a、b及びb’は同一もしくは異なり、0、1、2もしくは3の数を示し、 R8は水素又は炭素数が最高で3の直鎖状もしくは分枝鎖状アルキル を示し、 cは1もしくは2の数を示し、 R9及びR10は同一もしくは異なり、水素又は炭素数が最高で9の直鎖状もし くは分枝鎖状アルキルを示し、それは場合によりシクロプロピル、シクロブチル 、シクロペンチル、シクロヘキシル、シクロヘプチル又はナフチルもしくはフェ ニルにより置換されていることができ、それ自身はフッ素もしくは塩素により置 換されていることができるか、あるいは フェニル又はナフチルを示し、そのそれぞれは場合によりフッ素又は塩素によ り置換されていることができるか、あるいは シクロプロピル、シクロブチル、シクロペンチル、シクロヘキシル又はシクロ ヘプチルを示すか、あるいは R9及びR10は窒素原子と一緒になってモルホリン環又は式 の基を形成し、ここで あるいはベンジルもしくはフェニルを示し、ここで環系は場合によりフッ素又は 塩素により置換されていることができ、 R2及びR3が二重結合を含んでピリジル、ビリミジニル、ピラジニル又はビリ ダジニル環を形成し、そのそれぞれは場合によりホルミル、カルボキシル、ヒド ロキシル、メルカプチル、それぞれ炭素数が最高で5の直鎖状もしくは分枝鎖状 のアシル、アルキルチオ又はアルコキシカルボニル、ニトロ、シアノ、アジド、 フッ素、塩素、臭素又はそれ自身が ヒドロキシル、アミノ、カルボキシル、それぞれ炭素数が最高で4の直鎖状もし くは分枝鎖状のアシル、アルコキシもしくはアルコキシカルボニルにより置換さ れていることができるそれぞれ炭素数が最高で5の直鎖状もしくは分枝鎖状のア ルキルもしくはアルコキシにより最高で3回同一にもしくは異なって置換されて いることができ、 ならびに/あるいは上記の複素環は場合により式−NR12R13又は−S(O)c'N R9'R10'の基により置換されていることができ、ここで R12及びR13は同一又は異なり、水素又は炭素数が最高で4の直鎖状もしくは 分枝鎖状アルキルを示すか、あるいは R12は水素を示し、 R13はホルミルを示し、 c'、R9'及びR10'は上記のc、R9及びR10の意味を有し、これらと同一又 は異なっており、 ならびに/あるいは上記の複素環は場合によりフェニルにより置換されているこ とができ、それ自身はフッ素、塩素、臭素又はそれぞれ炭素数が最高で4の直鎖 状もしくは分枝鎖状のアルキルもしくはアルコキシにより置換されていることが でき、 ならびに/あるいは上記の複素環は場合により式 の基により置換されていることができ、ここで R14及びR15は水素又は炭素数が最高で4の直鎖状もしくは分枝鎖状アルキル を示し、 Aがチエニル、テトラヒドロピラニル、テトラヒドロフラニル、フェニル、モ ルホリニル、ピリミジル、ピラジニル、ピリダジニル又はピリ ジルを示し、そのそれぞれは場合によりヒドロキシル、ホルミル、カルボキシル 、それぞれ炭素数が最高で4の直鎖状もしくは分枝鎖状のアシル、アルキルチオ 、アルコキシアシル、アルコキシもしくはアルコキシカルボニル、フッ素、塩素 、臭素、ニトロ、シアノ、トリフルオロメチル又はそれ自身がヒドロキシル、カ ルボキシル、それぞれ炭素数が最高で4の直鎖状もしくは分枝鎖状のアシル、ア ルコキシもしくはアルコキシカルボニルにより置換されていることができるそれ ぞれ炭素数が最高で4の直鎖状もしくは分枝鎖状アルキルにより最高で2回同一 にもしくは異なって置換されていることができ、 ならびに/あるいは式−(CO)d−NR16R17の基により置換されており、 ここで dは0又は1の数を示し、 R16及びR17は同一もしくは異なり、水素、フェニル、ベンジル又はそれぞれ 炭素数が最高で4の直鎖状もしくは分枝鎖状のアルキル又はアシルを示す 請求の範囲第1項に記載の一般式(I)の化合物ならびにその異性体及び塩。 