Abstract

本发明涉及一种新型的、检测EB病毒抗体的显 色试剂及其制造方法。它由染色液和复染剂所组 成。染色液包括3-氨基-9-乙基咔唑(AEC)、N, N-二甲基甲酰胺、过氧化脲和醋酸缓冲液;复染剂为 0.5%固绿水溶液。使用该显色试剂,可提高被检血 清的阳性细胞百分率,避免假阳性,使判断更加客观、 准确,特别适用于基层医疗单位使用。

Description

本发明涉及一种新型的、检测EB病毒抗体的显色试剂及其制造方法。
以往检测EB病毒抗体的显色原理是用辣根过氧化物酶标记的羊抗人IgA(第二抗体)与过氧化氢反应释放氧原子作用于3,3-二氨基联苯胺后,根据某些靶细胞上出现棕色沉淀反应,以反应细胞的密集程度、颜色深浅,来判断不同释放度的被检血清内抗EB病毒IgA抗体(第一抗体)的存在及其强度,作为辅助诊断鼻咽癌的依据。在这种显色中,联苯胺为高致癌性化学物质,进行显色操作时需要一定的防护措施。过氧化氢在常温环境下不稳定,不能随试剂提供,要求用户在临用时按要求加入并避光使用。由于储藏后的过氧化氢浓度趋于降低,技术不熟练者难于掌握它的用量,而导致阳性细胞染色过浅或使靶片染色过深。前者将造成假阳性结果,后者可干扰阳性细胞与阴性本底的对比。故以往的EB病毒检测试剂难于对阳性结果的程度作出客观判断。
本发明的目的在于提供一种对人体无害、判断准确的检测EB病毒抗体的显色试剂及其制造方法。
本发明由染色液和复染剂所组成,染色液包括3-氨基-9-乙基咔唑(AEC)、N,N-二甲基甲酰胺、过氧化脲和醋酸缓冲液,复染剂为0.5%固绿水溶液。
本发明的制造方法为:(1)将AEC以4mg/ml溶于N,N-二甲基甲酰胺溶液内即成AEC原液,将过氧化脲以0.3mg/ml浓度溶于0.1M    pH5.0的醋酸缓冲液中即成底物缓冲液;(2)将AEC原液和底物缓冲液以1∶20混合即成染色液;(3)用0.5g的固绿溶于100ml蒸馏水即成复染剂。
该显色试剂的使用方法:在靶细胞片上滴加待检血清,37℃作用45分钟后用pH7.0磷酸盐缓冲液(PBS)洗三次,再滴加辣根过氧化物酶标记的羊抗人IgA,37℃,45分钟后用PBS洗三次即加上述染色液,常温下作用7~10分钟后用水冲洗靶片再加复染剂1~2分钟作用后冲去复染剂,在显微镜下观察结果。
该显色试剂使靶片内的阳性细胞按被检血清内抗体的多寡分别被染成浅红色,红色或棕红色,阳性细胞百分率也相应提高。经固绿复染后,靶片内的阴性细胞被染成兰绿色,与阳性细胞成鲜明的对比,完全覆盖了非特异性染色,避免了假阳性,使判断更加客观、准确。由于过氧化脲水溶液在常温环境下十分稳定,便于提供给用户,简化了操作并使实验的重复可比性增加,特别适用于基层医疗单位使用。

Claims (2)

1、一种检测EB病毒抗体的显色试剂,其特征在于它由染色液和复染剂所组成,染色液包括3-氨基-9-乙基咔唑(AEC)、N,N-二甲基甲酰胺、过氧化脲和醋酸缓冲液,复染剂为0.5%固绿水溶液。
2、一种检测EB病毒抗体的显色试剂的制造方法,其特征在于:
A.将AEC以4mg/ml溶于N,N-二甲基甲酰胺溶液内即成AEC原液,将过氧化脲以0.3mg/ml浓度溶于0.1M  pH5.0的醋酸缓冲液中即成底物缓冲液;
B.将AEC原液和底物缓冲液以1∶20混合即成染色液;
C.用0.5g的固绿溶于100ml蒸馏水即成复染剂。
