Abstract
The invention discloses a skin care composition for removing spots and resisting aging and a preparation method thereof, wherein the main components comprise butter fruit oil, gentian extract, sage extract, lysimachia christinae hance extract, flaxseed extract and acetyl tetrapeptide-11, the active components contained in the composition can quickly penetrate into skin epidermis, stimulate metabolism of skin cells, promote synthesis of collagen and elastin, and particularly the butter fruit oil and flaxseed extract have obvious antioxidation effect, can neutralize free radicals, reduce damage of skin cells and prevent skin aging. The skin care composition is rich in natural plant essence, has reasonable formula and no toxic or side effect, and can obviously lighten color spots and fine lines caused by ultraviolet irradiation after being used, and the skin is white, ruddy, compact and elastic.

The invention relates to the technical field of skin care cosmetic processing, in particular to a freckle-removing and anti-aging skin care composition and a preparation method thereof.
Background
Human skin is an organ that is prone to exhibit aging. With age, the aging characteristics of the skin also follow: increased wrinkles, loose, yellowish skin, reduced gloss, smoothness, coarser texture, increased stain, etc. It is currently widely accepted that the intrinsic cause of skin stain aging is mainly both endogenous and exogenous. Endogenous factors include genetic factors, hormonal shortages, and the like. The skin care products on the market at present often cannot achieve the aim of fundamentally protecting skin due to the problems of chemical agents, strong acid and alkali, chemical preservatives and the like, and can also lead the skin to be dependent or have strong irritation to the skin. Thus, the advent of a healthy, non-irritating skin care composition has become an urgent need. The invention takes the butter fruit tree fruit fat, the gentian root extract, the sage extract, the lysimachia christinae hance extract and the flaxseed extract as main components for removing spots and resisting aging, and achieves the purpose of removing spots and resisting aging of skin through the combination of the fruit fat substances and the active components of traditional Chinese medicines.

Compared with the prior art, the invention has the beneficial effects that:
(1) The main components of the anti-freckle and anti-aging composition are butter fruit, gentian extract, sage extract, lysimachia christinae hance extract, flaxseed extract and acetyl tetrapeptide-11, and the composition contains active components such as alkaloids, glycosides, flavonoids and the like, can quickly penetrate into the epidermis of the skin, stimulate metabolism of skin cells, promote synthesis of collagen and elastin, and particularly has obvious antioxidation effect, can neutralize free radicals, reduce injury of the skin cells and prevent skin aging.
(2) The flaxseed extract is obtained by adopting a cold grinding and enzymolysis technology, flaxseed cells are broken by rasping wall breaking to release active micromolecular substances, solid and liquid phases are separated by centrifugation, the enzymolysis releases flaxseed protein isolate and lignan, and a membrane filtration system is used for molecular screening, so that the obtained micromolecular substances are easy to be absorbed by skin. In the skin care composition test prepared in example 3, DPPH free radical scavenging ability was 75.2%, OH free radical scavenging ability was 63.2%, tyrosinase activity was 43.2%, and HaCaT cell survival rate was 90.1%.

As can be seen from comparison with comparative example 6, the flaxseed is subjected to rasping and centrifugation at low temperature to maintain the activity of small molecules, and the rich unsaturated fatty acid can help reduce the generation of free radicals and prevent oxidative damage of cells. .
Test example 2 tyrosinase inhibition assay
2.1 Test methods
Taking human A375 melanocytes in logarithmic growth phase, regulating cell concentration to 5×10 4/mL with complete culture medium, inoculating into 96-well plate, adding 100 μl of skin care composition per well for continuous culture for 48 hr, and setting control group with blank complete culture medium and positive group with arbutin, and setting 3 multiple wells per well. After incubation to time node, PBS was washed 2 times, 10Triton X100 solution 90. Mu.L was added to each well, the cells were lysed by shaking for 5min, and 1% L of Dopa 10. As can be seen from Table 2, example 3, comparative examples 1 to 6 and the positive control group have an inhibitory effect on tyrosine, wherein tyrosinase activity in example 3 is only 43.2%, indicating that the skin care composition prepared in example 3 has spot-removing ability.
Test example 3H 2O2 protection experiment against induced injury to HaCaT cells
3.1 Test methods
HaCaT cells are cultured by using a complete culture medium, logarithmic growth phase cells are inoculated into a 96-well plate according to 5X 10 4/mL, 100 mu L of each well is inoculated, after the cells are completely attached, the culture is continued for 4 hours by replacing the DMED culture medium containing 250 mu M hydrogen peroxide, the supernatant is discarded, PBS is used for three times, the culture is continued for 24 hours by adding the DMED complete culture medium containing the skin care composition, 4 mu g/mL of vitamin c is used as a positive control group, and distilled water is used as a control group. After incubation to time node, 10. Mu.L of CCK 8 was added to each well, incubated at 37℃for 20min, absorbance values of each well were measured at a wavelength of 450nm, and survival (%) of each well was calculated according to the following formula.

From table 3, it can be seen that the skin care composition has a better protection effect on HaCaT cells with H 2O2 induced oxidative damage, and the HaCaT cell survival rate of example 3 reaches 90.1%, which is significantly different from that of the positive control group, indicating that the skin care composition prepared in example 3 is more beneficial to resisting oxidative damage and aging effects of skin.

The freckle-removing and anti-aging skin care composition is characterized by comprising the following raw materials in parts by weight: 5 to 10 parts of nicotinamide, 2 to 8 parts of polydimethylsiloxane, 2 to 8 parts of polysorbate-60, 25 to 65 parts of traditional Chinese medicine extract, 3 to 10 parts of butter fruit, 1 to 3 parts of hydroxypropyl cyclodextrin, 1 to 3 parts of tetrahydropyrimidine carboxylic acid, 1 to 5 parts of sodium hydroxide, 2 to 10 parts of cyclopeptide-161, 1 to 5 parts of acetyl tetrapeptide-11, 3 to 10 parts of hydroxypropyl tetrahydropyran triol, 3 to 7 parts of retinol, 1 to 5 parts of ascorbic acid, 2 to 9 parts of 3-o-ethyl ascorbic acid, 3 to 10 parts of sodium hyaluronate, 4 to 10 parts of astaxanthin, 5 to 10 parts of essence and 80 to 100 parts of water;
The traditional Chinese medicine extract comprises the following raw materials in parts by weight: 10-20 parts of gentian root extract, 5-15 parts of sage extract, 5-20 parts of lysimachia christinae hance extract and 5-10 parts of flaxseed extract;
the preparation method of the butter tree fruit fat comprises the following steps:
s1, cutting the avocado pulp into pieces, and grinding the pieces into powder by using liquid nitrogen;
s2, adding the avocado powder into an ethanol water solution with the concentration of 60-75 v/v% and the total volume of 3-7 times, and carrying out ultrasonic vibration extraction for 20-40 min;
s3, carrying out reflux extraction in a water bath at the extraction temperature of 40-60 ℃ for 20-30 min, carrying out suction filtration, centrifuging, filtering, evaporating and recovering ethanol to obtain crude extract of the fruit fat;
S4, preparing the crude extract of the fruit fat into a solution of 6-8 mg/ml, enabling the solution to flow through NKA-II macroporous adsorption resin at a flow rate of 2-4 BV/h for adsorption for 50-60 min, collecting effluent liquid, and concentrating to obtain the fruit fat of the Butyrospermum parkii;
