Abstract
本发明涉及一种具有细胞水平修复作用的防晒组合物及其应用,所述组合物包括以下重量百分比含量的各组分:山茶提取物:15~25%;绿茶提取物:0.5~1.5%;海藻多糖:0.5~1.5%;光甘草定:0.5~1.5%;积雪草苷:0.5~1.5%;水:65~80%;乳化剂:1~5%。本发明的防晒组合物可从细胞水平对UVB造成的皮肤伤害进行修复,缓解UVB造成的皮肤屏障受损、皮肤老化、红斑等现象,还可通过协同作用,增加防晒剂防晒指数,提高产品防晒效果;通过乳化工艺,将水溶性和油溶性成分融合,便于活性物应用,此外该组合物作用效果温和,天然、安全低刺激。

技术领域
本发明涉及护肤品技术领域,具体地说,涉及一种防晒组合物及其应用,尤其涉及一种具有细胞水平修复作用的防晒组合物及其应用。
背景技术
UVB(290-320nm)和UVA(320-400nm)是导致皮肤晒伤、光老化及皮肤癌的重要因素。而其中的UVB对皮肤的损伤强度是同等剂量UVA的800-1000倍。角质形成细胞位于表皮层,是抵抗紫外线照射的第一道防线,也是UVB波长能量作用的主要靶部位。UVB辐射可引起永生化表皮角质形成细胞(HaCaT)辐射损伤,导致皮肤组织中的活性氧(ROS)水平和自由基的生成及肿瘤坏死因子-α(TNF-α)和白介素-6(IL-6)的增加,进而引起一系列的氧化损伤。UVB还可穿透表皮到达真皮上部,激活基质金属蛋白(MMPs)调节的信号通路,诱导皮肤光老化,甚至引起细胞DNA光加合物嘧啶二具体的形成,若修复不善,将可导致DNA突变及皮肤癌的发生。如果长期户外活动,受到UVB辐射后不能采取有效的紫外线防护措施,很容易引起细胞的损伤,并伴随肌肤的老化。
现有的防晒产品多采用化学防晒剂来抵抗紫外线的伤害,一般防晒指数SPF值越高,防晒效果越好,但同时添加的UVB吸收剂含量越高,对皮肤的副作用也越大。近些年,开发的防晒产品中也会添加一定的植物防晒剂来减少紫外线的伤害,或者通过植物防晒剂来代替化学防晒剂,来减少化学防晒剂的副作用,但是防晒指数偏低。如专利文献CN102302417A记载了将海洋贝类活性肽、海洋胶原蛋白肽、海藻多糖与防晒剂进行结合,在减少防晒剂用量的同时,提高了其防晒能力和产品安全性。专利文献CN201510104419.6提供了一种含有脐形紫菜提取物、盐生杜氏藻提取物、雨生红球藻提取物、海水珍珠粉的防晒化妆品组合物,但是其紫外线吸收、防晒能力并没有得到显著的提升,还仍然需要0.4~35%含量的双-乙基己氧苯酚甲氧苯基三嗪、二乙基氨基羟基苯甲酰苯甲酸己酯等有机紫外线吸收剂,才能达到符合要求的防晒指数。
专利文献CN 110151619A公开了一种含多种活性物的防晒组合物,具体含有生育酚乙酸酯、抗坏血酸四异棕榈酸酯、红球藻抗逆因子、麦角硫因以及植物提取复合物(光果甘草根提取物、茶叶提取物、迷迭香叶提取物、积雪草提取物、母菊花提取物、黄芩根提取物、虎杖根提取物),对皮肤刺激性少,保湿性好,有高的水分增长率,而且皮肤弹性增加率一直持续增长。但该防晒组合物存在以下不足:该组合仅具有缓解紫外线对皮肤的刺激,增加皮肤弹性及水分含量,不能同时具备降低紫外线辐射和修复紫外线对皮肤的损伤作用,不能从根本上减少紫外线对皮肤的伤害。
发明内容
