Abstract

技术领域
本发明涉及用于促进牙根或牙周组织的生长、或者预防和治疗牙周炎的药物组合物,所述药物组合物包含木兰花(Magnolia flower)提取物、其分离物(fraction)、或从其中分离的化合物作为活性成分。
背景技术
牙周组织是包含由牙龈(gingiva)(齿龈(gums))、牙骨质、牙周韧带和牙槽骨组成的软组织和硬组织二者的复杂的器官。牙龈是涉及牙齿的支持和咀嚼过程的简单的结缔组织,而牙周韧带是高度分化的细胞结缔组织,其位于牙齿和牙槽骨之间从而支持牙齿并且将牙齿连接至颌骨、抵抗沿各种方向的机械应力如咀嚼力、并且还具有感觉功能。牙周韧带包括各种细胞如在牙槽骨侧的成骨细胞和破骨细胞,在牙周韧带的中央的成纤维细胞、马拉色氏上皮剩余(epithelial rests of Malassez)、巨噬细胞、未分化的间充质细胞、神经因子、血管内皮细胞,以及在牙根侧的成牙骨质细胞,并且已知牙周韧带涉及牙槽骨、牙根和牙骨质的恢复和再生。
牙龈是在口腔中观察到的牙齿支持组织的一部分,由此发展疾病(龈炎)。当疾病恶化并且深入地扩散至支持组织中时,可以破坏结合至围绕牙根的骨组织的牙周韧带并且还可以破坏覆盖牙齿的牙槽骨,导致牙周疾病。
牙周疾病是一种由细菌引起的牙齿支持组织中的炎症,其可以分为龈炎和牙周炎。该疾病的原因是由于不良口腔卫生导致的由口腔细菌形成牙菌斑。牙菌斑是在使用包含在唾液中的粘性物质作为粘合剂粘附在牙齿的表面上之后在其上增殖的大量的细菌。如果对牙菌斑不进行治疗,则炎症发展从而引起牙龈出血和口臭,其称为龈炎。
一旦龈炎进展,牙齿和牙龈之间的间隙变得更深,结果,形成牙周袋,其中引起牙周疾病的细菌增殖从而引起牙周炎。随着牙周炎进展,例如刷牙等弱刺激可以引起牙龈出血和牙龈肿胀、急性炎症和疼痛。这样的炎症降低骨形成功能但是增强骨吸收功能,从而使牙槽骨逐渐降低,引起牙槽骨被破坏并且最终失去牙齿。进展的牙周炎可以引起牙齿脱落。在超过40岁的成年人中,牙周炎占拔牙的原因的40%,表明由于牙周炎的牙齿脱落是一个严重的问题。
牙周疾病中的牙周韧带和牙槽骨的破坏是由于胶原(这些组织的主要组分)的降解导致的。虽然这样的破坏的精确的机理尚未公开,但是已知胶原的降解是由宿主中分泌的胶原酶和由引起牙周疾病的细菌分泌的中性蛋白酶引起的。因此,已经进行了通过抑制胶原酶和蛋白酶的活性来抑制牙周疾病的进展的研究。
对于牙周疾病的治疗,已经尝试了患者的口腔卫生的改善、非手术或手术牙科刮治术(dental scaling)、牙根平整术(dental root planing)、牙龈刮除术(gingivalcurettage)和使用新的附着物(attachment)的牙周再生。然而,这样的手术治疗方法引起访问牙科诊所的不便并且无法保证有效的治疗且仅限于进展的疾病,表明使用该治疗没有预防效果。如果不治疗该疾病,则其在大多数情况下进展为慢性疾病。已经使用全身性抗生素(Systemic antibiotic)和缓释制剂作为补充治疗。然而,存在药物过多地递送至不必要的区域而导致副作用的严重的问题。最近,已经报道了对抗生素显示抗性的牙周致病菌(periodontopathic bacteria),这使情况更糟糕。当牙齿由于牙周疾病脱落时,需要假牙修复治疗(prosthodontic treatment)或牙科植入治疗(dental implant treatment),增加了患者的负担。特别地,其对于社会和经济上的弱势群体会是更高的医疗障碍。因此,如果受损的组织可以在牙齿脱落之前再生,则可以通过减少由牙齿脱落导致的对例如人造牙齿或假体等治疗的需求来缓解经济和社会负担。
