Abstract

The present invention describes a connectable fixation system 1 intended as the space for performing chemical, biological or physical fixation, or a combination thereof, wherein said fixation system 1 comprises a main holder and container body 2 having a connecting bottom 20 which is connectable to a flask and arranged to contain fixation liquid, said fixation system 1 also comprising a lid unit 3 which is connectable to a connecting portion 61 of an inner open portion 6 of the main holder and container body 2 to seal the inner open portion 6, wherein the lid unit 3 is a lid and filling unit.

Technical Background Different types of fixation systems are known. For instance, inWO2010/120230 there is disclosed a system setup for carrying out abiomethane potential test on a biosample, wherein the system setupcomprises a digester with good agitation, a measuring device or sensor formeasuring gas composition on-line or spot check in regular basis, a containerfor chemical fixation of other gases than biomethane gas, the other gasesbeing at least carbon dioxide, wherein the chemical fixation is based onirreversible reaction(s); and a gas flow measuring device, wherein thedigester is connected to the container ford chemical irreversible fixation andthe container for chemical irreversible fixation is connected to the gas flowmeasuring device, and wherein the measuring device or sensor formeasuring gas composition may be provided after the digester and/or afterthe container for chemical irreversible fixation.
One aim of the present invention is to provide a fixation system whichhas several different possible usages, i.e. being a possible multi usagesystem, and which fixation system is simple to use for a user.

According to one specific embodiment of the present invention, thesystem is used for chemical fixation, e.g. in biological aerobic respiration testapplications or methane applications, such as for an aerobic biodegradability,biochemical oxygen demand and biomethane potential test, to remove targetgaseous compounds from a headspace of container.
The system or device according to the present invention may also beapplied to evaporate desired gaseous compounds to the headspace of aflask, such as add moisture by holding water in the system or device.
The present invention is directed to a system providing high flexibilityfor a user. This will become evident when reviewing the description below.Moreover, the system according to the present invention comprises at leasttwo units being the main holder and container body and the |id unit, whichunits are intendgd to be connected to each other. The main holder andcontainer body is intended to be connected to a flask to provide a complete system for fixation. 3ln relation to the present invention, the lid unit seals the main holder and container body and thus a flask which is connected to the main holderand container body. According to the present invention, the lid unit is a lid andfilling unit and is open in the top end, and also has a funnel shape, to enableto fill the main holder and container body with fixation liquid. This type of lidand filling unit may then be replaced with an agitator and lid unit which thenseals the entire system, i.e. the main holder and container unit containing thefixation liquid and also the connected flask in the bottom of the entire system,but of course also from the top, i.e. where the agitator and lid unit is arranged.This will be further understood when reviewing the description below and thefigures.
The main holder and container body may be used to store chemicalseither in liquid or solid form for adsorbing or absorbing target gas molecularfrom the gaseous headspace of a laboratory flask. As mentioned above, theadsorption or absorption can be based on chemical reaction, biological,physical or any combination with chemical, biological and physical interactionwith reagent and target gas molecule, etc.
Specific embodiments of the invention Below, some specific embodiments of the present invention aredisclosed and discussed further.
As hinted above, the system according to the present inventionprovides great flexibility and can be used in different ways. For instance, thesystem according to the present invention may suitably also have anintegrated agitator unit. ln line with this, according to one specific embodimentthe present invention is directed to a connectable fixation system intended asthe space for performing chemical, biological or physical fixation, or acombination thereof, wherein said fixation system comprises a main holderand container body having a connecting bottom which is connectable to aflask and arranged to contain fixation liquid, said fixation system alsocomprising an agitator and lid unit which is connectable to a top of the mainholder and container body to seal the inner open portion, wherein the agitatorand lid unit comprises an agitator rod which extends into the flask when theagitator and lid unit is connected to the container body which in turn is 4connected to the flask, to form a closed and sealed fixation flask system with agitation capability. Furthermore, the main holder and container body isarranged so that a suitable length of an agitation rod may be inserted andused for agitation of the liquid, slurry or solid media inside a laboratory flask(bioreactor).
