Precisely absorbing rutin reference substances of 0, 1.0, 2.0, 3.0, 4.0, 5.0 and 6.0mL, respectively placing in a 25mL measuring flask, respectively adding 30% ethanol to 6mL, adding 5% sodium nitrite solution of 1mL, uniformly mixing, placing for 6min, adding 10% aluminum nitrate solution of 1mL, uniformly mixing, placing for 6min, adding sodium hydroxide solution of 10mL, adding 30% ethanol to the scale, uniformly mixing, placing for 15min, and measuring the absorbance at the wavelength of 500nm by using an ultraviolet spectrophotometer. A standard curve was plotted with the absorbance A as the ordinate and the mass concentration C (mg/mL) as the abscissa.
2.2.3 determination of Total Flavonoids content
Precisely absorbing 1mL of sample liquid, placing the sample liquid in a 25mL volumetric flask, measuring the absorbance according to the method from 'adding water to 6mL and adding 5% sodium nitrite to 1 mL' according to a 3.2.2 method, substituting the absorbance into a standard curve to obtain the concentration of the sample solution, and calculating to obtain the test solution.
2.3 preparation of blank plaster of HESHU weight-reducing and lipid-lowering navel plaster
2.3.1 orthogonal design experiment
According to the preliminary experiment research, determining mixed fatty glyceride as a matrix, lanolin as a tackifier, absolute ethyl alcohol as a dispersing agent, selecting the dosage of the mixed fatty glyceride, the lanolin and the absolute ethyl alcohol as an investigation factor, and adopting L9(34) Orthogonal tables were tested and the protocol is shown in table 3.

2.3.2 preparation method and Scoring Standard
The preparation method comprises the following steps: weighing a certain amount of azone, putting the azone into a certain amount of absolute ethyl alcohol, weighing a certain amount of mixed fatty glyceride and lanolin, putting the mixed fatty glyceride and lanolin into a beaker, melting the mixed fatty glyceride and lanolin in a water bath kettle at 60 ℃, weighing a certain amount of absolute ethyl alcohol, adding the absolute ethyl alcohol into the mixed fatty glyceride and lanolin, fully and uniformly mixing the mixture after the mixed fatty glyceride and lanolin are completely melted, pouring the mixture on a glass flat plate while the mixture is hot, and observing the state of the mixture.
Grading standard: the paste was rated according to the state of dispersion, fluidity, viscosity and appearance of the paste on a glass plate, and was rated as 1. The dispersion state is given 0-0.3 minutes from weak to strong, the fluidity is given 0-0.2 minutes from strong to weak, the viscosity is given 0-0.3 minutes from weak to strong, and the appearance is given 0-0.2 minutes according to the integrity difference. The combined results were scored according to these 4 criteria.
3. Results and analysis

Linear analysis was performed on the data obtained in table 4 and fig. 2, and the regression equation between the rutin concentration (mg/mL) and the absorbance (a) was obtained as follows: 10.5393x +0.0147, R2=0.9999。
The concentration of the rutin control substance is in a good linear relation within the range of 0.008-0.048 mg/mL as can be seen from a regression equation and a standard curve.
3.2 preparation and evaluation of HESHU navel patch for reducing weight and blood lipid
3.2.1 results of orthogonal experiments
The results of the orthogonality test are shown in Table 5.

As can be seen from the visual analysis, the sequence influencing the formulation forming factors is C>B>A; the variance result shows that C has influence on the forming of the navel patch; the optimal prescription of blank matrix is determined by comprehensive consideration as A2B2C1Preferably, the mixed fatty glyceride is 2g, and lanolin is 2gThe amount of absolute ethyl alcohol is 1 mL.
3.2.2 preparation of weight-losing and lipid-lowering navel patch
Preparing a plaster according to the optimal screened blank matrix prescription and the drug loading rate of 10 percent: dissolving a certain amount of refined total flavone active extract and azone in a certain amount of absolute ethyl alcohol according to a certain proportion, putting a certain amount of mixed fatty glyceride and lanolin in a beaker, melting the mixed fatty glyceride and lanolin in a water bath kettle at 60 ℃, measuring a certain amount of absolute ethyl alcohol after the mixed fatty glyceride and lanolin are completely melted, pouring the absolute ethyl alcohol into the mixed fatty glyceride and lanolin, fully mixing the mixed fatty glyceride and lanolin uniformly, and pouring the mixture into a blank paste while the mixture is hot to obtain the weight-reducing and lipid-lowering navel paste for the lotus art. The best preparation prescription obtained in this example is to prepare 10 pieces of navel plaster (each plaster contains about 0.09g of medicine).
Use introduction: 1. wiping the navel with warm water, removing dirt, and finally wiping the surface of the navel; 2. uncovering the paster, and aligning the ointment to the navel to make the medicine fully contact with the skin; 3. rub the hands with the heat, apply the palm to the navel, and gently soften for three minutes.
3.2.3 evaluation of Lotus technique fat-reducing navel plaster
And (3) optimal process verification: the optimal preparation process obtained by the test is repeated for 3 times, and the prepared lotus weight-losing and lipid-lowering navel patch is comprehensively evaluated, and the results are all in line with the expected effect.
Skin irritation test: selecting one healthy rabbit, unhairing two sides of the spinal column of the back before administration, and checking the integrity of the skin after unhairing, wherein the skin is ensured to be intact and not injured. The prepared navel patch is attached to the skin of a rabbit and is taken down for 2, 4, 6, 8, 10, 12 and hours respectively to observe the skin condition of the rabbit, the skin allergy and red swelling conditions are not found, and the patch is proved to not cause irritation to the skin and has certain use safety.
4. Results
4.1 determination of optimum Process conditions
The matrix of the lotus leaf weight-losing and lipid-lowering navel patch is optimized, the formability of the whole navel patch is inspected, and test results show that when the dosage of the mixed fatty glyceride is 2g, the dosage of the lanolin is 2g, and the dosage of the absolute ethyl alcohol is 1mL, the integrity of the patch is good, and the patch can be used for preparing the navel patch. Moreover, the stimulation condition of the navel patch to the skin is investigated, the skin is not stimulated, and the prepared navel patch is proved to have certain safety and certain market development and use prospects.

