Abstract

A method of detecting japonica cultivar rice seeds having no indica type genome domain which contaminate japonica cultivar rice seeds having the indica type genome domain is provided. This method comprises: i) constructing a pair of primers for amplifying the above-described japonica type genome domain or a part thereof based on the base sequence in the japonica type genome domain corresponding to the above-described indica type genome domain; ii) effecting a nucleic acid amplification reaction by using one or more japonica cultivar rice seed genomic DNAs as template(s); and iii) then judging that japonica cultivar rice seeds having no indica type genome domain contaminate, in case where nucleic acid amplification product(s) are detected. Moreover, an improved cetyltrimethylammonium bromide (CTAB) method is provided. This method comprises suspending ground brown rice grains in a buffer for CTAB extraction and then centrifuging at a low temperature to thereby remove starch.