为了克服现有产品的不足,本发明的目的是提供一种具有细胞水平修复作用的防晒组合物及其应用,以山茶提取物、绿茶提取物、海藻多糖、光甘草定、积雪草苷搭配组合,获得一种可以从细胞水平进行紫外线晒伤修复作用的组合物,从源头阻断紫外线的皮肤的伤害,并将其应用于防晒产品中,以提高化学防晒剂的防晒指数,进而获得一种高防晒指数、安全的、有效的防晒产品。

本发明还提供了一种具有细胞水平修复作用的防晒组合物的制备方法,包括以下步骤:
A、将绿茶提取物、海藻多糖、光甘草定、积雪草苷分别与水进行溶解稀释后,混合形成混合物X;
B、将山茶提取物升温加热,得油相;将氢化卵磷脂加水分散后,加热,得水相;
C、将水相加入到油相中,搅拌均匀后,均质,然后降温,加入混合物X再搅拌均匀,冷却,即得所述防晒组合物。
优选地,步骤B中,所述升温加热的温度为80±2℃。
优选地,步骤C中,所述均质的条件为:搅拌频率25±5Hz,均质频率40±5Hz,均质3分钟。
优选地,步骤C中,所述降温至温度为75±2℃。
本发明还提供了一种具有细胞水平修复作用的防晒组合物在护肤品中的应用。
优选地,所述护肤品的制备方法如下:
称取0.5%的乙基己基三嗪酮、0.5%奥克立林、0.5%甲氧基肉桂酸乙基己基,升温加热至80±2℃,即得油相;
将2%的氢化卵磷脂在96%水中充分分散后,水加热至80±2℃,即得水相;
把水相添加到油相中,搅拌均匀后,搅拌频率25±5Hz,40±5Hz均质3分钟;然后降温至75±2℃,加入0.5~5%的防晒组合物,搅拌均匀后,冷却即得所述护肤品。
山茶提取物:《中国药典》等记载,茶油可用于医治外伤、烫伤,可消炎生肌,抗紫外线作用。上海林清轩生物科技有限公司以山茶花、叶、籽为原材料,研究开发的山茶提取物(公司产品清轩萃),富含黄酮、多酚、VE、油酸、角鲨烯、植物甾醇等多种生物活性成分及钙、钾、镁、锌、锰等营养元素,既具有良好的修复皮肤屏障功效,又可清除自由基,具有抗氧化作用。此外,现代研究表明,山茶提取物中的低极性活性成分多酚和黄酮类化合物具有抵抗紫外线的作用。
积雪草苷:可有效清除紫外线辐射产生的自由基,减轻脂质过氧化反应产生对机体的损伤,还能使辐射后的细胞进行正常的DNA分裂,促进细胞周期平稳过度,提高细胞的生存活力和细胞活性,避免皮肤癌的产生。紫外线照射后,使用积雪草苷,还可增加皮肤的羟脯氨酸含量,改善皮肤的含水量和皮肤弹性,对短期高强度的UVB照射所造成的光老化的损害的有修复和保护作用。
海藻多糖:细胞自噬是真核生物中进化保守的对细胞内物质进行周转的重要过程。该过程中一些损坏的蛋白或细胞器被双层膜结构的自噬小泡包裹后,送入溶酶体中进行降解并得以循环利用。UVB辐射以非MTOR依赖性方式抑制角质形成细胞自噬,海藻糖是角质形成细胞的非MTOR依赖性自噬诱导剂。海藻糖对UVB损伤的角质形成细胞具有降低细胞死亡和改善细胞代谢的效应,展现了良好的细胞保护效应。
光甘草定:即甘草黄酮,具有抗氧化作用,能够清除自由基,同时能通过抑制caspase-12、CHOP的活化,进而阻断caspase-3的激活,抑制细胞的凋亡,从而减缓UVB辐射引起的皮肤光老化现象。
绿茶提取物:活性成分表没食子儿茶素没食子酸酯具有光保护作用,UVB辐射后,可通过下调修复相关调控分子和基因表达,降低永生角质形成细胞和成纤维细胞光产物的产生。还可保护树突状细胞免疫功能作用,这主要与保护其表面共刺激表达有关。可以通过不可逆损伤细胞的凋亡,减少UVB诱导的人皮肤成纤维细胞的突变,可通过保护作用减少UVB辐射诱导皮肤局部免疫抑制作用,可减少组织的慢性光损伤作用。在化妆品中加入茶多酚对皮肤无不良反应,而且具有润滑效果和祛斑效果。
