Abstract
The invention discloses a composition for treating alopecia areata and application thereof, and belongs to the technical field of alopecia areata treatment, wherein the composition comprises the following components in parts by weight: 2-50 parts of acidic substance, 5-30 parts of diethyl ether, 5-30 parts of ethanol, 5-20 parts of glycerol and 5-20 parts of benzyl alcohol. The invention solves the hair follicle spasm problem of the alopecia areata part by the mutual synergy of the components with the specific proportion, and is beneficial to promoting the absorption of hair follicle to hair growth nutrient substances and effective components of therapeutic drugs. When the composition is applied to preparing the medicament for treating alopecia areata, if the composition is matched with a traditional Chinese medicine composition capable of promoting hair growth, the problems that the conventional alopecia areata treatment method is long in treatment course, low in cure rate, large in side effect and incapable of timely judging whether the treatment effect is good or not by a patient can be effectively solved.
Description
Technical Field
The invention relates to the technical field of alopecia areata treatment, in particular to a composition for treating alopecia areata and application thereof.
Background
Alopecia areata (alopecia aerata, AA), commonly called "shaving head", is a skin disease characterized by sudden localized alopecia areata, in which the hair at the lesions drops off rapidly, and is circular or irregular, and a few people can drop off completely, called alopecia totalis. Modern medicine suggests that the etiology and pathogenesis of this disease may be related to autoimmune, genetic, mental, microcirculation disorders, other visceral functions, and endocrine disorders.
The main method for treating alopecia areata at home and abroad at present comprises the following steps:
1. the common external medicine is as follows:
(1) Minoxidil; 5% minoxidil cream or solvent, applied 1 or 2 times daily, may be associated with its vasodilation effect.
(2) Anthralin; 0.5 to 1 percent of anthralin ointment or cream is a primary stimulant. It is applied externally 1 to several times daily to limit the appearance of mild dermatitis on the local skin.
(3) A contact sensitizer; diphenyl propenone (DCP) is most commonly used.
(4) Glucocorticoids; potent hormones are administered topically or encapsulated. 0.05% dexamethasone and 50% dimethyl sulfoxide solution have better curative effect than cream and the like when being externally used.
2. The usual internal medications are as follows:
(1) Glucocorticoids; prednisone is taken orally, gradually reduced after several weeks, and then maintained at a small dose for 6 months. The glucocorticoid has good effect, but has great side effect, and is easy to relapse after stopping the medicine, so the glucocorticoid is not used as a conventional therapy. But can be used for treating acute alopecia areata to avoid the development of total alopecia or alopecia universalis.
(2) Cyclosporine; the treatment course is 6-12 months. Some cases are effective, e.g. no effect stops after 4 months.
(3) Thymopentin: intramuscular injection was continued for 3 weeks.
(4) Vasodilators: niacin is orally administered.
3. A local injection method;
Local injection of glucocorticoids is suitable for small-scale alopecia, or for important cosmetic parts (such as eyebrows) of patients with alopecia universalis. The alopecia area can be directly injected, or the periphery of the alopecia area can be injected, so as to control the continuous expansion of the alopecia area. Care is taken to avoid local skin atrophy and sagging that may be caused.
4. Nerve block therapy;
the occipital nerve is occluded, and after the nerve is occluded, the skin temperature in the dominant region increases, which is beneficial to hair regeneration.
At present, although a plurality of methods for treating alopecia areata at home and abroad exist, the treatment methods at least have the following problems:
(1) The treatment course is long.
(2) The treatment effect varies from person to person and the patient cannot judge the treatment effect in time.
(3) The problems of large side effect and easy repeated attack are treated.
(4) Low cure rate.
Disclosure of Invention
The invention aims to provide a composition for treating alopecia areata and application thereof, and aims to solve the technical problems that the conventional alopecia areata treatment method is long in treatment course, low in cure rate, large in side effect and incapable of timely judging whether the treatment effect is good or not by a patient.
In a first aspect, an embodiment of the present application provides a composition for treating alopecia areata, including, in parts by weight:
2-50 parts of acidic substance, 5-30 parts of diethyl ether, 5-30 parts of ethanol, 5-20 parts of glycerol and 5-20 parts of benzyl alcohol.
Further, the composition comprises, in parts by weight:
20-50 parts of acidic substances, 20-30 parts of diethyl ether, 10-20 parts of ethanol, 5-10 parts of glycerol and 5-10 parts of benzyl alcohol.
