Abstract

L'invention concerne des mutants de transposase (Tnp) Tn5 présentant une activité de transposase plus élevée que celle de la Tnp du type sauvage. Les mutants de TnpTn5 diffèrent de la Tnp du type sauvage aux positions d'acide aminé 54, 242 et 372 et présentent, par rapport à la Tnp du type sauvage, une avidité supérieure vis-à-vis d'une séquence d'extrémité extérieure de Tn5 du type sauvage telle que définie par SEQ ID NO:3 et/ou une séquence d'extrémité extérieure de Tn5 modifiée telle que définie par SEQ ID NO:5. L'invention concerne également différents systèmes et procédés pour l'utilisation des mutants de Tnp à des fins de transposition in vitro ou in vivo.
