Abstract

재조합으로 제조된 혈청 알부민은 옴이온-교환 흡착제와의 선택적인 배양, 소수성 고체상 및 수성상의 탈착제로서 수용성 지질양이온을 사용하는 친화 크로마토그래피의 여러단계로 정제된다. 상기 고정상은 2-메르캅토 또는 2-히드록시 알카노산에 결합된 운반체를 포함한다.

혈청 알부민의 정제방법

제1도 : 환원 조건하에서의 SDS-PAGE 9CBB R-250 염색). 분자량 마커 : 줄 1 및 5(24㎍). Q-세파로스 및 친화 크로마토그래피 후의 정제된 제조합 HSA : 줄 3(30㎍). 상업적으로 구입가능한 사람 혈액 기원 HSA(시그마 사람 알부민 NO. A-8763: 로트: 109F-9304) : 줄 4 (30㎍), 제6도 : 등전집적 (CBB R-205 염색). 마커 (줄 1 및 4), Q-세파로스 및 친화 크로마토그래피로 정제된 재조합 HSA(줄 2(20㎍)) 및 상업적으로 구입가능한 사람 혈액 기원 HSA(줄 3 (20㎍)).

Claims (13)

  1. 재조합 DNA 기술로 만들어진 형질전환된 미생물 또는 세포주를 이용한 발효로 제조한 정제된 혈청 알부민의 생산방법에 있어서, 상기 혈청 알부민을 함유하는 청정화되 발표 액체배지 또는 세포용해물로 출발하는 단계; 필요한 경우, 생성물배지의 pH를 대략적인 생리 pH로 변화시키는 단계; 배지를 음이온-교환배지와 배양하고 산성 pH의 완충액으로 세척하여 혈청 알부민을 용출시키는 단계; 필요한 경우, 생성물배지의 pH를 대략적인 생리 pH로 변화시키는 단계; 상기의 농축된 배지를 2-메르캅토 또는 2-히드록시 C4-C14 알카노산, 또는 그것의 염 또는 에스테르에 결합된 운반체를 함유하는 지방친화성 고정상과 함께 크로마토 그래피한 후, 탈착제인 수용성 지질 음이온을 수상에 첨가하여 알부민을 용출시키는 단계; 및 본질적으로 오염물질이 없는 정제된 혈청 알부민을 상기의 미생물 또는 세포주로 부터 분리하는 단계로 이루어지는 것을 특징으로 하는 방법.
  2. 제1항에 있어서, 상기 운반체는 에폭시드 화합물에 의해 활성화되는 것을 특징으로 하는 방법.
  3. 제2항에 있어서, 아가로스 운반체는 하기 구조의 에폭시드 화합물에 의해 활성화되는 것을 특징으로 하는 방법 :
  4. 상기에서 X는또는 할로이고; 및 만약 X가 할로이고, n이 1일때를 조건으로 하여 n 및 m은 각각, 개별적으로 0 내지 10이다.
  5. 제3항에 있어서, 에폭시 에테르는 디글리시딜에테르 (즉, n는 1이고, X는이다,) 또는 에피클로로히드린 (즉, X는 클로로이다) 인것을 특징으로 하는 방법.
  6. 상기 청구항중의 어느 한항에 있어서, 상기의 정제된 혈청 알부민을 투석시키는 부가적 단를 포함하는 것을 특징으로 하는 방법.
  7. 상기 청구항중 어느 한항에 있어서, 상기 혈청 알부민은 사람 혈청 알부민인 것을 특징으로 하는 방법.
  8. C4-C14 2-메르캅토 또는 2-히드록시 알카노산에 결합되어 있는 크로마토그래피 운반체
  9. 제7항에 따른 에폭시-활성화 운반체
  10. 99.9%이상 순수한, 특이하게는 99.95% 이상 순수한 분리된 사람 혈청 알부민 조성물.
  11. 제9항에 있어서, 의약용도의 분리된 혈청 알부민 조성물.