As notable from above, according to this embodiment, the systemaccording to the present invention also provides agitation possibility inaddition to providing the actual fixation and a tight sealing.
Regardless if the present system comprises only a lid and filling unit oralso an integrated agitator and lid unit, these may be connected to a mainholder and container body which in turn may be connected to a standard flaskof any type, e.g. such a flask intended for laboratory analysis. Furthermore,this also implies that the present invention suitably comprises two different lidunits which are exchangeable with each other. The lid and filling unitaccording to the present invention is intended to enable to fill the main andholder body with fixation liquid, and the agitator and lid unit is intended toprovide agitation and full sealing of the entire system. Furthermore, the entiresystem may suitably also comprise a gas inlet and outlet arrangement as isfurther shown in fig. 3.
According to the present invention, the lid unit is a lid and filling unitand has a funnel shape with an open funnel end and a sealing end, whereinthe sealing end is connectable to the inner open portion of the main holderand container body, wherein the lid unit comprises a slot arranged above thesealing end, said slot enabling to fill the main holder and container body fromthe open funnel end of the lid unit. One such alternative is shown in fig. 1 and2 where the lid unit is a lid and filling unit and is in the form of an open funnelenabling to fill the main holder and container body with fixation liquid withoutcoming into contact with the content of a connected flask in the bottom. Theslot is in this case one possible solution to enable sealing of the flask and atthe same time filling of the main holder and container body.
Furthermore, according to the present invention, the sealing end of thelid unit and the connecting portion of the inner open portion of the main holderand container body are threaded to be connected. According to the present 5invention, the sealing end of the lid unit is insertable into the inner open portion of the main holder and container body. ln relation to above and also below, when the connection features ontwo units are threaded this implies that these two units may be screwedtogether.
According to the present invention, the lid unit is a lid and filling unitand comprises an extended portion arranged below the slot, said extendedportion being threaded to be locked into the inner open portion of the mainholder and container body when connected to the same. One such alternativeis shown in fig. 1 in a cross section.
Moreover, the main holder and container body suitably has somecertain features. According to one specific embodiment of the presentinvention, also supported by fig.1, the main holder and body has a bottomwhich is located beneath the top of a flask when these are connected to eachother, such as by being threaded. This enables to fill the main holder andcontainer body with fixation liquid when the lid and filling unit is connected, bitalso ensure that the fixation liquid is kept inside of the mina holder andcontainer body when the lid and filling unit is exchanged with an agitator andlid unit. ln line with this, according to one embodiment of the presentinvention, the main holder and container body is connected onto the flask asan outer container cylinder so that a container bottom of the main holder andcontainer body is positioned at a distance below a top of the flask. This typeof outer cylinder shape of the main holder and container body is shown incross section in fig. 1.
Furthermore, also the lid and filling unit may also have further featuresto enable filling of the main holder and container body in a suitable andeffective way. ln line with this, according to the present invention, the lid unitis a lid and filling unit and comprises a bottom sealing portion which isarranged to be continued into the slot enabling a sealing function towards aflask but being open via the slot towards a main holder and container bodywhen being connected to the same. Moreover, according to yet anotherspecific embodiment, the bottom sealing portion is inclined towards the slot.
One alternative thereof is shown in fig. 1. 6Furthermore, according to the present invention, the main holder and container body threaded to be connected to the top of a flask.Furthermore, according to one embodiment of the present invention, a bottomconnecting portion of the main holder and container body connectableon top of and outside of the top of a flask. This feature supports the feature ofthe main holder and container body being an outer cylinder onto the top of theflask so that a container bottom of the main holder and container body ispositioned at a distance below the top of the flask.