Further, the acidic substance includes at least one of carbolic acid, salicylic acid, and fruit acid.
In a second aspect, the embodiment of the application provides the application of the composition in the first aspect in preparing a medicament for treating alopecia areata.
In a third aspect, embodiments of the present application provide a medicament for treating alopecia areata, the medicament comprising:
(i) The composition of any one of the first aspects; and
(Ii) A Chinese medicinal composition for promoting hair growth.
Further, the traditional Chinese medicine composition comprises the following components in parts by weight:
10-30 parts of polygonum multiflorum, 10-30 parts of cortex mori radicis, 5-20 parts of frankincense, 10-20 parts of ligusticum wallichii and 5-20 parts of trogopterus dung.
Further, the traditional Chinese medicine composition comprises the following components in parts by weight:
30 parts of polygonum multiflorum, 30 parts of white mulberry root-bark, 20 parts of frankincense, 10 parts of ligusticum wallichii and 10 parts of trogopterus dung.
In a fourth aspect, an embodiment of the present application provides a method for preparing the drug for treating alopecia areata according to any one of the third aspects, the method comprising:
mixing acidic substances, diethyl ether, ethanol, glycerol and benzyl alcohol according to a proportion to obtain a first mixture;
Mixing the components in the traditional Chinese medicine composition according to a proportion to obtain a second mixture;
and mixing the first mixture and the second mixture, processing, and filtering to obtain the medicine for treating alopecia areata.
Further, the steps of mixing and processing the first mixture and the second mixture, and filtering to obtain the medicine for treating alopecia areata comprise the following steps:
and standing the fully dissolved first mixture for 24-48 hours, then adding the second mixture, fully stirring, stirring once every 12-16 days, sealing, standing for 80-100 days, and filtering to obtain the medicine for treating alopecia areata.

Detailed Description
In order to make the objects, technical solutions and advantages of the embodiments of the present invention more clear, the technical solutions of the embodiments of the present invention will be clearly and completely described below. The specific conditions are not noted in the examples and are carried out according to conventional conditions or conditions recommended by the manufacturer. The reagents or apparatus used were conventional products commercially available without the manufacturer's attention.
It should be noted that, without conflict, the embodiments of the present application and features of the embodiments may be combined with each other. The present application will be described in detail with reference to specific examples.
The general idea of the technical scheme provided by the embodiment of the invention is as follows:
In a first aspect, an embodiment of the present application provides a composition for treating alopecia areata, including, in parts by weight:
2-50 parts of acidic substance, 5-30 parts of diethyl ether, 5-30 parts of ethanol, 5-20 parts of glycerol and 5-20 parts of benzyl alcohol.

The composition for treating alopecia areata provided by the embodiment of the application has the following functions:
1. Acidic substance: the main function is to remove the cuticle of the follicular orifice, so that the medicine can permeate into the follicular orifice, and the effects of diminishing inflammation and sterilizing are achieved.
2. Diethyl ether: relieving spasm and relieving pain; deep into hair follicle thoroughly relieves the symptoms of cramps and reduces the feeling of pain caused by removal of the stratum corneum.
3. Benzyl alcohol: because benzyl alcohol has a certain anesthetic effect, penetration into hair follicles can have a certain antispasmodic effect, and also has the effect of eliminating inflammation.
4. Ethanol: since benzyl alcohol is dissolved in ethanol, the solubility of benzyl alcohol and the like is increased, so that several mixtures are sufficiently dissolved.
5. Glycerol: the function is to protect the local skin from moisture and chapping.
Therefore, the application combines the acidic substance, ethanol, diethyl ether, benzyl alcohol and glycerol with the specific proportion to achieve the effect under the combined action.
As an embodiment of the present examples, the composition comprises, in parts by weight:
20-50 parts of acidic substances, 20-30 parts of diethyl ether, 10-20 parts of ethanol, 5-10 parts of glycerol and 5-10 parts of benzyl alcohol.
The application further adopts the acidic substance, ethanol, diethyl ether, benzyl alcohol and glycerol with the specific proportion for matching use, and the effect is better.
As an embodiment of the present embodiment, the acidic substance includes at least one of carbolic acid, salicylic acid, and fruit acid.