Claims (10)
1. A preparation method of a lotus leaf, rhizoma atractylodis and tripe navel paste for losing weight and reducing fat is characterized in that the preparation method of the lotus leaf, rhizoma atractylodis and tripe navel paste for losing weight and reducing fat comprises the following steps:
step one, mixing raw medicinal materials according to a ratio, processing the raw medicinal materials, then putting the processed raw medicinal materials into a grinder for grinding, and sieving the obtained powder to obtain medicinal material powder;
step two, taking the prepared medicinal material powder to prepare a crude extract by a reflux extraction method;
purifying the crude extract by using macroporous adsorption resin to prepare an active extract;
step four, dissolving the active extract and the azone obtained in the step three in a certain amount of absolute ethanol according to a certain proportion to prepare a medicine-containing mixed ethanol solution;
dissolving a certain amount of mixed fatty glyceride and lanolin at 60 ℃, adding the medicine-containing ethanol solution according to a proportion after the mixed fatty glyceride and lanolin are completely dissolved, and fully and uniformly mixing;
and step six, dripping the blank paste into the navel paste according to the volume of 1ml of each paste when the paste is hot, condensing, forming and packaging to obtain the lotus leaf weight-losing and lipid-lowering navel paste.
2. The preparation method of the hilum plaster with holy surgery weight-losing and lipid-lowering functions as claimed in claim 1, wherein in the first step, the raw materials are mixed according to the following ratio: 9g of lotus leaf, 9g of fried bighead atractylodes rhizome, 6g of scorched hawthorn fruit, 6g of coix seed, 3g of dried orange peel and 3g of cassia seed; the resulting powder was sieved through a 80 mesh sieve.
3. The method for preparing the hilum plaster with holy surgery weight-losing and lipid-lowering functions as claimed in claim 1, wherein in the second step, the preparation of the sample solution by the reflux extraction method comprises the following steps:
firstly, fully and uniformly mixing medicinal fine powder in a mortar according to a proportion, putting 70.0g of the uniformly mixed medicinal fine powder into a round-bottomed flask, adding 20 times of 80% ethanol, carrying out reflux extraction on the mixture for 2 times at 90 ℃ in a water bath kettle for two hours in total, combining the two crude extract solutions, centrifuging to obtain a filtrate, fixing the volume, and shaking uniformly to obtain an extract;
and secondly, carrying out reduced pressure concentration on the extracting solution in a rotary evaporator to obtain a crude extract of the Heshu weight-losing and lipid-lowering prescription.
4. The method for preparing the hilum plaster with holy surgery weight-losing and lipid-lowering functions as claimed in claim 1, wherein in the third step, the method for purifying the sample liquid comprises the following steps: loading a sample with the mass concentration of 0.75mg/mL and the pH value of 3.5, wherein the loading amount of the resin is 4.4mL/g, the volume flow of the loaded sample is 1.5BV/h, sequentially eluting with 2.0BV of water and 3.0BV of 60% ethanol, concentrating the collected eluent under reduced pressure in a rotary evaporator, transferring the concentrated eluent to an evaporation dish, evaporating the eluent on a water bath kettle to dryness, finally transferring the eluent to a reduced-pressure dryer, and drying the eluent for 8 to 12 hours.
5. The method for preparing the HESHU JIANFEI JIANGZHI MAIQI Patch as claimed in claim 1, wherein,
1g of total flavone extract in the fourth step and 2ml of absolute ethyl alcohol;
in the fifth step, the dosages of the mixed fatty glyceride, the lanolin and the absolute ethyl alcohol are as follows: the dosage of the mixed fatty glyceride is 2g, the dosage of the lanolin is 2g, and the dosage of the absolute ethyl alcohol is 1 mL; putting the mixed fatty glyceride and the lanolin into a beaker, and melting the mixed fatty glyceride and the lanolin in a water bath kettle at the temperature of 60 ℃;
dissolving the mixed fatty glyceride and the lanolin at 60 ℃, adding the medicine-containing ethanol solution according to the proportion after the mixed fatty glyceride and the lanolin are completely dissolved, and fully and uniformly mixing;
and sixthly, dripping the blank paste into the patch while the patch is hot according to the volume of 1ml per patch, condensing, forming and packaging to obtain the lotus leaf weight-losing and lipid-lowering navel patch.
6. A lotus leaf, rhizoma atractylodis and bellied navel paste with weight losing and lipid lowering functions, which is prepared by the preparation method of the lotus leaf, rhizoma atractylodis and bellied navel paste with weight losing and lipid lowering functions of any one of claims 1 to 5.
7. The hilum plaster for holy surgery to reduce weight and fat of claim 6, wherein the drug-loading rate of the hilum plaster for holy surgery to reduce weight and fat is 10%.
8. The use of the navel patch for losing weight and reducing fat of lotus leaf as claimed in claim 6 in weight-reducing medicine.
9. The use of the navel patch for weight losing and lipid lowering according to claim 6 in a lipid lowering medicine.
10. The use of the navel patch for losing weight and reducing fat of lotus leaf as claimed in claim 6 in a medicine for reducing blood pressure.