Furthermore, as hinted above, the connectable fixation systemaccording to the present invention suitably comprises both a lid and filling unitand an agitator and lid unit. ln line with this, according to one specificembodiment of the present invention, the fixation system also comprises anagitator and lid unit which is connectable to a connecting top of the mainholder and container body, wherein the agitator and lid unit comprises anagitator rod which extends into the flask when the agitator and lid unit isconnected to the container body which in turn is connected to the flask, toform a closed and sealed fixation flask system with agitation capability, andwherein the agitator and lid unit also comprises a sealing cap arranged toseal the connecting top of the main holder and container body, said sealingcap being arranged with a through hole into which the agitator rod may bepassed through, and an agitator rod tube intended to hold the agitator rodinside of the flask. One alternative showing this embodiment is shown in fig.3.
Furthermore, it should be noted that also in this case it suitably is athreaded connection between the main holder and container body and theagitator and lid unit.
Moreover, according to yet another embodiment of the presentinvention, the agitator and lid unit also comprises a motor and motor holderunit to which a connecting side or piece of the agitator rod is connectable, anda locking unit arranged to lock the sealing cap onto the connecting top of theof the main holder and container body, wherein the motor and motor holder unit is connectable to the locking unit. 7lt may further be said that the agitation system according to the present invention suitably contains a motor holder with a female screw thread so thatit may be screwed on top of the main holder and container body. The centraltunnel is designed with extra space, so that the agitator rod may loosely passthrough the central tunnel and so that gas may easily move and diffusebetween the headspace of a laboratory flask (bioreactor) and headspace ofthe main holder and container body. This may be important since the fixationtakes place on gas-liquid interface of the headspace compartment of the mainholder and container body, and therefore the gas must be easily diffused frombioreactor headspace to the main holder and container body headspace.
The present invention also refers to a flask system comprising aconnectable fixation system according to the present invention and a flaskconnectable to the main holder and container body of the connectable fixationsystem.
Moreover, the present invention also refers to a method comprisingusing a flask system according to above, for chemical fixation of at leastcarbon dioxide for performing a biological methane potential test or anaerobic respiration/biodegradability assay.
Detailed description of the drawinqs and further explanations ln fig. 1 there is shown a cross sectional view of a connectable fixationsystem 1 according to one embodiment of the present invention. Theconnectable fixation system 1 comprises a main holder and container body 2and a lid unit 3, in this case a lid and filling unit. The main holder andcontainer body 2 comprises a connecting bottom 20 intended to be connectedto the top of flask 80. ln this case the top of the flask is threaded to beconnected to the main holder and container body 2. The lid unit (3) isconnectable to a connecting portion 61 of an inner open portion 6 of the mainholder and container body 2 to seal the inner open portion 6.
Furthermore, in this specific case, the lid unit 3 has an open funnel end4 and a sealing end 5, wherein the sealing end 5 is connectable to the inneropen portion 6 of the main holder and container body 2. ln this case the lidunit 3 comprises a slot 7 arranged above the sealing end 5, said slot 7enabling to fill the main holder and container body 2 from the open funnel end 84 of the lid unit 3. Also in this case the sealing end 5 of the lid unit 3 and the connecting portion 61 of the inner open portion 6 of the main holder andcontainer body 2 are threaded to be connected to one another. As notable, inthis case it is the sealing end 5 of the lid unit 3 that is insertable into the inneropen portion 6 of the main holder and container body 2.
Moreover, the lid unit 3 is a lid and filling unit and comprises anextended portion 8 arranged below the slot 7, said extended portion (8) beingthreaded to be locked into the inner open portion 6, and thus connectingportion 61, of the main holder and container body 2 when connected to thesame.
Furthermore, the main holder and container body 2 is connected ontothe flask as an outer container cylinder so that a container bottom 50 of themain holder and container body 2 is positioned at a distance below a top ofthe flask. This enables to keep the content inside of the main holder andcontainer body 2 also after removal of the lid unit 3, when such should bereplaced with an agitator and lid unit 30 (see fig. 3).