为了克服手术治疗的局限性和使用抗生素的问题从而增强牙周疾病的预防和治疗的效果,迫切地需要开发在恢复受损和失去的牙周组织方面有效的新的药物。牙周组织均具有不同的结构特征从而难以建立有效的治疗方法来使由牙周疾病损坏或破坏的损伤再生。通过例如斑块和结石等炎性结缔组织的非手术或手术移除可以使牙周疾病改善至愈合阶段。然而,由于形成牙周组织的各组分无法完全恢复,因此被破坏的牙周组织无法通过这样的传统的治疗方法彻底恢复。
木兰花是属于木兰科(Magnoliaceae)的木兰的花芽的名称,其味道微辣。在韩国,使用玉兰(Magnolia denudata Desrousseaux)或其同属的植物的干燥的花芽。在日本,使用玉兰(M.denudata Desr)、望春花(M.flower Pamp)、日本辛夷(M.kobus DC)、柳叶木兰(M.salicifolia Maxim)和武当玉兰(M.sprengeri Pamp)。在中国,使用玉兰、望春花和武当玉兰。在东方医学(oriental medicine)中,已知的是木兰花性温并且在治疗头痛、下背痛、鼻炎以及鼻窦炎方面是有效的。根据之前的报道,其还具有止痛的活性。
美国专利申请No.US 13/739,340描述了促进成骨作用的药物组合物,其源自属于木兰科的那些植物的叶或树皮的提取物,这是因为已经已知它们没有副作用但是可以促进骨生长和增加骨密度。人类骨骼借助成骨细胞和破骨细胞的作用通过骨构建(bonemodeling)和骨重建(bone remodeling)来维持其体内平衡(homeostasis)。在成骨作用的早期,透明软骨以骨的形式形成并且其后钙沉积,其间软骨消失、骨生长并且软骨细胞变成成骨细胞。
牙齿通过与以上骨形成过程不同的过程形成。特别地,当细胞在出生6周时开始发育时,在内陷的结缔组织中,表面上皮开始侵入并且细胞开始增殖。随着内陷的细胞在上皮中生长,牙周组织在下方形成。上皮上的细胞分化为成釉细胞从而覆盖牙齿,而形成牙本质的成牙质细胞和各种牙髓细胞在内部分化。如上所述,骨形成和牙齿形成中涉及的机理和细胞彼此完全不同。
在寻找可以预防由牙周炎引起的牙周组织的丧失和促进牙周组织的再生的材料的研究的过程中,本发明人确认从木兰花提取物分离的辛夷脂素可以促进牙根的生长并且有效地在牙齿周围诱导牙槽骨形成,导致了本发明的完成。
发明内容
本发明的目的在于提供用于促进牙根或牙周组织的生长、或者预防或治疗牙周炎的药物组合物,其包含木兰花提取物、其分离物或从其中分离的化合物作为活性成分。

2)通过将包含乙酸乙酯和己烷的混合溶剂添加至步骤1)的甲醇分离物中来获得洗脱液;和
3)使步骤2)的洗脱液中的晶体析出,随后将其用过滤器过滤。

下文中,详细地描述本发明。
本发明提供用于促进牙根或牙周组织的生长、或者预防或治疗牙周炎的药物组合物,其包含木兰花提取物或从其中分离的分离物作为活性成分。
本发明还提供用于促进牙根或牙周组织的生长、或者预防或治疗牙周炎的方法,其包括将药学上有效的剂量的木兰花提取物或从其中分离的分离物给予受试者的步骤。
本发明还提供包含木兰花提取物或从其中分离的分离物作为活性成分的药物组合物用于促进牙根或牙周组织的生长、或者预防或治疗牙周炎的用途。
木兰花可以购买或栽培。可以使用木兰花的所有部分如茎、叶、根和花芽等,但是在本发明的优选的实施方案中,使用花芽。