3.R1がイミダゾリル、フリル、ピリジル、ピロリル、ピラジニル、ピリミジ ル、イソオキサゾリル、オキサゾリル又はチアゾリルを示し、それらのそれぞれ は場合によりホルミル、フッ素、塩素、アミノ、メルカプチル、シアノ、それぞ れ炭素数が最高で4の直鎖状もしくは分枝鎖状のアシル、アルキルチオ、アルコ キシもしくはアルコキシカルボニル又はそれ自身がヒドロキシル、カルボキシル 、アミノ、アジド、それぞ れ炭素数が最高で3の直鎖状もしくは分枝鎖状のアシル、アルコキシ、アルコキ キシル、ヒドロキシル、それぞれ炭素数が最高で4の直鎖状もしくは分枝鎖状の アシル、アルコキシ、アルキルチオ又はアルコキシカルボニル、ニトロ、シアノ 、フッ素、塩素又はそれ自身がヒドロキシル、アミノ、カルボキシル、それぞれ 炭素数が最高で3の直鎖状もしくは分枝鎖状のアシル、アルコキシもしくはアル コキシカルボニルにより置換されていることができるそれぞれ炭素数が最高で3 の直鎖状もしくは分枝鎖状のアルキルもしくはアルコキシにより最高で3回同一 にもしくは異なって置換されていることができ、 ならびに/あるいは複素環は場合によりアミノ、N,N−ジメチルアミノにより 又は式−NH−CHOもしくは−N=CH−N(CH3)2の基により及び/又は フェニルにより置換されていることができ、それ自身は式−O(CH2)2−CH3 の基により置換されていることができ、 Aがテトラヒドロピラニル、フェニル、ピリミジル、チエニル又はピリジルを 示し、そのそれぞれは場合によりホルミル、カルボキシル、それぞれ炭素数が最 高で3の直鎖状もしくは分枝鎖状のアシル、アルキルチオ、アルキルオキシアシ ル、アルコキシもしくはアルコキシカルボニル、フッ素、塩素、臭素、ニトロ、 シアノ、トリフルオロメチル又はそれ自身がヒドロキシル、カルボキシル、それ ぞれ炭素数が最高で3の直鎖状もしくは分枝鎖状のアシル、アルコキシもしくは アルコキシカルボニルにより置換されていることができる炭素数が最高で3の直 鎖状もし くは分枝鎖状アルキルにより最高で2回同一にもしくは異なって置換されている ことができる請求の範囲第1項に記載の一般式(I)の化合物ならびにその異性 体及び塩。 4.R2及びR3のもとに定義される複素環の種々の意味に依存して、 [A]一般式(II) R1−D (II) [式中、 R1は上記で示した意味を有し、 Dは式 の基を示し、 ここで R18はC1−C4−アルキルを示す] の化合物を不活性溶媒中で、適宜塩基の存在下に一般式(III) A−CH2−NH−NH2 (III) [式中、 Aは上記で示した意味を有する] の化合物と反応させることにより一般式(IV)又は(IVa)[式中、 A及びR1は上記で示した意味を有する] の化合物に転化し、 一般式(IVa)の化合物の場合には、カルボン酸、ニトリル、ホルムアミド又 はグアニジニウム塩を用いて環化し、 一般式(IV)の化合物の場合には、酸の存在下及び適宜マイクロ波下で、1, 3−ジカルボニル誘導体、その塩、互変異性体、エノールエーテルもしくはエナ ミンを用いて環化するか、 あるいは [B]R2及びR3が一緒になってピラジン環を形成する場合、一般式(IV)の 化合物を最初にニトロソ化により一般式(V) [式中、 A及びR1は上記で示した意味を有する] の化合物に転化し、 第2段階で、還元により一般式(VI) [式中、 A及びR1は上記で示した意味を有する] の化合物を製造し、 