Moreover, the lid unit also comprises a bottom sealing portion 9 whichis inclined and which is arranged to be continued into the slot 7 enabling asealing function towards a flask but being open via the slot 7 towards themain holder and container body 2 when being connected to the same.
Furthermore, in fig. 1 there is also shown a connecting bottom 20 ofthe main holder and container body 2, said connecting bottom beingconnectable and sealable to a flask. Moreover, the main holder and containerbody 2 also comprises a connecting top 21 which is connectable to anagitator and lid unit 30. ln fig. 2, there is shown one embodiment of a main holder andcontainer body 2 and a lid unit 3, the latter a so called lid and filling unit, whichtwo units are shown in separate mode.
Furthermore, in fig. 3 there is shown another specific embodimentaccording to the present invention. ln this case there is shown a flask system100 comprising a system 1 in which there is arranged an agitator and lid unit30 connected to a main holder and container body 2, which in turn isconnected to a flask to provide the flask system 100. As notable, the flask 9system 100 also comprises the flask 80. This type of lid unit is intended to be used to seal the entire system with a connected flask 80 and which flasksystem 100 also has both fixation capability and agitation capability. ln this specific case the agitator and lid unit 30 comprises an agitatorrod 301 which extends into the flask when the agitator and lid unit 30 isconnected to the main holder and container body 2 which in turn is connectedto the flask, to form a closed and sea|ed fixation flask system 1 with agitationcapability.
Moreover, the agitator and lid unit 30 is connectable to a connectingtop 21 of the main holder and container body 2. Furthermore, the agitator andlid unit 30 also comprises a sealing cap 302 arranged to seal a connecting top21 of the main holder and container body 2, said sealing cap 302 beingarranged with a through hole 303 into which the agitator rod 301 may bepassed through, and an agitator rod tube 304 intended to hold the agitator rod301 inside of the flask. Furthermore, the agitator and lid unit 30 alsocomprises a motor and motor holder unit 305 to which a connecting side orpiece 3010 of the agitator rod 301 is connectable. A locking unit 306 isarranged to lock the sealing cap 302 onto the connecting top 21 of the mainholder and container body 2, where the motor and motor holder unit 305 isconnectable to the locking unit 306.
Furthermore, the entire system 100 suitably also comprise a gas inletand outlet arrangement. ln the embodiment shown in fig. 3 this is arranged asgas/liquid inlet and outlet ports in the sealing cap 302.
As should be understood from the figures, the entire system, i.e. alsowith the connected flask may be seen as a flask system comprising aconnectable fixation system according to the present invention and a flaskconnectable to the main holder and container body 2 of the connectablefixation system 1. lt should be noted that the connectable fixation system 1 according tothe present invention comprises a main holder and container body 2 and a lidunit 3, suitably in the form of a lid and filling unit, and/or an agitator and lidunit 30. The different types of lid units are intended to be replaced dependingon what is needed in the fixation system, i.e. filling of fixation substances or agitation during trials. lt should be noted that fixation substances may be needed to be filled first. Then it is switched to the agitator and |id unit forproviding mixing for chemcial/biological reactions in the reactor/flask.Therefore, the selection of the |id could be due to the need of the sequence ofpreparation of analysis or reaction tasks.
When the agitator and |id unit 30 is combined with the main holder andcontainer body, which latter is also connected to a flask, then the entire flasksystem is sealed and gas tight. Again, the sealing cap 302 suitably comprisesgas/liquid inlet and outlet ports to create a completely gas tight system whichstill is connectable to e.g. a gas source.
Moreover, and as mentioned above, the different connections may alsobe threaded to ensure a tight sealing. This also implies that suitable materialwith good gas sealing should be used. For instance, the main holder andcontainer body 2 and the |id unit 3 may be made in a plastic material which isresistance to strong acids and alkali solutions, and to strong oxidant reagents,such as FeClzlFeCla. The material may also be selected to ensure certainchemical resistance and thermo-stability to stand for organic solvents, as wellas chemicals in solid form. This may open up for other potential applications.
