Background
After a large amount of color cosmetics stay on the skin, the color cosmetics easily bring burden to the metabolism of the skin and delay the updating time of the color cosmetics. Especially in the eye, the appearance of aging is very pronounced, and over time the skin becomes aged and develops fine lines. In addition to accelerating aging, color cosmetics also deposit a large amount of pigment on the skin, which may eventually lead to a deepening of the color of the lips and even the color of the eyelids, and the skin color is remarkably deepened after a long time. For people who make up frequently, the common facial cleanser cannot clean the makeup residues on the skin, so that the condition of the skin is greatly changed, and the skin is dark and striking in a large oil field and acne muscles. Therefore, the skin is primarily cleaned, the deep cleaning is not thorough, and the skin care product cannot be well absorbed subsequently.
Skin care products and cosmetics are different in nature, the skin absorbs nutrients in the skin care products and is healthier and healthier, but if too much chemicals in the cosmetics with heavy texture are absorbed, pores are blocked, and the skin cannot permeate air gradually, so that the phenomena of acnes and large pores are caused. The makeup product has the advantages that the makeup product can immediately play the roles of skin color modification and flaw covering when being coated on the face, but the makeup product has good skin grinding effect and high pigment content, if makeup is not removed for a long time, the pigment in the makeup product can be deposited on the skin, the skin color slowly becomes dull and yellow, and the aging sign is generated.
Therefore, the design of the repairing and relieving cleaning formula is based on the dual effects of repairing and relieving the skin, and does not damage the skin and the skin barrier on the basis of cleaning the skin. For people who make up frequently and have incomplete cleaning, it is important to develop a composition which can clean the skin thoroughly and has no damage, repair and relieving effects on the skin.

(2) Subject selection
The selection standard is that the age is between 18 and 60 years, healthy male or female; those with positive lactic acid stinging; one of the symptoms of impaired skin barrier function such as dryness, desquamation, and redness of the skin is perceived; understanding the test course, voluntarily taking part in the trial and signing an informed consent form; the product can be used in required amount.
Exclusion criteria were pregnant or lactating women, or those who had a pregnancy plan recently; patients with psoriasis, eczema, atopic dermatitis, etc.; at present or within about one month, the patients are orally taken or externally applied with anti-inflammatory drugs such as corticosteroid hormone; other clinical trial participants are present or nearly one month old; the users do not participate voluntarily and cannot finish the required content on time; other clinical assessments considered unsuitable for participants;
withdrawal criteria are subjects who are considered inappropriate for continuing the study, e.g., by presenting an adverse event during the study or violating the study protocol; in other cases, the clinical study was withdrawn and the reason for withdrawal is indicated.
And (4) safety evaluation: all adverse skin reactions such as redness, stinging, itching, burning etc. that occurred during the study should be recorded and reported in time for severe adverse reactions.
The adverse reaction is judged according to the following three-level standard:
mild-easily tolerated, causes little discomfort, and does not affect normal life.
Moderate-enough to affect normal life.
Severe-hampering normal life.
Adverse reactions relationship to study:
irrelevant-clearly related to other factors; not possible-most likely related to other factors; possibly-after use, but also possibly due to other factors; it is possible-to occur after use, but not explained by other factors; probably-it occurs after use but cannot be explained by other factors and occurs again after use.
Subject: 2 patients with addict, 2 patients with abscission, 30 effective cases, 0 male and 32 female; minimum age: 18 years old, maximum age: age 57; the mean age was 33.09 years.
(3) Measurement procedure
At the first visit, subjects were instructed to perform the trial and signed written informed consent. Subjects were asked a series of questions about disease history, health status, etc., based on inclusion and exclusion criteria, etc., and compliance assessments were made and recorded.
Before the test is started, a face cleaning product without any efficacy is provided by a test center to clean the tested part of the testee; clean skin was tested after sitting still in a satisfactory room for at least 30 minutes. The factors of the cleaning product, the water temperature and the like should be kept consistent every return visit.
Pre-product skin baseline evaluation and testing, including instrumental testing and standard image capture and data recording, were performed on the enrolled qualified subjects. The same test was performed again on day 7 and day 14 after the product was used.
