Claims (18)
1. Izolirani ili rekombinantni polipeptid koji sadrži polipeptidnu sekvencu koja se razlikuje od SEQ ID NO:36 u ne više od 6 aminokiselinskih ostataka i koja uključuje ostatke na položajima 24, 30, 32, 41, 50, 54, 55, 56, 64, 65, 70, 85, 104 i 106 koji su identični sa aminokiselinskim ostatcima na odgovarajućim položajima u SEQ ID NO:36, naznačen time da polipeptid veže humani CD80 ili humani CD86 ili izvanstaničnu domenu bilo kojeg od njih i ima sposobnost inhibicije imunološkog odgovora koja je veća od sposobnosti inhibicije imunološkog odgovora polipeptida LEA29Y koji sadrži polipeptidnu sekvencu koja je prikazana u SEQ ID NO:168.
2. Polipeptid prema zahtjevu 1, naznačen time da se polipeptid razlikuje od SEQ ID NO:36 u ne više od 1, 2, 3, 4, ili 5 aminokiselinskih ostataka.
3. Polipeptid prema zahtjevu 1, naznačen time da polipeptid sadrži polipeptidnu sekvencu koja je prikazana u SEQ ID NO:36.
4. Izolirani ili rekombinantni dimerni polipeptid koji sadrži dva polipeptida prema bilo kojem od zahtjeva 1-3, naznačen time da dimer ima veću sposobnost za suzbijanje imunološkog odgovora nego dimer koji sadrži dva polipeptida LEA29Y, pri čemu svaki polipeptid LEA29Y sadrži polipeptidnu sekvencu koja je prikazana u SEQ ID NO:168.
5. Polipeptid prema zahtjevu 1, koji je izolirani ili rekombinantni fuzijski protein naznačen time da sadrži (a) polipeptid koji sadrži polipeptidnu sekvencu koja se razlikuje od SEQ ID NO:36 u ne više od 6 aminokiselinskih ostataka i sadrži ostatke na položajima 24, 30, 32, 41, 50, 54, 55, 56, 64, 65, 70, 85, 104 i 106 koji su identični aminokiselinskim ostatcima na odgovarajućim položajima u SEQ ID NO:36 i (b) drugi polipeptid, pri čemu je drugi polipeptid Ig Fc polipeptid, te pri čemu fuzijski protein ima veću sposobnost za suzbijanje imunološkog odgovora nego LEA29Y-Ig fuzijski protein, pri čemu LEA29Y-Ig fuzijski protein sadrži polipeptidnu sekvencu prema SEQ ID NO:166.
6. Fuzijski protein prema zahtjevu 5, naznačen time da fuzijski protein sadrži polipeptidnu sekvencu koja je prikazana u SEQ ID NO:197 ili 211.
7. Dimerni polipeptid prema zahtjevu 4, koji je izolirani ili rekombinantni dimerni fuzijski protein koji sadrži dva monomerna fuzijska proteina povezana preko barem jedne disulfidne veze koja je formirana između dva cisteinska ostatka koji se nalaze u svakom monomernom mutiranom fuzijskom proteinu, naznačen time da svaki monomerni fuzijski protein sadrži (a) polipeptid sa polipeptidnom sekvencom koja se razlikuje od SEQ ID NO:36 u ne više od 6 aminokiselinskih ostataka i koja sadrži ostatke na položajima 24, 30, 32, 41, 50, 54, 55, 56, 64, 65, 70, 85, 104 i 106 koji su identični aminokiselinskim ostatcima na odgovarajućim položajima u SEQ ID NO:36 i (b) Ig Fc polipeptid, pri čemu dimerni fuzijski protein ima veću sposobnost za suzbijanje imunološkog odgovora nego dimerni fuzijski protein LEA29Y-Ig, pri čemu dimerni fuzijski protein LEA29Y-Ig sadrži dva monomerna fuzijska proteina LEA29Y-Ig koji svaki sadrži polipeptidnu sekvencu prema SEQ ID NO:166.
8. Dimerni fuzijski protein prema zahtjevu 7, naznačen time da svaki monomerni fuzijski protein sadrži polipeptidnu sekvencu koja je prikazana u SEQ ID NO:197 ili 211.