Furthermore, the material chosen for both the main holder andcontainer body 2 and the |id unit 3 may be a plastic which is translucent or atleast semi-translucent. Then the level of chemicals inside of the main holderand container body 2 will be visible from the outside. Furthermore, certainchemical indicators, such as a pH indicator, may also be added to thechemical reagent for indicating the usage or capacity level of the chemicalfixation. ln relation to when the |id unit 3 is a |id and filling unit, then this isintended to facilitate in pouring liquid fixation chemicals into the compartment.lt may also be used for fine powder of a solid chemical. Moreover, itdiminishes the risk for chemical spilling during the setting up of the systemand when removing the device once the analysis is done. Therefore, the |idunit 3 in form of a |id and filling unit improves user friendliness of handling and safety. 11The system 1 according to the present invention may be used for different types of applications. lt can be used to fix target gas moleculesproduced from biological fermentation processes, and this may include but isnot limited to C02, H28, 02, etc., and by using the system together with asuitable gas flow one can selectively measure target gas production in term ofvolume and flow rate.
The system 1 can also be used as a container for water inside alaboratory flask, so that the moisture content can be well maintained if certainprocesses demands this. One example may be the analysis of biologicalrespiration of soil or samples in solid form. The moisture condition can also beimportant to ensure the biological reactions. ln relation to the above it should be mentioned that the presentinvention also covers methods involving using the system for theseapplications mentioned above.
Furthermore, more than one device can be connected together, so thatmultiple chemical reagents can be used for fixing different target gaseousmolecules from the same biological fermentation or any other chemcial andphyscial processes where certain gas may be released and consumed. Stillthese chemical reagents are well separated each other. This also applies to acombination of one system 1 with water and one system with a chemicalreagent according to the present invention. ln relation to the above it shouldbe said that the system 1 may be connected to other systems 1 according tothe present invention.
The system 1 and method according to the present invention haveseveral advantages. First of all, the device system 1 can be easily fixed ontop of a flask, e.g. a GL45 flask or any other size of standard flask, with orwithout connecting to an agitation system as explained above. This offers agreat flexibility for various potential applications including biologicalfermentations, chemical reactions, as well as biological samples andchemicals storage under certain conditions in terms of moisture content,headspace gas composition, etc. ln relation to this it should be understood that also a lid unit 3 may be a sealing lid which seals an entire flask system. 12Therefore, the lid unit 3 can be such sealing lid, a filling lid as explained above or an agitation and sealing lid 30 as explained above.
Secondly, the system 1 offers a great flexibility with possibility of usingone or multiple systems 1 for desired purpose.
Moreover, as the system 1 is fixed directly on top of a laboratory flask,this greatly improves the gas diffusion and makes the fixation process muchmore efficient with less dead volume between a reactor and fixation unit (mainholder and container body).
Furthermore, the system is designed for e.g. GL45 flasks with varioussizes. The most common sizes are from 50 ml up to 10 L.
Moreover, even though one suggested design according to the presentinvention is for GL45 screw threads, the present invention also is applicableto other sizes of GL thread laboratory flasks both in glass and plastic, as well as various sizes of flasks.

Claims (15)

Claims
1. Connectable fixation system (1) intended as the space for performingchemical, biological or physical fixation, or a combination thereof, whereinsaid fixation system (1) comprises a main holder and container body (2)having a connecting bottom (20) which is connectable to a flask and arrangedto contain fixation liquid, said fixation system (1) also comprising a lid unit (3)which is connectable to a connecting portion (61) of an inner open portion (6) of the main holder and container body (2) to seal the inner open portion (6).
2. Connectable fixation system (1) intended as the space for performingchemical, biological or physical fixation, or a combination thereof, whereinsaid fixation system (1) comprises a main holder and container body (2)having a connecting bottom (20) which is connectable to a flask and arrangedto contain fixation liquid, said fixation system (1) also comprising an agitatorand lid unit (30) which is connectable to a connecting top (21) of the mainholder and container body (2) to seal the inner open portion (6), wherein theagitator and lid unit (30) comprises an agitator rod (301) which extends intothe flask when the agitator and lid unit (30) is connected to the container body(2) which in turn is connected to the flask, to form a closed and sealed fixation flask system with agitation capability.