用于木兰花的提取的方法可以为本领域接受的常规的方法之一,其例举为过滤、热水提取、脂吸法(enfleurage)、回流提取和超声提取。当提取通过热水提取进行时,提取次数优选为1~5次并且更优选3次,但是不总是限于此。可以将提取溶剂按干燥的木兰花的体积的0.1~10倍的体积添加至干燥的木兰花中、并且更优选按干燥的木兰花的体积的0.3~5倍添加。提取温度优选为20℃~40℃并且提取时间优选为12~48小时。
木兰花提取物或其分离物可以通过包括以下步骤的制备方法来制备:
1)将木兰花干燥从而除去水分然后将干燥的木兰花粉碎;
2)将干燥的木兰花在室温下用提取溶剂提取;
3)将步骤2中获得的提取物过滤,随后是减压浓缩;和
4)通过添加有机溶剂将提取物分离。
在以上方法中,步骤1)中的干燥优选通过减压干燥、真空干燥、沸腾干燥(boilingdrying)、喷雾干燥或冷冻干燥来进行,但是不总是限于此。
在以上方法中,步骤2)的提取溶剂可以为水、C1~C2低级醇、或其混合溶剂。
在本方法中,步骤3)中的减压浓缩优选通过使用真空浓缩器或真空旋转蒸发器来进行,但是不总是限于此。
在以上方法中,步骤4)的有机溶剂可以为乙酸乙酯、己烷、苯或氯仿。
所述分离物可以通过以下过程来制备:将木兰花提取物通过使用乙酸乙酯分离然后通过使用甲醇用分离物进行另外的分离;或者通过使用以3:7或5:5的比例包含乙酸乙酯和己烷的混合溶剂用分离物进行另外的分离。
在本发明的优选的实施方案中,本发明人确认当将牙齿包埋在包含木兰花提取物的琼脂糖珠中然后培养时,显著地促进了牙根的生长并且在牙齿周围新形成牙槽骨(见图4),表明木兰花提取物在促进牙根或牙周组织的生长方面是有效的。
关于上述实施方案的结果,当将牙齿从5日龄的小鼠拔出时,因为牙齿是在其处于在牙槽骨的内部的状态下取出的,因而不仅牙齿被拔出,而且可以帮助牙齿和附着至牙齿的牙根的生长的牙周韧带也被拔出。牙根和牙周组织通过拔出的牙齿中的样本(sample)生长。此时,包括在牙周韧带中的各种细胞相互作用从而帮助牙根的生长。牙根的外表面覆盖有称为牙骨质的硬组织,其连接至牙周韧带从而固定牙齿。牙骨质也由牙周韧带细胞形成。所以,包括在牙周韧带中的各种细胞之间的相互作用不仅诱导牙根的生长还围绕牙根形成牙骨质。因此,牙周韧带对于牙根和牙周组织的生长是重要因素。
因此,木兰花提取物或其分离物可以有效地用作用于促进牙根或牙周组织的生长、或者预防或治疗牙周炎的药物组合物。
本发明的组合物优选以组合物的总重量的0.1~99.9重量%包括本发明的木兰花提取物,但是不总是限于此。所述组合物可以另外包括药学上可接受的载体、赋形剂或稀释剂。
本发明的组合物可以口服地或胃肠外地给予和以药物制剂的一般形式使用。即,本发明的组合物可以通过与通常使用的稀释剂或赋形剂如填料、增量剂(extender)、粘合剂、润湿剂、崩解剂和表面活性剂混合来制备用于口服或胃肠外给药。用于口服给药的固体制剂为片剂、丸剂、散剂、颗粒剂和胶囊剂。通过将所述化合物与一种以上的适宜的赋形剂如淀粉、碳酸钙、蔗糖或乳糖、明胶等混合来制备这些固体制剂。除了简单的赋形剂以外,可以使用润滑剂例如硬脂酸镁、滑石等。用于口服给药的液体制剂为混悬剂、溶液剂、乳剂和糖浆剂,并且除了例如水和液体石蜡等通常使用的简单的稀释剂以外,上述制剂可以包含各种赋形剂如润湿剂、甜味剂、芳香剂和防腐剂。用于胃肠外给药的制剂为灭菌的水性溶液剂、水不溶性赋形剂、混悬剂、乳剂、冻干制剂和栓剂。
除了一种或多种活性化合物以外,水不溶性赋形剂和混悬剂可以包含丙二醇、聚乙二醇、植物油如橄榄油、可注射的酯如油酸乙酯等。除了一种或多种活性化合物以外,栓剂可以包含witepsol、聚乙二醇、吐温61、可可脂、月桂油、甘油明胶等。