The application further adopts acidic substances such as carbolic acid, salicylic acid, fruit acid and the like, has the main effects of removing the cuticle of the hair follicle, allowing the medicine to permeate in, and simultaneously has the effects of diminishing inflammation and sterilizing, and the effect is better. In some embodiments, fruit acids such as glycolic acid, lactic acid, malic acid, tartaric acid, citric acid, mandelic acid, and the like may be used.
In a second aspect, the embodiment of the application provides the application of the composition in the first aspect in preparing a medicament for treating alopecia areata.
When the composition of the first aspect of the application is applied to the preparation of the medicament for treating alopecia areata, if the composition is matched with a traditional Chinese medicine composition capable of promoting hair growth, the problems of long treatment course, low cure rate, large side effect and incapability of timely judging the treatment effect of a patient in the existing alopecia areata treatment method can be effectively solved.
In a third aspect, embodiments of the present application provide a medicament for treating alopecia areata, the medicament comprising:
(i) The composition of any one of the first aspects; and
(Ii) A Chinese medicinal composition for promoting hair growth.
The embodiment of the application provides a medicament for treating alopecia areata, which is realized based on the composition of any one of the first aspect, and at least has all the beneficial effects brought by the technical scheme of the embodiment because the medicament adopts part or all of the technical scheme of the medicament, which are not repeated herein.
The traditional Chinese medicine composition for promoting hair growth provided by the application specifically refers to a traditional Chinese medicine composition capable of providing nutrition for hair growth and/or playing a role in promoting hair growth. In some specific embodiments, the traditional Chinese medicine composition may be as disclosed in the prior art (chinese patent CN104800263a, a traditional Chinese medicine composition for treating alopecia areata, and a preparation method and application thereof), and the traditional Chinese medicine composition comprises the following components in parts by weight: 30 parts of safflower, 30 parts of biota orientalis and 30 parts of ginseng leaf; or a traditional Chinese medicine composition which is disclosed in the second prior art (Chinese patent CN114984144A, a traditional Chinese medicine composition for treating alopecia areata) and consists of 20 parts of semen cuscutae, 15 parts of Chinese angelica, 10 parts of prepared fleece-flower root, 10 parts of fructus psoraleae, 15 parts of medlar, 20 parts of prepared rehmannia root, 15 parts of achyranthes root, 15 parts of Chinese yam, 15 parts of fructus corni, 12 parts of rhizoma alismatis, 15 parts of poria cocos, 10 parts of gastrodia elata, 10 parts of radix sileris and 15 parts of cortex moutan.
In some embodiments, the weight ratio of the composition of any of the first aspects to the hair growth promoting traditional Chinese medicine composition may be (3-5): (1-2) or the like, such as 3:1, 4:1, 5:1, 5:2 or the like.
As an implementation mode of the embodiment of the application, the traditional Chinese medicine composition comprises the following components in parts by weight:
10-30 parts of polygonum multiflorum, 10-30 parts of cortex mori radicis, 5-20 parts of frankincense, 10-20 parts of ligusticum wallichii and 5-20 parts of trogopterus dung.
The application adopts the traditional Chinese medicine composition composed of 10-30 parts of polygonum multiflorum, 10-30 parts of white mulberry root-bark, 5-20 parts of frankincense, 10-20 parts of ligusticum wallichii and 5-20 parts of trogopterus dung, has better synergistic effect with the composition in any one of the first aspect, and can effectively solve the problems of long treatment course, low cure rate, large side effect and incapability of timely judging the treatment effect of patients in the existing alopecia areata treatment method.
The functions of the components in the traditional Chinese medicine composition are as follows:
1. polygonum multiflorum Thunb: increasing hair nutrition and promoting hair growth.
2. Cortex Mori: increasing hair nutrition and promoting hair growth.
3. Mastic gum: promoting blood circulation.
4. Ligusticum wallichii: promoting blood circulation.
5. Trogopterus dung: promoting blood circulation and relieving pain.
Therefore, the application combines the polygonum multiflorum, the white mulberry root-bark, the frankincense, the ligusticum wallichii and the trogopterus dung which are prepared in the specific proportion into the traditional Chinese medicine composition, and the traditional Chinese medicine composition is matched with the composition in any one of the first aspect, and achieves the effect of effectively treating alopecia areata by solving the problems of hair follicle spasm, hair follicle nutrition supply and absorption.