与现有技术相比,本发明具有如下的有益效果:
本发明的防晒组合物可从细胞水平对UVB对造成的皮肤伤害进行修复,缓解UVB造成的皮肤屏障受损、皮肤老化、红斑等现象,还可通过协同作用,增加防晒剂防晒指数,提高产品防晒效果;通过乳化工艺,将水溶性和油溶性成分融合,便于活性物应用,此外该组合物作用效果温和,天然、安全低刺激。
具体实施方式
下面结合具体实施例对本发明进行详细说明。以下实施例将有助于本领域的技术人员进一步理解本发明,但不以任何形式限制本发明。应当指出的是,对本领域的普通技术人员来说,在不脱离本发明构思的前提下,还可以做出若干变形和改进。这些都属于本发明的保护范围。
实施例1-5和对比1-7
实施例1-5和对比例1-7提供了一种具有细胞水平修复作用的防晒组合物及其制备方法,所述防晒组合物的各组分及含量如表1所示。所述防晒组合物的制备方法如下:
将新鲜绿茶叶高温杀青后,烘干粉碎过筛,按照绿茶粉:水:乙醇=1:10:10的比例在40℃下,超声萃取30min,静置30min后,取上清液用0.3μm微孔滤膜过滤取得澄清液,浓缩即得绿茶提取物。
以褐藻提取物为原材料,包括齿缘墨角藻、钝马尾藻,烘干粉碎过筛,按照海藻粉:水:乙醇=1:10:10的比例在40℃下,超声萃取30min,静置30min后,取上清液用0.3μm微孔滤膜过滤取得澄清液,真空浓缩滤液至原体积的1/4,加入3倍体积的乙醇,放置24h,离心过滤得沉淀物,得海藻多糖。
将绿茶提取物、海藻多糖、光甘草定、积雪草苷和水分别按照1:5的比例混合后,再将溶解稀释后的各提取物混合,即得混合物X。
称取一定量的山茶提取物(如上海林清轩生物科技有限公司生产的清轩萃,其山茶籽油(wt%):山茶花提取物(wt%):山茶叶提取(wt%)物的比例为95:2.5:2.5),升温加热至80±2℃,即得油相。将氢化卵磷脂在水中充分分散后,加热至80±2℃,即得水相。把水相添加到油相中,搅拌均匀后,搅拌频率25±5Hz,均质频率40±5Hz,均质3分钟。降温至75±2℃,加入上述提取物混合液,搅拌均匀后,冷却即得防晒组合物。

实施例4
本实施例与实施例1的防晒组合物及其制备方法基本相同,不同之处仅在于:本实施例中采用的清轩萃为山茶籽油(wt%):山茶花提取物(wt%):山茶叶提取物(wt%)=90:7:3。
实施例5
本实施例与实施例1的防晒组合物及其制备方法基本相同,不同之处仅在于:本实施例中采用的清轩萃为山茶籽油(wt%):山茶花提取物(wt%):山茶叶提取物(wt%)=98:1:1。
对比例6
本对比例与实施例3的防晒组合物及其制备方法基本相同,不同之处仅在于:本实施例中采用山茶籽油(wt%):山茶花提取物(wt%)=95:5的组合物代替清轩萃。
对比例7
本对比例与实施例3的防晒组合物及其制备方法基本相同,不同之处仅在于:本实施例中采用山茶籽油(wt%):山茶叶提取物(wt%)=95:5的组合物代替清轩萃。
功效验证试验:
1、对DPPH自由基清除能力测定
将2mL待测样品中加入0.2mmol/L DPPH-乙醇(乙醇体积分数:60%)溶液2.0mL,混匀,静置30min,于517nm波长处测定吸光度A1。以待测样品与体积分数60%乙醇溶液的混合液为对照,测定吸光度A2;以DPPH溶液与体积分数60%乙醇溶液的混合液为空白,测定吸光度A0。计算公式如下:
DPPH自由基清除率/%=(1-A1-A2/A0)*100
结果如表2所示。

2、对细胞影响
采用DMEM培养基培养HaCaT细胞,SUV-100日光紫外线模拟器及UVB辐射度监示器进行实验。按照下表(见表3)设计开展实验,收集各组细胞进行相关指标检测。

2.1对细胞增殖的影响
采用MTT法检测细胞存活率。将不同组别的细胞接种于96孔板中,每组设置5个复孔,孔加入100μL DMEM完全培养基与10μL 5mg/mL MTT混合液,置于37℃、5%CO2培养箱中孵育4h。4h后小心吸去上清液,加入150μL DMSO,避光静置10min后在酶标仪上测定其在570nm波长处OD值,OD值越大表明存活率越高。
细胞相对存活率/%=OD样品组/OD对照组*100
式中:OD样品组、OD对照组分别为样品组、对照组在570nm波长处OD值。
2.2细胞内脂质过氧化物(MDA)含量的测定
将分组的细胞培养24h后,按照试剂盒说明书进行操作,测定MDA含量浓度。
2.3细胞内酶活力的测定
将分组的细胞培养24h后,按照试剂盒说明书测定细胞内SOD、CAT活力。
2.4ELISA法检测MMP-1含量、TNF-α及IL-6质量浓度
将分组的细胞培养24h后,按照试剂盒说明书检测MMP-1含量、TNF-α及IL-6质量浓度。
各测试结果如表4所示。

3安全性评估
1、选取40名受试者曲臂内侧作为测试区域。10人一组,分区标记位置如下:在距手腕三指宽处划定一个3cm×3cm区域,标记为样品修复;另一条手臂在样品修复组对应区域划定一个3cm×3cm区域,标记为自然修复。自然修复不涂抹任何样品。
②样品修复使用0.5%SLS溶液200μl单次诱导,刺激2h后去除斑贴器(包含半小时的环境适应时间)进行测试。仪器测试结束后再使用各实施例和对比例制备的防晒组合物样品,在样品区域滴一滴,然后小心涂抹在区域内。志愿者应在每天洁肤后在样品区域涂抹样品,于24h、48h进行回访,回访时先洗去测试样品,然后适应环境30分钟后再进行仪器测试(EI值),数据为每组受试者测试数据的平均值。结果如表5所示。

4对防晒指数的影响
配方A:称取0.5%的乙基己基三嗪酮、0.5%奥克立林、0.5%甲氧基肉桂酸乙基己基,升温加热至80±2℃,即得油相。将2%的氢化卵磷脂在96.5%水中充分分散后,水加热至80±2℃,即得水相。把水相添加到油相中,搅拌均匀后,搅拌频率25±5Hz,均质频率40±5Hz,均质3分钟。冷却即得配方A。
配方B:称取0.5%的乙基己基三嗪酮、0.5%奥克立林、0.5%甲氧基肉桂酸乙基己基,升温加热至80±2℃,即得油相。将2%的氢化卵磷脂在91.5%水中充分分散后,水加热至80±2℃,即得水相。把水相添加到油相中,搅拌均匀后,搅拌频率25±5Hz,均质频率40±5Hz,均质3分钟。降温至75±2℃,加入5%实施例1制备的防晒组合物,搅拌均匀后,冷却即得配方B。
配方C:称取0.5%的乙基己基三嗪酮、0.5%奥克立林、0.5%甲氧基肉桂酸乙基己基,升温加热至80±2℃,即得油相。将2%的氢化卵磷脂在93.5%水中充分分散后,水加热至80±2℃,即得水相。把水相添加到油相中,搅拌均匀后,搅拌频率25±5Hz,均质频率40±5Hz,均质3分钟。降温至75±2℃,加入3%实施例1制备的防晒组合物,搅拌均匀后,冷却即得配方C。