本发明的组合物可以口服地或胃肠外地给予。本文中的胃肠外给药可以通过皮肤外应用(external skin application)、腹膜内注射、直肠内注射、静脉内注射、肌内注射、皮下注射、子宫内注射或脑室内注射来进行。其中,皮肤外应用是最优选的。
本发明的组合物的有效剂量可以根据体重、年龄、性别、健康状况、膳食、给药频率、给药方法、排泄和疾病的严重性来确定。剂量为每天0.01~1000mg/kg,优选每天30~500mg/kg,并且更优选每天50~300mg/kg,并且给药频率优选为一天1~6次。
本发明的组合物可以单独给予或与外科手术(surgical operation)、放射疗法、激素疗法、化学疗法和生物调节剂一起治疗。
本发明还提供用于促进牙根或牙周组织的生长、或者预防或改善牙周炎的准药物组合物,其包含木兰花提取物或从其中分离的分离物作为活性成分。

准药物组合物优选为口腔用组合物制剂如牙膏、漱口水或口腔清洁剂。
本发明的包含木兰花提取物或其分离物的准药物组合物的制剂不受限制并且可以以例如牙膏、漱口水或口腔清洁剂等常规的形式配制。本发明中提供的口腔用组合物可以包含制剂所需要的各种基质和添加剂,并且这些成分的种类和量可以由本领域技术人员容易地选择。例如,当将口腔用组合物配制为牙膏时,其可以包括研磨剂、润湿剂、发泡剂、粘合剂、甜味剂、pH调节剂、防腐剂、药用成分、矫味剂、增白剂、着色剂和溶剂。

本发明的健康功能食品可以另外包括各种调味剂(flavor)或天然碳水化合物。上述天然碳水化合物可以为以下的一种:单糖如葡萄糖和果糖,二糖如麦芽糖和蔗糖,多糖如糊精和环糊精,以及葡萄糖醇类如木糖醇、山梨糖醇和赤藓糖醇。此外,可以包括例如奇甜蛋白(thaumatin)和甜叶菊(stevia)提取物等天然甜味剂、和例如糖精和阿斯巴甜等合成甜味剂作为甜味剂。天然碳水化合物的含量在100重量份的本发明的健康功能食品中优选为0.01~0.04重量份并且更优选0.02~0.03重量份。
除了上述成分以外,本发明的健康功能食品可以包括各种营养物、维生素、矿物质、调味剂、着色剂、果胶酸和其盐、海藻酸和其盐、有机酸、保护性胶体增粘剂、pH调节剂、稳定剂、防腐剂、甘油、醇类、用于添加至苏打中的碳酸化剂等。所有提及的成分可以单独或一起添加。那些成分的混合比例实际上无关紧要,但是一般地,可以以相对于100重量份的本发明的健康功能食品为0.01~0.1重量份添加每一种。

在本发明的优选的实施方案中,本发明人确认当将牙齿包埋在包含从木兰花提取物分离的辛夷脂素的琼脂糖珠中然后培养时,显著地促进了牙根的生长并且在牙齿周围新形成牙槽骨(见图3),表明由式1或式2表示的化合物在促进牙根或牙周组织的生长、或者预防或治疗牙周炎方面是有效的。
本发明的由式1或式2表示的化合物可以以药学上可接受的盐的形式使用,其中所述盐优选为由药学上可接受的游离酸形成的酸加成盐。所述酸加成盐可以从无机酸如盐酸、硝酸、磷酸、硫酸、氢溴酸、氢碘酸、亚硝酸和亚磷酸,或者无毒的有机酸如脂肪族单/二羧酸盐、苯基取代的链烷酸盐、羟基链烷酸盐、链烷二酸盐(alkandioate)、芳香酸和脂肪族/芳香族磺酸获得。药学上无毒的盐例举为:硫酸盐、焦硫酸盐、硫酸氢盐、亚硫酸盐、亚硫酸氢盐、硝酸盐、磷酸盐、磷酸一氢盐、磷酸二氢盐、偏磷酸盐、焦磷酸盐、氯化物、溴化物、碘化物、氟化物、乙酸盐、丙酸盐、癸酸盐(decanoate)、辛酸盐、丙烯酸盐、甲酸盐、异丁酸盐、癸酸盐(caprate)、庚酸盐、丙炔酸盐(propiolate)、草酸盐、丙二酸盐、琥珀酸盐、辛二酸盐、癸二酸盐(cabacate)、富马酸盐、马来酸盐、丁炔-1,4-二酸盐、己烷-1,6-二酸盐、苯甲酸盐、氯苯甲酸盐、甲基苯甲酸盐、二硝基苯甲酸盐、羟基苯甲酸盐、甲氧基苯甲酸盐、邻苯二甲酸盐、对苯二甲酸盐、苯磺酸盐、甲苯磺酸盐、氯苯磺酸盐、二甲苯磺酸盐、苯乙酸盐、苯丙酸盐、苯丁酸盐、柠檬酸盐、乳酸盐、羟基丁酸盐、乙醇酸盐、苹果酸盐、酒石酸盐、甲磺酸盐、丙磺酸盐、萘-1-磺酸盐、萘-2-磺酸盐和扁桃酸盐。