9. Izolirana ili rekombinantna nukleinska kiselina naznačena time da sadrži polinukleotidnu sekvencu koja kodira polipeptid prema bilo kojem od zahtjeva 1-3, dimerni polipeptid prema zahtjevu 4, fuzijski protein prema zahtjevu 5 ili zahtjevu 6, ili dimerni fuzijski protein prema zahtjevu 7 ili zahtjevu 8, ili njihovu komplementarnu polinukleotidnu sekvencu.
10. Vektor naznačen time da sadrži nukleinsku kiselinu prema zahtjevu 9.
11. Izolirana ili rekombinantna stanica domaćin naznačena time da sadrži:
(a) polipeptid prema bilo kojem od zahtjeva 1-3;
(b) dimerni polipeptid prema zahtjevu 4;
(c) fuzijski protein prema zahtjevu 5 ili zahtjevu 6;
(d) dimerni fuzijski protein prema zahtjevu 7 ili zahtjevu 8;
(e) nukleinsku kiselinu prema zahtjevu 9; i/ili
(f) vektor prema zahtjevu 10.
12. Farmaceutski pripravak naznačen time da sadrži farmaceutski prihvatljivu pomoćnu tvar ili farmaceutski prihvatljiv nosač i jedno ili više od slijedećeg:
(a) polipeptid prema bilo kojem od zahtjeva 1-3;
(b) dimerni polipeptid prema zahtjevu 4;
(c) fuzijski protein prema zahtjevu 5 ili zahtjevu 6;
(d) dimerni fuzijski protein prema zahtjevu 7 ili zahtjevu 8;
(e) nukleinsku kiselinu prema zahtjevu 9;
(f) vektor prema zahtjevu 10; i/ili
(g) stanicu domaćina prema zahtjevu 11.
13. Farmaceutski pripravak prema zahtjevu 12, naznačen time da sadrži farmaceutski prihvatljivu pomoćnu tvar ili farmaceutski prihvatljiv nosač i dimerni fuzijski protein prema zahtjevu 8.
14. Polipeptid prema bilo kojem od zahtjeva 1-3, dimerni polipeptid prema zahtjevu 4, fuzijski protein prema zahtjevu 5 ili zahtjevu 6, dimerni fuzijski protein prema zahtjevu 7 ili zahtjevu 8, nukleinska kiselina prema zahtjevu 9, vektor prema zahtjevu 10, ili stanica prema zahtjevu 11, naznačeni time da su za uporabu za:
(i) profilaktičko ili terapijsko liječenje za sprečavanje ili suzbijanje imunološkog odgovora kod sisavca;
(ii) liječenje bolesti ili poremećaja imunološkog sustava; ili
(iii) sprječavanje odbacivanja transplantiranog tkiva ili organa kod sisavca.
15. Dimerni fuzijski protein za uporabu prema zahtjevu 14, naznačen time da dimerni fuzijski protein je dimerni fuzijski protein prema zahtjevu 8.
16. Uporaba polipeptida prema bilo kojem od zahtjeva 1-3, dimernog polipeptida prema zahtjevu 4, fuzijskog proteina prema zahtjevu 5 ili zahtjevu 6, dimernog fuzijskog proteina prema zahtjevu 7 ili zahtjevu 8, nukleinske kiseline prema zahtjevu 9, vektora prema zahtjevu 10, ili stanice prema zahtjevu 11 naznačena time da se koristi za proizvodnju lijeka za:
(i) profilaktičko ili terapijsko liječenje za sprečavanje ili suzbijanje imunološkog odgovora kod sisavca;
(ii) liječenje bolesti ili poremećaja imunološkog sustava; ili
(iii) sprječavanje odbacivanja transplantiranog tkiva ili organa kod sisavca.
17. Uporaba dimernog fuzijskog proteina prema zahtjevu 16, naznačena time da dimerni fuzijski protein je dimerni fuzijski protein prema zahtjevu 8.
18. Postupak za dobivanje polipeptida naznačen time da sadrži kultiviranje stanice domaćina prema zahtjevu 11 u kultivacijskom mediju i izolacije polipeptida koji je stanica eksprimirala.