As an implementation mode of the embodiment of the application, the traditional Chinese medicine composition comprises the following components in parts by weight:
30 parts of polygonum multiflorum, 30 parts of white mulberry root-bark, 20 parts of frankincense, 10 parts of ligusticum wallichii and 10 parts of trogopterus dung.
The application adopts the polygonum multiflorum, the white mulberry root-bark, the frankincense, the ligusticum wallichii and the trogopterus dung which are prepared into the traditional Chinese medicine composition with the specific proportion, and the synergistic effect of the traditional Chinese medicine composition and the composition in any one of the first aspect is better.
In a fourth aspect, an embodiment of the present application provides a method for preparing the drug for treating alopecia areata according to any one of the third aspects, as shown in fig. 1, where the preparation method includes:
mixing acidic substances, diethyl ether, ethanol, glycerol and benzyl alcohol according to a proportion to obtain a first mixture;
Mixing the components in the traditional Chinese medicine composition according to a proportion to obtain a second mixture;
and mixing the first mixture and the second mixture, processing, and filtering to obtain the medicine for treating alopecia areata.
The embodiment of the application provides a preparation method of the medicine for treating alopecia areata, which is simple to operate without additional special equipment, and is characterized in that the first mixture and the second mixture are mixed and processed and filtered, and the obtained filtrate is the medicine.
As an implementation manner of the embodiment of the present application, the steps of mixing and processing the first mixture and the second mixture, and filtering to obtain the medicine for treating alopecia areata include:

In addition, the application method of the medicine for treating alopecia areata provided by the embodiment of the application is very simple, and the medicine can specifically comprise the following steps:
1. Firstly, medical alcohol is used for wiping the alopecia areata and the alopecia part for disinfection.
2. The medicine for treating alopecia areata provided by the application is dipped with a medical cotton swab, slightly wiping the alopecia areata part once, staying for ten seconds, then dipping the medicine again with a new cotton swab, smearing the medicine on the alopecia areata part again, repeatedly and uniformly wiping with a medical cotton swab, staying for ten seconds, repeating the process of dipping the medicine for the second time for the third time, at the moment, the scalp color is seen to turn white, the follicular orifice is clearly visible and can be stopped, if the scalp is not clearly repeated once again, and after staying for one minute, the residue on the alopecia areata part is removed with a medical gauze.
In summary, the present invention at least solves the following problems for patients suffering from alopecia areata:
1. the treatment cycle is long; the invention does not need oral medicine, is directly applied to the alopecia areata part by local external application, and can be solved in a few minutes, and has the advantages of simple operation, convenient use, quick and obvious effect and short treatment process.
2. The curative effect is not definite; the invention can clearly tell the patient that the hair cannot grow out by observing the skin change, and the growth cycle can also be determined, and the hair can grow out by clearly seeing the penetration of the hair follicle, and the normal hair can naturally grow out within 7-90 days.
3. The problem of large side effects; the invention has the advantages of local treatment, only removal of horny layer, no atrophy and depression, no hormone influence on liver and kidney functions and small side effect.
4. The problem of easy recurrence; the invention expands a little range in the smearing process, has good treatment effect, and solves the problem that the expansion of the alopecia areata range is easy to repeat.
It should be noted that:
1. Five days after the treatment of the alopecia areata part, the bathing, the soaking, and the like are not needed, so as to prevent bacteria from entering hair follicles to cause infection.
2. The medicine for treating alopecia areata provided by the embodiment of the application has poor effects on alopecia totalis and alopecia universalis.
3. The medicine for treating alopecia areata provided by the embodiment of the application is not suitable for eyebrows, beard, body hair and other parts.
The features and capabilities of the present invention are described in further detail below in connection with the examples.
Examples 1 to 9 and comparative examples 1 to 5 provide a composition for treating alopecia areata, and the specific amounts of each of the components in examples 1 to 5 and comparative examples 1 to 5 in parts by weight (unit: g) are shown in Table 1; wherein the acidic material in examples 1-5, example 9 and comparative examples 1-5 is carbolic acid (i.e., phenol), the acidic material in example 6 is salicylic acid, and the acidic material in example 8 is citric acid.