本发明中的酸加成盐可以通过本领域技术人员已知的常规方法来制备。例如,将本发明的由式1或式2表示的化合物溶解在有机溶剂如甲醇、乙醇、丙酮、二氯甲烷或乙腈中,向其中添加有机酸或无机酸从而诱导析出。然后,将析出物过滤并且干燥从而得到盐。或者将溶剂和过量的酸在减压下蒸馏,并且干燥,从而得到盐。或者使析出物在有机溶剂中结晶从而得到盐。
药学上可接受的金属盐可以通过使用碱来制备。碱金属或碱土金属盐通过以下过程获得:将化合物溶解在过量的碱金属氢氧化物或碱土金属氢氧化物溶液中;将不溶性化合物盐过滤;将剩余的溶液蒸发并且将其干燥。此时,优选以钠、钾或钙盐的药学上适宜的形式制备金属盐。并且通过碱金属或碱土金属盐与合适的银盐(例如硝酸银)的反应来制备相应的银盐。
本发明不仅包括由式1或式2表示的化合物和其药学上可接受的盐,还包括可能由其产生的溶剂化物、水合物和异构体。
本发明还提供用于促进牙根或牙周组织的生长、或者预防或改善牙周炎的准药物组合物,其包含由式1或式2表示的化合物、其药学上可接受的盐、或其光学异构体作为活性成分。
本发明还提供包含由式1或式2表示的化合物、其药学上可接受的盐、或其光学异构体作为活性成分的准药物组合物用于促进牙根或牙周组织的生长、或者预防或改善牙周炎的用途。
本发明还提供用于促进牙根或牙周组织的生长、或者预防或改善牙周炎的健康功能食品,其包含由式1或式2表示的化合物、其药学上可接受的盐、或其光学异构体作为活性成分。
本发明还提供包含由式1或式2表示的化合物、其药学上可接受的盐、或其光学异构体作为活性成分的健康功能食品用于促进牙根或牙周组织的生长、或者预防或改善牙周炎的用途。
本发明还提供用于促进牙根或牙周组织的生长或者改善牙周炎的化妆品组合物,其包含由式1或式2表示的化合物、其药学上可接受的盐、或其光学异构体作为活性成分。
本发明还提供包含由式1或式2表示的化合物、其药学上可接受的盐、或其光学异构体作为活性成分的化妆品组合物用于促进牙根或牙周组织的生长或者改善牙周炎的用途。
在本发明的优选的实施方案中,本发明人确认当将牙齿包埋在包含从木兰花提取物分离的辛夷脂素的琼脂糖珠中然后培养时,显著地促进了牙根的生长并且在牙齿周围新形成牙槽骨(见图3),表明由式1或式2表示的化合物可以有效地用作用于促进牙根或牙周组织的生长、或者预防或改善牙周炎的准药物组合物、化妆品组合物或健康功能食品的活性成分。
此外,本发明提供用于分离由以上式1或式2表示的化合物的方法,其包括以下步骤:
1)通过将甲醇添加至通过用乙酸乙酯分离木兰花提取物生成的乙酸乙酯层中来生产甲醇分离物;
2)通过将包含乙酸乙酯和己烷的混合溶剂添加至步骤1)的甲醇分离物中来获得洗脱液;和
3)使步骤2)的洗脱液中的晶体析出,随后将其用过滤器过滤。
优选通过使用水、C1~C2低级醇、或其混合溶剂来提取以上木兰花提取物。步骤2的乙酸乙酯和己烷的混合溶剂以3:7或5:5的比例包含乙酸乙酯和己烷。
本发明的实际的和当前优选的实施方案如以下实施例中所示进行说明。
然而,将理解的是,本领域技术人员考虑本公开内容,可以在本发明的精神和范围内进行修饰和改善。
实施例1:木兰花提取物、分离物和化合物的制备