(4) The result of the calculation
Descriptive statistics: descriptive statistics are performed on the measured values of each test area, including quantity, mean, standard deviation, minimum, median, maximum.
And (3) differential analysis: the change rates of the TEWL average values on the sample application side and the control side were calculated, respectively, and then the difference between the sample application area and the control area at different measurement time points was statistically analyzed using the change rates. If the test data is normal distribution, performing statistical analysis by adopting a t test method; if the test data is in a non-normal distribution, a rank sum test method is adopted for statistical analysis. The statistical methods all adopt a two-tail test, and the test level is alpha =0.05.
(5) Determination of results
Positive results: compared with the control side, the change rate of the TEWL value of the sample smearing side is remarkably different, P is less than 0.05, and the tested sample has a promotion effect on the recovery of the transepidermal water loss of the skin with the damaged barrier.
Negative results: compared with the control side, the change rate of the TEWL value of the sample application side has no significant difference P which is more than or equal to 0.05, and the change rate indicates that the tested sample has no promotion effect on the recovery of the transepidermal water loss of the skin with damaged barrier.
TABLE 2 TEWL values of the test samples at different times
Statistical analysis of the experimental results in table 2 shows that the comparative examples and the examples have repairing effects to different degrees, but the change rate of the TEWL value of the water loss through the epidermis of all experimental groups is far higher than that of all experimental groups. Example 16 the transepidermal water loss value decreased by 11% after 7 days of use compared to before the sample, with a very significant difference, indicating that the composition had a repairing effect after 7 days of use. The proportion of the experimental group has a certain reduction rate, but the reduction degree of the experimental group has no significant difference. The control group data also shows almost no repair efficacy, and all the examples in the experimental group have better repair efficacy than the comparative examples, which proves that the repair efficacy is greatly enhanced by the synergistic cooperation of various substances in the composition.
3. Test for soothing efficacy
The method is carried out according to a clinical evaluation standard method of T/CNMIA 0015-2020 allergy-relieving type functional skin care products.
1. Lactic acid sting test
(1) Test samples: the test samples prepared by adding the compositions of examples 1 to 16 and comparative examples 1 to 6 to a common emulsion external base, wherein the stable cleansing composition having the repairing and soothing effects accounts for 5% of the total mass of the emulsion.
(2) The test method comprises the following steps: screening of lactate stinging sensitive muscles: a10% lactic acid solution (50. Mu.L) was dropped on a filter paper having a diameter of 8mm, and placed in the nasolabial folds at both sides of the subject, and the discomfort levels of itching, tingling, and burning sensation at the test site were evaluated by the subject at 30S, 2.5min, and 5min, respectively. Scoring was done on a 4 point scale (0 points no sensation, 1 points mild, 2 points moderate, 3 points severe). The patients with positive lactic acid stabbing pain are classified as if the sum of stabbing pain scores of 2.5min and 5min is more than or equal to 3. The subjects were divided on average into 22 groups (5 persons per group) based on the lactic acid sting screening results, and each group was smeared with a different test sample. The test product was used 1 time each day, morning and evening during the trial run. Taking the lactic acid sting screening score as a basic value, performing the lactic acid sting test again 28 days after the product is used, and performing statistical analysis on the lactic acid sting score before and after the product is used by using SPSS, wherein P <0.05 is a difference and has statistical significance.
TABLE 3 lactic acid sting scores for test samples at different times
As can be seen from the experimental results in table 3, examples 1 to 16 achieved the expected effect, and the stimulation of lactic acid to the skin was reduced, and after Wilcoxon paired rank sum test, the test level α =0.05, p <0.001 was obtained, which indicates that the lactic acid prick score of the subject who continuously used the test samples of examples 1 to 16 for 28 days was significantly different from the base value, and the prick degree was greatly reduced; while comparative examples 1 to 6 are improved compared with those before use, the expected effect is not achieved, the effect is only half of that of the examples, and the fact that the comparative examples may still have sensitivity in a small proportion is shown.
