after the experiment is started, the hairs on the back of the trunk of the animal are removed, the hair-removed area accounts for about 10% of the surface area of the body of a rat, then the hemorrhoid comforting ointment prepared by the invention is uniformly smeared on the hair-removed area, the dose is 5000mg/kg of the body weight, a layer of thin film is used for covering, a non-irritant adhesive plaster is used for fixing, and then observation is carried out for 24 hours; after the observation is finished for 24 hours, cleaning the tested rat by using clear water, and continuously observing for 14 days; the tested rat has no poisoning and death phenomenon within 24h, and has no tremor, convulsion, salivation, diarrhea, lethargy, coma and other phenomena, the appearance, fur and skin color of the animal have no abnormal change, no abnormal breathing exists, and no abnormal secretion exists in the nose, eyes and oral cavity. And the behavioral activities in the 14d later are not abnormal and have no great difference with the blank control group; the once-limiting method is adopted to detect that the test object with low toxicity does not cause animal death, which indicates that the hemorrhoid comforting ointment prepared by the invention can be used in large dose.
II, anti-inflammatory experiment:
50 SD rats with the weight of 180-. Marking the ankle joints of the right hind limbs of the rats by using a marker pen before the experiment, and administering corresponding medicines of each group at the marked positions for 3-4 days continuously; the foot swelling degree (the swelling degree of foot is the volume of foot after inflammation-volume of foot before inflammation) is calculated by a daily foot volume measuring device in the experiment. The corresponding drugs are respectively administered at the marked positions of rats at the present day, and after lh administration of the drugs, 0.1 lml/rat of 1% carrageenan is injected into the right plantar skin marked by each rat intradermally to cause inflammation; the volume of the right hind paw of the mouse is measured 30min, 60min and 120min after inflammation, and the swelling degree of the hind paw after inflammation is recorded for 30min, 60min and 120 min. Shearing inflammatory feet below the ankle joint, weighing, placing in 2ml of physiological saline, fully shearing, homogenizing, centrifuging (3000rpm for 5min), taking supernatant, detecting SOD and MDA contents with a kit, and detecting IL-1 content of the supernatant by an ELISA method; the measurement results are shown in tables 1 and 2:

By contrast, after equal amounts of hemorrhoid comforting ointment, marigold and Jiuhua ointment are used, the swelling degree of feet is relieved to a certain extent, which shows that the medicines of the examples and the comparative examples can inhibit acute inflammation of rats caused by carrageenan, the relieving degree of the examples at 120min is better than that of the comparative examples, and the inhibiting effect of the examples is better and the inflammation is relieved more quickly.

The results in table 2 show that the SOD content of the hemorrhoid comforting ointment prepared by the invention is obviously higher than that of the ointment in the market, and the MDA and IL-1 are obviously lower than that of long-term ointment, which shows that the hemorrhoid comforting ointment of the invention has more excellent inhibition effect on inflammatory cells.
Thirdly, pharmacodynamic experiment:
the clinical observation of the hemorrhoid comforting ointment prepared by the invention is compared with the hemorrhoid ointment on the market, and the comparison is as follows:
selecting 180 cases with mixed hemorrhoids, wherein the market of the ointment is better in 4-8 days, and the medicine is exchanged for 1 time each day; the ointment for treating hemorrhoid is improved after 2-5 days, and the ointment is changed for 1 time every day, and pain is relieved on the day. 230 anal fissures, the market ointment improves in 5-15 days, the medicine is changed for 1 time every day, the hemorrhoid comforting ointment is changed for 4-10 days, and the medicine is changed for 1 time every day. The perianal abscess can be treated by exchanging the medicine for 1 time every day for 4-20 days. The hemorrhoid comforting ointment of Shengshichuang is 2-12, the medicine is changed 1 time every day, and the whole case is treated according to the disease condition by matching with the operation.