用于本发明的木兰花提取物、分离物和化合物通过以下方法来制备。
<1-1>木兰花提取物的制备
将甲醇以10倍花芽体积的体积添加至中国木兰花(购自Osaka,日本)中,随后提取3~7天。将提取物过滤从而除去不溶性残渣,并且将经过滤的提取物使用真空浓缩器浓缩。然后,固体木兰花提取物通过将浓缩的提取物冷冻干燥来获得。
<1-2>木兰花分离物的制备
将实施例<1-1>中获得的提取物用乙酸乙酯和蒸馏水分离。然后,将蒸馏水层用丁醇分离,并且将乙酸乙酯层用90%甲醇和己烷进一步分离。将90%甲醇层(21.17g)使用硅胶与己烷和乙酸乙酯的混合溶剂(乙酸乙酯:己烷)分离成7份洗脱液(1:9、2:8、3:7、5:5、100%乙酸乙酯,和100%甲醇)。
<1-3>化合物分离
在以上实施例<1-2>中获得的7份洗脱液中,使两种洗脱液(乙酸乙酯:己烷=3:7和5:5)中的晶体析出。将晶体用过滤器过滤,作为结果,获得0.44g和0.53g的辛夷脂素。
木兰花提取物、分离物和所述化合物的制备过程在图1中说明(图1)。
实施例2:用于增加木兰花提取物中的活性成分含量的溶剂分离
为了增加木兰花提取物中的木酚素(lignan)化合物辛夷脂素和木兰脂素的含量,用如下所示的不同的条件获得分离物并且测量辛夷脂素和木兰脂素的含量。
<2-1>通过pH进行溶剂分离
将1.2L的70%乙醇(v/v)添加至100g的切碎的木兰花中,随后在85℃下回流提取6小时。然后,将提取物过滤,并且将残渣用70%乙醇洗涤。收集滤液并且减压浓缩。结果,获得了17g的70%乙醇提取物。将水添加至以上获得的提取物中并且完全溶解。然后,根据条件(pH 5.0、pH 3.0和pH 9.0)调节pH。将正己烷添加至水中,随后进行数次溶剂分离。在减压下进行浓缩。辛夷脂素和木兰脂素的含量通过辛夷脂素含量试验和指纹图谱分析(fingerprint pattern assay)来分析。
作为结果,如表1中所示,在其中未调节pH的对照组中,在己烷分离物和水分离物二者中均检测到辛夷脂素和木兰脂素。此时,水分离物的回收率高于己烷分离物的回收率(表1)。

如表2中所示,当分离在酸性条件(pH 3.0)下进行时,总体回收率下降,这是因为木酚素的双四氢呋喃环(furofuran ring)开环并且分解。

如表3中所示,当分离在碱性条件(pH 9.0)下进行时,与酸性条件下的总体回收率相比,总体回收率增加,并且相对含量由于其它材料对碱性条件反应而增加。

实验例1:从木兰花提取物分离的辛夷脂素对牙根或牙周组织的生长促进效果
为了研究从木兰花提取物分离的辛夷脂素对牙根或牙周组织的生长的促进效果,进行以下实验。
特别地,将实施例1中获得的辛夷脂素以80μm的终浓度在PBS(DainipponPharmaceuticals Inc.,日本)中稀释,并且将4个琼脂糖珠(Affi-Gel Blue Gel,#153-7301,Bio-Rad Laboratories Inc.,日本)用PBS洗涤,随后反应至少60分钟从而使辛夷脂素吸收在那些珠中。对于对照,使其它4个珠仅与PBS反应。将5日龄的C57BL/6小鼠(SankyoLabo Service Corporation,日本)处死,将臼齿从其下颌骨拔出。将拔出的臼齿的牙髓包埋在包含辛夷脂素的琼脂糖珠中。对于对照,将拔出的臼齿的牙髓包埋在仅包含PBS的琼脂糖珠中。将琼脂糖珠各自植入至8周龄的C57BL/6宿主小鼠(雌性,Sankyo Labo ServiceCorporation,日本)肾脏的囊(capsule)中。将宿主小鼠在12h/12h光照和黑暗循环、室温和自由进食的条件下饲养。3周后,将小鼠处死,从其中摘除肾脏。然后,观察牙齿。牙齿的3D图像通过显微CT(Rigaku R_mCT2)来拍摄。