最後に、1,2−ジカルボニル化合物、好ましくはグリオキサール水溶液を用い て環化するか、 あるいは [C]一般式(VII) [式中、 A1、R2及びR3は上記で示した意味を有し、 Lは式−SnR19R20R21、ZnR22の基、ヨウ素、臭素又はトリフレートを示 し、 ここで R19、R20及びR21は同一又は異なり、炭素数が最高で4の直鎖状もしくは分 枝鎖状アルキルを示し、 R22はハロゲンを示す] の化合物を一般式(VIII) R1−T (VIII) [式中、 R1は上記で示した意味を有し、 L=SnR19R20R21又はZnR22の場合、 Tはトリフレート又はハロゲン、好ましくは臭素を示し、 L=ヨウ素、臭素又はトリフレートの場合、 Tは式SnR19'R20'R21'、ZnR22'又はBR23'R24'の基を示し、 ここで R19'、R20'、R21'及びR22'は上記で示したR19、R20、R21及びR22の意 味を有し、それらと同一又は異なり、 R23'及びR24'は同一又は異なり、ヒドロキシル、炭素数が6〜10のアリー ルオキシ又はそれぞれ炭素数が最高で5の直鎖状もしくは分枝鎖状のアルキル又 はアルコキシを示すか、あるいは一緒になって5−もしくは6−員の炭素環式環 を形成する] の化合物と、不活性溶媒中で、適宜塩基の存在下においてパラジウム−触媒反応 において反応させるか、 であり、 ここで R25は場合によりハロゲンで置換されていることができる(C1−C6)−アル キルを示す場合、 一般式(IX) [式中、 A、R2及びR3は上記で示した意味を有する] の化合物をNaOCO−CH3/N−メチルピロリジンの系中で直接又は式(X )[式中、 R25は上記で示した意味を有する] の化合物との反応により一般式(Ia) [式中、 R2、R3、A及びR25は上記で示した意味を有する] の化合物に転化し、 次いで、メタノール中における水酸化カリウムの作用によりアセチル基を除去す るか、 あるいは 最初に、一般式(IX)の化合物と式(X)の化合物の反応により一般式(XI ) [式中、 R2、R3、A及びR25は上記で示した意味を有する] の化合物を製造し、 さらなる段階において、水酸化カリウムの作用によりヒドロキシメチル化合物を 製造し、 適宜、通常の方法に従うアルキル化により対応するアルコキシ化合物に転化し、 −S(O)cNR9R10及び−S(O)c'NR9'R10'の基の場合、一般式(I)の非 置換化合物から出発して、最初にチオニルクロリドとそして第2段階で適したア ミンと反応させ、 適宜、R1、R2、R3及び/又はAのもとに挙げた置換基を通常の方法に従って 、好ましくは塩素化、接触水素化、還元、酸化、保護基の除去及び/又は求核置 換により変化させるか又は導入することを特徴とする請求の範囲第1項に記載の 一般式(I)の化合物の製造法。 5.請求の範囲第1項に記載の一般式(I)の化合物の少なくとも1種を含む薬 剤。 6.請求の範囲第1項に記載の式(I)の化合物の少なくとも1種を、適宜通常 の助剤及び添加剤を用いて、適した投与形態に転換することを特徴とする薬剤の 製造法。 7.請求の範囲第1項に記載の一般式(I)の化合物の少なくとも1種を有機ナ イトレート又はNOドナーと組み合わせて含む薬剤。 8.請求の範囲第1項に記載の一般式(I)の化合物の少なくとも1種をサイク リックグアノシン−リン酸(cGMP)の分解を阻害する化合物と組み合わせて 含む薬剤。 9.心臓血管障害の処置のための薬剤の製造における請求の範囲第1項に記載の 一般式(I)の化合物の使用。 10.血栓塞栓性障害及び虚血の処置のための薬剤の製造における請求の範囲第 1項に記載の一般式(I)の化合物の使用。