配方D:称取0.5%的乙基己基三嗪酮、0.5%奥克立林、0.5%甲氧基肉桂酸乙基己基,升温加热至80±2℃,即得油相。将2%的氢化卵磷脂在96%水中充分分散后,水加热至80±2℃,即得水相。把水相添加到油相中,搅拌均匀后,搅拌频率25±5Hz,均质频率40±5Hz,均质3分钟。降温至75±2℃,加入上述混0.5%实施例1制备的防晒组合物,搅拌均匀后,冷却即得配方D。
测试方法:使用紫外分光光度计,在短波紫外线范围内(290-320nm)选择7个波长点,分别将加入配方组分的样品溶液与60%的乙醇按照5:1的比例混合后,置于石英比色皿内,以未加配方组分的60%的乙醇为空白,进行吸光值的测定。吸光度<1.0,表明无防晒效果;吸光度=1.0,表示有低级防晒效果;1.0<吸光度<2.0,表示有中级防晒效果;吸光度>2.0,表示有高级防晒效果。参照《化妆品管理及安全性和功效评价》(主编:秦钰慧)。结果如表6所示。

本发明具体应用途径很多,以上所述仅是本发明的优选实施方式。应当指出,以上实施例仅用于说明本发明,而并不用于限制本发明的保护范围。对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以做出若干改进,这些改进也应视为本发明的保护范围。
Claims (10)
2.根据权利要求1所述的具有细胞水平修复作用的防晒组合物,其特征在于,所述山茶提取物包括以下质量比的各组分:山茶籽油:山茶花提取物:山茶叶提取物的比例为90~99:1~7:0.5~3.5。
3.根据权利要求1所述的具有细胞水平修复作用的防晒组合物,其特征在于,所述的乳化剂选自氢化卵磷脂、磷脂酰胆碱、鲸蜡硬脂基葡糖苷中的至少一种。
5.一种根据权利要求1所述的具有细胞水平修复作用的防晒组合物的制备方法,其特征在于,包括以下步骤:
A、将绿茶提取物、海藻多糖、光甘草定、积雪草苷分别与水进行溶解稀释后,混合形成混合物X;
B、将山茶提取物升温加热,得油相;将氢化卵磷脂加水分散后,加热,得水相;
C、将水相加入到油相中,搅拌均匀后,均质,然后降温,加入混合物X再搅拌均匀,冷却,即得所述防晒组合物。
6.根据权利要求5所述的具有细胞水平修复作用的防晒组合物的制备方法,其特征在于,步骤B中,所述升温加热的温度为80±2℃。
7.根据权利要求5所述的具有细胞水平修复作用的防晒组合物的制备方法,其特征在于,步骤C中,所述均质的条件为:搅拌频率25±5Hz,均质频率40±5Hz,均质3分钟。
8.根据权利要求5所述的具有细胞水平修复作用的防晒组合物的制备方法,其特征在于,步骤C中,所述降温至温度为75±2℃。
9.一种根据权利要求1-4所述的具有细胞水平修复作用的防晒组合物在护肤品中的应用。
10.根据权利要求9所述的应用,其特征在于,所述护肤品的制备方法如下:
称取0.5%的乙基己基三嗪酮、0.5%奥克立林、0.5%甲氧基肉桂酸乙基己基,升温加热至80±2℃,即得油相;
将2%的氢化卵磷脂在96%水中充分分散后,水加热至80±2℃,即得水相;
把水相添加到油相中,搅拌均匀后,搅拌频率25±5Hz,40±5Hz均质3分钟;然后降温至75±2℃,加入0.5~5%的防晒组合物,搅拌均匀后,冷却即得所述护肤品。