作为结果,如图3中所示,在PBS对照组中几乎观察不到牙根和骨的生长,而在辛夷脂素实验组中,牙根的生长是显著的并且观察到牙槽骨在植入的牙齿周围的形成(图3)。因此,确认的是从本发明的木兰花提取物分离的辛夷脂素化合物在促进牙根的生长或牙周组织的形成方面是有效的。
实验例2:木兰花提取物对牙根和牙周组织的生长促进效果
为了研究木兰花提取物对牙根和牙周组织的生长的促进效果,进行以下实验。
特别地,将实施例1中获得的木兰花提取物以50和500μg/ml的终浓度在PBS中稀释。将4个琼脂糖珠(Affi-Gel Blue Gel,#153-7301,Bio-Rad Laboratories Inc.,韩国)用PBS洗涤,随后反应至少60分钟从而使辛夷脂素吸收在那些珠中。对于对照,使其它4个珠仅与PBS反应。对于比较组,将珠如实验例1中所示与辛夷脂素化合物反应。将5日龄的C57BL/6小鼠(DBL Co.,Ltd.,韩国)处死,将臼齿从其下颌骨拔出。将拔出的臼齿的牙髓包埋在包含辛夷脂素或木兰花提取物的琼脂糖珠中。对于对照,将拔出的臼齿的牙髓包埋在仅包含PBS的琼脂糖珠中。将琼脂糖珠各自植入至8周龄的C57BL/6宿主小鼠(雌性,DBLCo.,Ltd.,韩国)肾脏的囊中。将宿主小鼠在12h/12h光照和黑暗循环、室温和自由进食的条件下饲养。3周后,将小鼠处死,从其中摘除肾脏。然后,观察牙齿。牙齿的3D图像通过显微CT来拍摄。
作为结果,如图4中所示,在PBS对照组中几乎观察不到牙根和骨的生长,而牙根的生长在实验组中是木兰花提取物剂量依赖性地显著的,并且观察到牙槽骨在植入的牙齿周围的形成(图4)。因此,确认的是木兰花提取物在促进牙根的生长和牙周组织的形成方面是有效的,与辛夷脂素化合物相当或更好。
下文中,描述了包含本发明的木兰花提取物、其分离物、或从其中分离的化合物作为活性成分的药物组合物、准药物组合物和化妆品组合物的制造例。
制造例1:药物制剂的制备
<1-1>片剂的制备(直接加压)
根据用于制备片剂的常规的方法,将所有以上组分以规定的量添加、均一地混合并且压制从而制备片剂。
<1-2>片剂的制备(浆料制粒(slurry granulation))
在将活性成分筛分之后,添加并且混合乳糖和淀粉。在将聚山梨醇酯溶解在纯水中之后,将适量的溶液添加至活性成分、乳糖和淀粉的混合物中,随后雾化。在干燥之后,将颗粒筛分并且与胶体二氧化硅和硬脂酸镁混合。将颗粒加压从而制备片剂。
制造例2:准药物组合物的制备
<2-1>口腔含漱液的制备

Claims (27)
1.一种用于促进牙根或牙周组织的生长、或者预防或治疗牙周炎的药物组合物,其包含木兰花提取物或从其中分离的分离物作为活性成分。
2.根据权利要求1所述的用于促进牙根或牙周组织的生长、或者预防或治疗牙周炎的药物组合物,其中通过使用水、C1~C2低级醇、或其混合溶剂作为提取溶剂来提取所述木兰花提取物。
3.根据权利要求1所述的用于促进牙根或牙周组织的生长、或者预防或治疗牙周炎的药物组合物,其中通过将所述木兰花提取物用乙酸乙酯、己烷、苯或氯仿分离来获得所述分离物。
4.根据权利要求1所述的用于促进牙根或牙周组织的生长、或者预防或治疗牙周炎的药物组合物,其中所述分离物通过以下来获得:将所述木兰花提取物用乙酸乙酯分离然后通过使用甲醇用分离物进行另外的分离;或者通过使用以3:7或5:5的比例包含乙酸乙酯和己烷的混合溶剂用分离物进行另外的分离。