2. Hyaluronidase inhibition assay
Hyaluronidase is a hydrolase for degrading hyaluronic acid, is a main component of extracellular matrix of connective tissue of an organism and is related to most allergic reactions, so that the hyaluronidase in vitro inhibition experiment can react the effects of relieving, resisting allergy, resisting inflammation and the like of a product on skin to a certain extent; hyaluronic acid is a high-order polysaccharide consisting of D-glucuronic acid and N-acetylglucosamine, and is capable of undergoing a color reaction according to the Elson-morgan reaction. The inhibition rate of hyaluronidase in the sample can be determined by this reaction; the greater the inhibition rate, the better the soothing, anti-allergy or sensitivity inhibition effect.
(1) Test samples: the test samples were samples of examples 1 to 16 and comparative examples 1 to 6, and the concentrations were 0.5%, 4%, and 8%, respectively.
(2) The test method comprises the following steps: the test was performed according to the ELSON-MORGAN modified method.
TABLE 4 inhibition of hyaluronidase in samples at different concentrations (unit:%)
As can be seen from the experimental results in Table 4, the hyaluronidase inhibition results of examples 1-16 are significantly higher than those of comparative examples 1-6, indicating that the soothing, anti-allergy or sensitivity inhibition effects of examples 1-16 are better than those of comparative examples 1-6. Although the groups of comparative examples 1 to 6 also have a certain soothing and anti-allergy effect, the soothing effect is not obvious and is far from the examples, and the highest inhibition rate of the comparative example is 9.48% at a concentration of 8%, while the example is 75.17%, which is almost close to 8 times different. The synergistic effect of the components and the content of the components is obvious and obvious.
3. Visia image analysis method
(1) Test samples: the test samples prepared by adding the compositions of examples 1 to 16 and comparative examples 1 to 6 to a general emulsion external base, wherein the stable cleansing composition having the repairing and soothing effects accounts for 5% of the total mass of the emulsion.
(2) Subject selection: same lactic acid stabbing pain selected subjects
(3) Measurement procedure
Visia pictures were taken of D0 before using the sample, and Visia pictures were taken of D7 and D14 at 7 and 14 days, respectively. The results of analyzing the Visia facial red region picture with Image-Pro Plus 7.0C to obtain the red region a value are shown in Table 5.
TABLE 5 facial Red zone a values at different times for test samples
Statistical analysis of the experimental results in table 5 shows that the a values of examples 1-16 are significantly reduced and significantly different from those of 0 days in 7 days and 14 days, and P is less than 0.001. The compositions of examples 1 to 16 are described to have a promoting effect on the recovery of facial inflammation. The recovery effect of comparative examples 1 to 6 on inflammation is not obvious, the recovery effect of examples 1 to 16 on facial inflammation is obviously better than that of comparative examples 1 to 6, the red zone a value is reduced by 13% after the sample is used for 7 days, compared with the value before the sample is used, the sample has a very significant difference, the sample has an obvious effect of relieving facial redness after the sample is used for 7 days, the sample has been reduced to 12.85 on the 14 th day, and the reduction rate reaches 34%. The effective relieving capacity of the Chinese medicinal composition is proved, and the importance and the necessity of the synergistic effect of the formula in solving the problem of skin inflammation are further proved.
In conclusion, the caprylic/capric triglyceride, the PEG-20 glyceryl triisostearate, the boswellia resin extract, the tripeptide-1 copper, the camellia oil, the jojoba oil, the avocado oil, the shea butter, the jatropha wax and the rosa roxburghii seed oil are matched and act synergistically, and the results of a combination stability test, a repair efficacy test and a relieving efficacy test show that in the stability test, although the examples and the comparative examples are stable, the stability of the examples is superior to that of the comparative examples; in the test of repairing efficacy and relieving efficacy, the effect of the embodiment is obviously better than that of the comparative example, the comparative example may have sensitivity in a smaller proportion, and the effect of the embodiment is enhanced by about 8-10 times compared with that of the comparative example, which shows that the effect of the composition in repairing and relieving aspects of the invention is very obvious.
