Technical Field
The invention belongs to the technical field of disinfectants, and particularly relates to a novel coronavirus (SARS-CoV-2) disinfectant prepared from natural plant extract essential oil and application thereof.
Background
The novel coronavirus is a single-stranded positive-strand RNA virus, has strong infectivity, and can be transmitted from person to person through respiratory droplets, direct contact, aerosol and other ways. Meanwhile, the novel coronavirus can survive in an in vitro environment for up to one month, and virus pollution in a public environment is one of important factors causing the new crown to be pandemic. Therefore, the rapid and effective inactivation of the virus in the transmission environment and the blocking of the transmission of the virus from person to person are one of the important working points for controlling the new crown epidemic situation.
The novel coronavirus is sensitive to heat and ultraviolet light, and in addition conventional disinfectants such as alcohol, chlorine-containing disinfectants, peroxides can inactivate the virus. However, the traditional chemical disinfectant has certain toxicity and strong irritation, is easy to damage skin tissues, respiratory systems and the like after long-term use, and can damage the environment in the production and use processes. There are also safety hazards associated with the use of chemical disinfectants in large quantities, and there is a risk of explosion caused by the large quantities of disinfectant, such as alcohol, sprayed in public areas. Disinfectants containing chlorine or peroxides also present residual problems, with potential risks to both humans and the environment. Therefore, it is necessary to develop new green, environment-friendly and effective coronavirus disinfectants for blocking virus transmission and protecting human health.
The natural plant extract essential oil is a plant metabolic product extracted from plants by a specific method, is rich in active substances such as ketone, terpene, aldehyde, alcohol and the like, and is widely applied to products such as medicines, antibacterial agents, antiviral agents and the like due to the activity of enhancing immunity, bacteriostasis, antiviral agents and the like. The natural plant extract essential oil is obtained by a general method, such as geranium oil also called geranium oil obtained by steam distillation of fresh leaves of geranium, cedar leaf oil obtained by extracting cedar, red pine or red cedar leaves by steam distillation or solvent extraction, rosemary oil obtained by steam distillation of erect or creeping whole plants of rosemary, orange flower essential oil obtained by steam distillation of petals of bitter orange, sweet orange or lemon tree, and sage oil obtained by steam distillation of dried sage. In addition, the experiment result also proves the inactivation effect of the natural plant extract essential oil disinfectant on the novel coronavirus, and the experiment also proves that the natural plant extract essential oil disinfectant has no cytotoxicity. The natural plant extract essential oil is widely applied in the field of air purifiers, is nontoxic and harmless to human bodies, has aromatic components, is environment-friendly and has no safety risk.
Disclosure of Invention
The invention aims to provide a novel natural plant extract essential oil coronavirus disinfectant which is prepared by taking natural plant extract essential oil with different proportions as raw materials, wherein the raw materials are simple and easy to obtain, nontoxic, harmless, green and environment-friendly, the preparation method is simple to operate, and the obtained novel coronavirus disinfectant is convenient to use, remarkable in effect and free of environmental pollution and potential safety hazards.
The invention discloses a novel natural plant extract essential oil coronavirus disinfectant which is prepared from the following natural plant extract essential oil in parts by weight: 10-40 parts of geranium oil, 7-37 parts of cedar leaf oil, 13-43 parts of neroli essential oil, 3-33 parts of rosemary oil and 1-22 parts of sage oil.
In a preferred embodiment of the invention, the novel natural plant extract essential oil coronavirus disinfectant is prepared by combining the following natural plant extract essential oils in parts by weight: 20-30 parts of geranium oil, 17-27 parts of cedar leaf oil, 23-33 parts of neroli essential oil, 13-23 parts of rosemary oil and 3-12 parts of sage oil.
In a preferred embodiment of the invention, the novel natural plant extract essential oil coronavirus disinfectant is prepared by combining the following natural plant extract essential oils in parts by weight: 25 parts of geranium oil, 22 parts of cedar leaf oil, 28 parts of neroli essential oil, 18 parts of rosemary oil and 7 parts of sage oil.
The invention also aims to provide a preparation method of the novel natural plant extract essential oil coronavirus disinfectant, which is realized by the following steps: selecting corresponding natural plant extract essential oil pelargonium roseum oil, cedar leaf oil, neroli essential oil, rosemary oil and sage oil according to the weight parts in the formula, sequentially adding the natural plant extract essential oil pelargonium roseum oil, the cedar leaf oil, the neroli essential oil, the rosemary oil and the sage oil into a mixing kettle, and stirring and mixing for 0.1-1h at the rotating speed of 200-600r/min under magnetic stirring to fully mix; ultrasonically dispersing the prepared plant extract essential oil mixed solution, and standing for more than 1-2h to obtain plant extract essential oil disinfectant; and detecting, filling and packaging the plant extract essential oil disinfectant finished product to obtain the novel natural plant extract essential oil coronavirus disinfectant for sale.
The invention also aims to provide the application of the novel natural plant extract essential oil coronavirus disinfectant in novel coronavirus disinfection in the environment, wherein the coronavirus is human coronavirus SARS-CoV-2.