5.一种用于促进牙根或牙周组织的生长、或者预防或改善牙周炎的准药物组合物,其包含木兰花提取物或从其中分离的分离物作为活性成分。
6.根据权利要求5所述的用于促进牙根或牙周组织的生长、或者预防或改善牙周炎的准药物组合物,其中所述准药物组合物为牙膏、漱口水或口腔清洁剂的形式。
7.一种用于促进牙根或牙周组织的生长、或者预防或改善牙周炎的健康功能食品,其包含木兰花提取物或从其中分离的分离物作为活性成分。
8.一种用于促进牙根或牙周组织的生长或者改善牙周炎的化妆品组合物,其包含木兰花提取物或从其中分离的分离物作为活性成分。
9.一种用于促进牙根或牙周组织的生长、或者预防或治疗牙周炎的药物组合物,其包含由以下式1或式2表示的化合物、其药学上可接受的盐、或其光学异构体作为活性成分:

12.根据权利要求11所述的用于促进牙根或牙周组织的生长、或者预防或改善牙周炎的准药物组合物,其中所述准药物组合物为牙膏、漱口水或口腔清洁剂的形式。
13.一种用于促进牙根或牙周组织的生长、或者预防或改善牙周炎的健康功能食品,其包含由式1或式2表示的化合物、其药学上可接受的盐、或其光学异构体作为活性成分。
14.一种用于促进牙根或牙周组织的生长或者改善牙周炎的化妆品组合物,其包含由式1或式2表示的化合物、其药学上可接受的盐、或其光学异构体作为活性成分。
15.一种用于分离由以上式1或式2表示的化合物的方法,其包括以下步骤:
1)通过将甲醇添加至通过用乙酸乙酯分离木兰花提取物生成的乙酸乙酯层中来生产甲醇分离物;
2)通过将包含乙酸乙酯和己烷的混合溶剂添加至步骤1)的所述甲醇分离物中来获得洗脱液;和
3)使步骤2)的所述洗脱液中的晶体析出,随后将其用过滤器过滤。
16.根据权利要求15所述的用于分离由式1或式2表示的化合物的方法,其中通过使用水、C1~C2低级醇、或其混合溶剂作为提取溶剂来提取所述木兰花提取物。
17.根据权利要求15所述的用于分离由式1或式2表示的化合物的方法,其中步骤2的所述混合溶剂以3:7或5:5的比例包含乙酸乙酯和己烷。
18.一种用于促进牙根或牙周组织的生长、或者预防或治疗牙周炎的方法,其包括将药学上有效的剂量的木兰花提取物或从其中分离的分离物给予受试者的步骤。
19.包含木兰花提取物或从其中分离的分离物作为活性成分的药物组合物用于促进牙根或牙周组织的生长、或者预防或治疗牙周炎的用途。
20.包含木兰花提取物或从其中分离的分离物作为活性成分的准药物组合物用于促进牙根或牙周组织的生长、或者预防或改善牙周炎的用途。
21.包含木兰花提取物或从其中分离的分离物作为活性成分的健康功能食品用于促进牙根或牙周组织的生长、或者预防或改善牙周炎的用途。
22.包含木兰花提取物或从其中分离的分离物作为活性成分的化妆品组合物用于促进牙根或牙周组织的生长或者改善牙周炎的用途。
23.一种用于促进牙根或牙周组织的生长、或者预防或治疗牙周炎的方法,其包括将药学上有效的剂量的由式1或式2表示的化合物、其药学上可接受的盐、或其光学异构体给予受试者的步骤。
24.包含由式1或式2表示的化合物、其药学上可接受的盐、或其光学异构体作为活性成分的药物组合物用于促进牙根或牙周组织的生长、或者预防或治疗牙周炎的用途。
25.包含由式1或式2表示的化合物、其药学上可接受的盐、或其光学异构体作为活性成分的准药物组合物用于促进牙根或牙周组织的生长、或者预防或改善牙周炎的用途。
26.包含由式1或式2表示的化合物、其药学上可接受的盐、或其光学异构体作为活性成分的健康功能食品用于促进牙根或牙周组织的生长、或者预防或改善牙周炎的用途。
