Przedmiotem wynalazku jest sposób przystosowania komórek macierzystych dla celów medycyny regeneracyjnej, poprzez genetyczną modyfikację macierzystych komórek mięśniowych oraz ich hodowlę. Wynalazek polega na wprowadzeniu do komórki macierzystej z mięśni poprzecznie prążkowanych genu GJA1, który zawiera sekwencję w co najmniej 95% zgodną z Sekwencją 1, przy czym korzystnie sekwencja użytego genu GJA1 powinna być zgodna w co najmniej 98% z Sekwencją 1.The subject of the invention is a method of adapting stem cells for the purposes of regenerative medicine, through genetic modification of muscle stem cells and their culture. Stem cells of muscle cells for the purposes of regenerative medicine are characterized by maintaining their pro-regenerative capacity while reducing possible side effects resulting from the need for direct interaction of two different types of muscle cells (myoblasts and cardiomyocytes) and their cooperation in the production of synchronous heart contraction. The invention consists in introducing into the stem cell from striated muscles the GJA1 gene which contains a sequence at least 95% compatible with Sequence 1, preferably the sequence of the GJA1 gene used should be at least 98% compatible with Sequence 1.