Experiments show that the SARS-CoV-2 virus smeared on the surface of the carrier has obvious inhibition effect on the activity of the SARS-CoV-2 virus after being exposed in the atmosphere of the natural plant extract essential oil disinfectant for 15min, 30min, 45min and 60 min. Meanwhile, the same treatment mode has no obvious toxic effect on Vero-E6 cells. This indicates that the natural plant essential oils in these formulations are safe and effective and can be used as disinfectants for the disinfection of the new coronavirus (SARS-CoV-2) in the environment. The natural plant extract essential oils mentioned in the present invention also have novel coronavirus inhibitory activity either alone or in any combination,
the application is that the novel coronavirus disinfectant is placed in an environment needing disinfection, and the effective components volatilized by the disinfectant are utilized to kill coronavirus.
The application is that the novel coronavirus disinfectant is matched with equipment such as a humidifier, an air conditioner, an air purifier and the like to be used for disinfecting indoor environment.
The application is that the novel coronavirus disinfectant is used for preparing daily washing, protecting and disinfecting products, wherein the daily washing, protecting and disinfecting products comprise antiviral spray, hand sanitizer, skin care products and wet tissues.
The invention has the beneficial effects that: the natural plant extract essential oil is used as raw material, and a novel disinfectant capable of inactivating a novel coronavirus (SARS-CoV-2) is prepared according to a proportion. The components in the formula are natural components, the raw materials are easy to obtain, and the novel disinfectant is simple in preparation process. The obtained natural plant extract essential oil disinfectant can effectively inactivate novel coronavirus without toxicity, and can be directly applied to disinfection of novel coronavirus (SARS-CoV-2) in the environment.
Detailed Description
The present invention will now be described more fully hereinafter with reference to the accompanying examples, in which the specific examples are shown and described, simply by way of illustration and not by way of limitation.
Example 1:
according to the proportion and the components in the formula, 25 g of geranium oil, 125 g of cedar leaf oil, 22 g of cedar leaf oil, 28 g of orange blossom essential oil, 18 g of rosemary oil and 90 g of sage oil are sequentially added into a mixing kettle, the mixture is stirred and mixed for 0.5h at the rotating speed of 400r/min under the magnetic stirring, the mixture is fully mixed, the prepared plant extract essential oil mixed solution is subjected to ultrasonic dispersion for 1.5 and then stands to obtain plant extract essential oil disinfectant, and the plant extract essential oil disinfectant prepared in the last step is subjected to detection, filling and packaging to obtain the novel coronavirus disinfectant for natural plant extract essential oil which can be sold.
Example 2:
exposing SARS-CoV-2 virus (strain number BetaCoV/JS02/Human/2019) coated on the surface of the carrier to the natural plant extract essential oil disinfectant and rapeseed oil volatilization atmosphere or normal air environment for 15min, 30min, 45min and 60min, eluting the virus on the surface of the carrier by using virus growth liquid and culturing for 18-20 h, and carrying out cytotoxicity detection by adopting a CCK-8 method. All experiments were repeated in triplicate.
Example 3:
cytotoxicity experiments: the MTT method was used to determine the cytotoxicity of the plant extract disinfectant and negative control rapeseed oil of each formulation against Vero-E6. Cells were seeded in 96-well plates and incubated until the number of cells per well reached about 1X 104And (4) respectively. Cells were then incubated with 2mL of each formulation of disinfectant and negative control canola oil for 24 hours. MTT solution (5 mg. mL)-1) The medium is removed by adding each well separately and incubating for an additional 4 hours at 37 deg.C, and the low insoluble purple formazan crystals in the wells are solubilized with 100 μ L of dimethyl sulfoxide. The plates were then shaken for 30 minutes and the absorbance of the solution at 570nm was measured in each well and the cell viability in each formulation was calculated by comparison of absorbance values against negative cells without any phytodisinfectant or rapeseed oil. All experiments were repeated in triplicate.

The experiment result shows that the natural plant extract essential oil disinfectant with the formula has different inhibition rates on viruses under different time periods of 0 to 60 minutes. The formula disinfectant has 89.65% of inhibition rate on the novel coronavirus after 15 minutes of treatment, and the formula has 98.93% of inhibition rate on the novel coronavirus after 60 minutes of treatment. Negative control rapeseed oil had no inhibitory effect on the virus.

The experimental result shows that the cell survival rate is not influenced under the treatment of the plant extract essential oil disinfectant and the negative control rapeseed oil with the formula, and the natural plant extract essential oil disinfectant has no cytotoxicity.
Therefore, the novel natural plant extract essential oil coronavirus disinfectant prepared by the invention can effectively kill novel coronavirus, and simultaneously, because chemical reagents such as alcohol, oxidant, chloride and the like are not contained in the components, the defects of irritation, corrosivity and the like of the traditional disinfectant do not exist. The novel natural plant extract essential oil coronavirus disinfectant in the formula also has no cytotoxicity and potential safety hazard. Meanwhile, the disinfectant has aromatic smell, can inactivate novel coronavirus and eliminate peculiar smell in the air, and does not influence normal production and living order of a common area.