Connectable fixation system (1) according to claim 1 or 2, wherein the lidunit (3) is a lid and filling unit and has a funnel shape with an open funnel end(4) and a sealing end (5), wherein the sealing end (5) is connectable to theinner open portion (6) of the main holder and container body (2), wherein thelid unit (3) comprises a slot (7) arranged above the sealing end (5), said slot(7) enabling to fill the main holder and container body (2) from the open funnel end (4) of the cap unit (3).
4. Connectable fixation system (1) according to any of claims 1-3, wherein thesealing end (5) of the lid unit (3) and the connecting portion (61) of the inner 13open portion (6) of the main holder and container body (2) are threaded to be connected.
5. Connectable fixation system (1) according to any of c|aims 1-4, wherein thesealing end (5) of the cap unit (3) is insertable into the inner open portion (6)of the main holder and container body (2).

Connectable fixation system (1) according to any of c|aims 1-5, wherein the|id unit (3) is a |id and filling unit and comprises an extended portion (8)arranged below the slot (7), said extended portion (8) being threaded to belocked into the inner open portion (6) of the main holder and container body(2) when connected to the same.
7. Connectable fixation system (1) according to any of c|aims 1-6, wherein themain holder and container body (2) is connected onto the flask as an outercontainer cylinder so that a container bottom (50) of the main holder andcontainer body (2) is positioned at a distance below a top of the flask.

Connectable fixation system (1) according to any of c|aims 1-7, wherein the|id unit (3) is a |id and filling unit and comprises a bottom sealing portion (9)which is arranged to be continued into the slot (7) enabling a sealing functiontowards a flask but being open via the slot (7) towards a main holder andcontainer body (2) when being connected to the same.
9. Connectable fixation system (1) according to any of c|aims 1-8, wherein the bottom sealing portion (9) is inclined towards the slot (7).
10. Connectable fixation system (1) according to any of c|aims 1-9, whereinthe main holder and container body (2) are threaded to be connected to the top of a flask.
11. 1411. Connectable fixation system (1) according to any of claims 1-10, wherein a bottom connecting portion (10) of the main holder and container body (2)are connectable on top of and outside of the top of a flask.

Connectable fixation system (1) according to any of claims 1-11, saidfixation system (1) also comprising an agitator and |id unit (30) which isconnectable to a connecting top (21) of the main holder and container body(2), wherein the agitator and |id unit (30) comprises an agitator rod (301)which extends into the flask when the agitator and |id unit (30) is connected tothe container body (2) which in turn is connected to the flask, to form a closedand sealed fixation flask system with agitation capability, and wherein theagitator and |id unit (30) also comprises a sealing cap (302) arranged to sealthe connecting top (21) of the main holder and container body (2), saidsealing cap (302) being arranged with a through hole (303) into which theagitator rod (301) may be passed through, and an agitator rod tube (304)intended to hold the agitator rod (301) inside of the flask.

Connectable fixation system (1) according to claim 12, wherein theagitator and |id unit (30) also comprises a motor and motor holder unit (305)to which a connecting side or piece (3010) of the agitator rod (301) isconnectable, and a locking unit (306) arranged to lock the sealing cap (302)onto the connecting top (21) of the main holder and container body (2),wherein the motor and motor holder unit (305) is connectable to the lockingunit (306).
14. Flask system comprising a connectable fixation system (1) according toany of claims 1-13 and a flask connectable to the main holder and containerbody (2) of the connectable fixation system (1).
15. A method comprising using a flask system (100) according to claim 14, forchemical fixation of at least carbon dioxide for performing a biological methane potential test or an aerobic respiration/biodegradability assay.