Examples 10 to 20 and comparative examples 6 to 10 provide a medicament for treating alopecia areata, wherein: examples 10 to 18 provide a medicament for treating alopecia areata, and the compositions for treating alopecia areata, which are provided in examples 1 to 9, are used together with a traditional Chinese medicine composition capable of promoting hair growth, respectively, so as to obtain the medicament for treating alopecia areata; examples 19 and 20 the composition for treating alopecia areata provided in example 9 was used in combination with a hair growth promoting Chinese medicinal composition; comparative examples 1 to 5 provide a composition for treating alopecia areata, which is used as comparative examples 6 to 10 in combination with a Chinese medicinal composition for promoting hair growth, respectively. The medicaments in examples 10 to 20 and comparative examples 6 to 10 comprise, in parts by weight (unit: g):
(i) Any of examples 1-9 and comparative examples 1-5 provide a composition for treating alopecia areata; (ii) a Chinese medicinal composition for promoting hair growth.
The specific amounts of the components in examples 10 to 20 and comparative examples 6 to 10 are shown in Table 2.

The preparation method of the drug for treating alopecia areata in the above examples 10 to 20 and comparative examples 6 to 10 comprises:
Mixing the composition for treating alopecia areata according to a proportion to obtain a first mixture;
mixing and crushing the components in the traditional Chinese medicine composition according to a proportion to obtain a second mixture;
And standing the fully dissolved first mixture for 24 hours, mixing the first mixture and the second mixture according to the weight ratio of 3:1, fully stirring, stirring once every 15 days, sealing, standing for 90 days, filtering, and collecting filtrate to obtain the medicine for treating alopecia areata.
Test case
In this example, the drugs for treating alopecia areata provided in examples 10 to 20 and comparative examples 6 to 10 were subjected to treatment effect test.
The testing method comprises the following steps: the medicine is rubbed on the alopecia areata part of the patient, the medicine is rubbed once in the first day, the medicine is not used for the second time if the hair follicle is observed to grow out by naked eyes after 20 days, and the medicine is rubbed again after 30 days apart if no new hair grows out. Each wiping process is as follows:
1. firstly, wiping the alopecia areata and the alopecia part of the patient with medical alcohol for disinfection.
2. The medicine for treating alopecia areata is dipped with a medical cotton swab, the alopecia areata is gently wiped once (the cotton swab is fully wetted), after the medicine is stopped for ten seconds, the medicine is dipped with a new cotton swab again and smeared on the alopecia areata, the wiping process is repeated for 3 times, and finally the medicine gauze is used for removing residues on the alopecia areata after the medicine is stopped for one minute.
Test object: 240 qualified subjects suffering from alopecia areata (at least 30% of scalp area affected, patients without alopecia totalis and alopecia universalis) were randomly divided into 16 groups of 15 persons each, each group being rubbed with the drugs for treating alopecia areata provided in examples 10 to 20 and comparative examples 6 to 10, respectively.
The test results after the subjects rubbed the drugs for treating alopecia areata provided in examples 10 to 20 and comparative examples 6 to 10 are shown in table 3.

(Ii) A hair growth promoting Chinese medicinal composition;
The composition comprises the following raw materials in parts by weight: 20-50 parts of acidic substances, 20-30 parts of diethyl ether, 10-20 parts of ethanol, 5-10 parts of glycerol and 5-10 parts of benzyl alcohol; the acidic substance is one of phenol, salicylic acid and citric acid;
the traditional Chinese medicine composition for promoting hair growth comprises the following raw materials in parts by weight: 10-30 parts of polygonum multiflorum, 10-30 parts of cortex mori radicis, 5-20 parts of frankincense, 10-20 parts of ligusticum wallichii and 5-20 parts of trogopterus dung.
2. The medicine for treating alopecia areata of claim 1, wherein the hair growth promoting Chinese medicinal composition comprises the following raw materials in parts by weight: 30 parts of fleece-flower root, 30 parts of white mulberry root-bark, 20 parts of frankincense, 10 parts of szechuan lovage rhizome and 10 parts of trogopterus dung.
3. A method for preparing the medicament for treating alopecia areata according to any one of claims 1 to 2, which is characterized by comprising the following steps:
mixing acidic substances, diethyl ether, ethanol, glycerol and benzyl alcohol according to a proportion to obtain a first mixture;
Mixing the components in the traditional Chinese medicine composition according to a proportion to obtain a second mixture;
mixing the first mixture and the second mixture, processing, and filtering to obtain a medicine for treating alopecia areata;