Improvement: the pain is relieved, the pain is not felt when walking, and the hemorrhoids are reduced;
and (3) curing: the hemorrhoids disappear, the pain stops, the stool does not bleed, and the swelling disappears.
The result shows that the hemorrhoid comforting ointment prepared by the invention has quick curative effect and short cure time, and has more excellent effect compared with the long-time ointment in the prior art.
Fourthly, clinical trial:
certain Liu, woman, age 33; admission at 11/9/2016 and discharge at 3/17/2016; and (3) diagnosis: anal fissure, mixed hemorrhoid; the total treatment time is 8 days, the pain is improved after the patient is admitted for 2 days, and the pain is relieved in the same day; the patient is discharged after 8 days; no relapse occurs after 2 years of study.
White certain, female, age 67; diagnosis, anal fistula, mixed hemorrhoids; the patient is admitted at 10 days of 12 months and 10 months in 2016, and is discharged at 10 days of 12 months and 20 months in 2016; the total treatment time is 10 days by using the hemorrhoid comforting ointment; after the patient is admitted, the flaccid tube mixed hemorrhoid resection is carried out, then the medicine is exchanged every day, the pain is relieved on the day of postoperative medicine exchange, the patient is completely cured and discharged after 10 days, and no relapse occurs after 2 years of inert visit.
Zhangyi, woman, age 55; diagnosis, mixed hemorrhoid, perianal abscess; admission at 2016, 11, 28, 12, and discharge at 2016, 12, 7, 9; the hemorrhoids-relieving ointment is used for treating the hemorrhoids and is discharged after 8 days of hospital recovery, and no relapse occurs after 2 years of study.
Yan somebody, woman, 26 years old; and (3) diagnosis: anal fissure is admitted at 2016, 12 and 9 days, and discharged at 2017, 1 and 15 days, and the total number of hospitalization days is 15 days. The ointment in the ointment market is less than Jiuhua ointment, and the Mayilong hemorrhoids ointment has no relapse after more than 2 years of inert visit.
Comparative group 2: von of someone, women, age 42; diagnosing mixed hemorrhoid and perianal abscess; the method records that the patient is admitted in 2016, discharged in 19 days in 1 month and 2017, and discharged in 20 days in total. The treatment medicine is Jiuhua plaster, Mayilong hemorrhoid plaster, and has no recurrence after 2 years of study.
Clinical experiments show that the hemorrhoid comforting ointment generally needs 8-10 days for curing, and the market medicines < Jiuhua ointment and Mayilong hemorrhoid ointment > for curing generally need 15-20 days, so that the hemorrhoid comforting ointment prepared by the invention is also verified to have quick effect, no recurrence is seen in 2 years, and the curative effect is good.

Claims (4)
1. The hemorrhoid comforting ointment is characterized by comprising the following components in parts by weight: 61-63 parts of coptis chinensis, 15-16 parts of borax, 30-32 parts of indigo naturalis, 30-32 parts of golden cypress, 30-32 parts of gallnut, 30-32 parts of angelica dahurica and 2-3 parts of borneol.
2. The hemorrhoid comforting cream according to claim 1, which is prepared from the following components in parts by weight: 63 parts of coptis chinensis, 16 parts of borax, 32 parts of indigo naturalis, 32 parts of golden cypress, 32 parts of gallnut, 32 parts of angelica dahurica and 3 parts of borneol.
3. The method for preparing hemorrhoid relaxing cream according to claim 1, comprising the steps of:
step one, weighing the following raw materials in parts by weight: 61-63 parts of coptis chinensis, 15-16 parts of borax, 30-32 parts of indigo naturalis, 30-32 parts of golden cypress, 30-32 parts of gallnut, 30-32 parts of angelica dahurica and 2-3 parts of borneol for later use;
step two, drying the coptis chinensis, the golden cypress, the gallnut and the angelica dahurica together, crushing and sieving by a sieve of 60-200 meshes to obtain fine powder;
step three, uniformly mixing the heated vaseline and the fine powder obtained in the step two, cooling to paste, adding borax, borneol and indigo naturalis, and uniformly mixing to obtain a plaster;
wherein the mass ratio of the total weight of the raw materials to the vaseline is 5: 1-1.5;
step four, adding sesame oil into the plaster obtained in the step three to obtain hemorrhoid comforting plaster;
wherein the mass ratio of the total weight of the raw materials and the vaseline to the sesame oil is 10: 0.15-0.25;
and fifthly, instantly sterilizing at high temperature and bottling.
4. The method for preparing hemorrhoid comforting ointment according to claim 3, wherein the high-temperature instant sterilization conditions in the fifth step are as follows: keeping the temperature at 110-120 ℃ for 2-3 minutes until the finished product reaches a commercial sterile state.
