Abstract

Procédé permettant de réguler la synthèse de biopolymères par détermination de la génétique et de l'enzymologie de la biosynthèse de polyhydroxybutyrate (PHB) au stade moléculaire. Les enzymes et gènes purifiés constituent les moyens permettant de mettre au point de nouveaux biopolymères de type PHB ayant des squelettes de polyester. Les objectifs spécifiques sont 1) de commander la longueur de la chaine des polymères produits dans des procédés de fermentation par manipulation génétique, 2) d'introduire différents monomères dans les polymères afin de produire des copolymères ayant différentes propriétés physiques, et 3) d'examiner les propriétés physiques/rhéologiques de ces nouveaux biopolymères afin de mettre au point davantage de critères de conception au stade moléculaire. Le procédé permettant la mise au point de synthèse biopolymère consiste : à isoler et caractériser des gènes pour les enzymes dans la trajectoire synthétique (beta-cetothiolase, réductase d'acétoacétyle CoA et synthétase PHB), à cloner les gènes dans un vecteur(s), à placer le vecteur(s) sous commande d'activateurs régulés, à exprimer les gènes, à déterminer la fonction et l'utilisation d'autres facteurs tels que la spécificité du substrat dans la production et la composition de polymères, et à isoler ainsi qu'à procéder à l'analyse physique et chimique des polymères ainsi obtenus.
