Abstract

1328205 Oxazolo-triazines LILLY INDUSTRIES Ltd 13 Oct 1970 [5 Nov 1969] 54286/69 Heading C2C Novel oxazolo-triazines of the Formula I in which R<SP>1</SP> and R<SP>2</SP> each are H, C 1-6 alkyl, phenyl or substituted phenyl, and when R<SP>2</SP> is H R<SP>1</SP> may additionally be CF 3 , or COX, when X is NH 2 or C 1-6 alkoxy, and R is aryl or substituted aryl are prepared by reacting oxazoles of the Formula II above in which Y is H, acyl or -CONHR with isocyanates of the formula RNCO. Alternatively compounds of Formula I above in which R 1 is CONH 2 may be made by reacting the appropriate compound in which R 1 is C 1-6 alkoxycarbonyl with ammonia. 2 - Acetamido - 4 - methyloxazole, 2 - acetamido - 4 - trifluoromethyloxazole, 2 - acetamidooxazole - 4 - carboxamide, and 2 - acetamido - 4- ethoxycarbonyloxazole are obtained by reacting the appropriate 2-amino-oxazoles with acetic anhydride. 2 - Amino - 4 - methyloxazole is made by reacting cyanamide with hydroxyacetone. 2- Amino - 4 - trifluoromethyloxazole and 2- amino - 4 - ethoxycarbonyloxazole are obtained by reacting urea with bromotrifluoracetone and ethyl bromopyruvate respectively. Pharmaceutical compounds, having antiinflammatory and diuretic activity and suitable for oral, parenteral or rectal administration, contain the above novel oxazolo-triazines and pharmaceutically acceptable carriers or diluents.
