실시예 1~3: 오크라, 몰로키아, 귀리, 밀 및 캐롭콩 혼합추출물의 제조Examples 1-3: Preparation of mixed extracts of okra, molokia, oats, wheat and carob beans
상기 비교예 2와 비교예 5에서 제조한 추출물을 하기 표 2의 중량비로 혼합하여 오크라, 몰로키아, 귀리, 밀 및 캐롭콩 혼합추출물을 제조하였다.The extracts prepared in Comparative Examples 2 and 5 were mixed in the weight ratio shown in Table 2 below to prepare a mixed extract of okra, molokhia, oats, wheat, and carob beans.
시험예 1: 보습 인자 HAS-3 발현 효과 Test Example 1: Effect of moisturizing factor HAS-3 expression
LSGS가 첨가된 Midium 106 배지를 이용하여 섬유아세포(CCD-986Sk)를 5×105 cell/well로 조절하여 12 well plate에 접종하였다. 접종된 배지를 18시간 동안 배양한 후, 본 발명 실시예 및 비교예의 시료에 같은 농도로 처리하고 24시간 추가 배양하였다. 배양된 세포에서 RNA를 추출하여 RT-PCR(Reverse transcription polymerase chain reaction)을 통해 HAS-3(Hyaluronan Synthase-3)의 발현 증가율을 확인하였다. HAS-3의 발현 증가율을 하기 식에 의하여 산출하였으며, 그 결과를 하기 표 3에 나타내었다. Using Medium 106 medium supplemented with LSGS, fibroblasts (CCD-986Sk) were adjusted to 5× 105 cells/well and seeded in a 12-well plate. 상기 표 3에서 확인되는 바와 같이, 제1 추출물과 제2 추출물의 혼합물인 본 발명의 실시예의 시료에서 각각의 추출물인 비교예의 시료에 비하여 높은 HAS-3 발현율을 나타내었다. 그 중 실시예 3에서 가장 우수한 보습효과를 나타내었다. 상기 비교예와 실시예에서 제조된 오크라, 몰로키아, 귀리, 밀 및 캐롭콩 혼합추출물을 시료로 사용하여 Hyaluronic acid 생성량을 측정하였다. 상기 시험방법은 HaCaT 세포를 이용하여 생성된 hyaluronic acid를 측정하여 보습능력을 평가하는 것으로, 먼저 HaCaT 세포를 6 well plate에 1×106cells/well의 농도로 분주하여 배양하였다. 배양 후 FBS가 첨가되지 않은 배지로 교체하여 18시간 starvation 시키고, 24시간 동안 추가 배양하였다. HaCaT 세포에 각 시료를 같은 농도로 처리하고, 24시간 동안 배양 한 후 회수한 배양액은 ELISA kit를 이용하여 hyaluronic acid의 생성량을 측정하였다.The amount of hyaluronic acid produced was measured using the mixed extracts of okra, molokia, oat, wheat and carob beans manufactured in the comparative examples and examples above as samples. 시험예 3: 피부 장벽 강화 인자 Filaggrin 발현 효과 Test Example 3: Effect of expression of skin barrier strengthening factor Filaggrin
각질형성세포(HaCaT)를 10% FBS가 첨가된 DMEM 배지를 이용하여 5×105 cell/well로 조절한 후, 12 well plate에 접종하고 18시간 동안 배양한 후 200mJ UVB와 시료를 처리하고 24시간 동안 배양하였다. 배양된 세포에서 RNA를 추출하여 RT-PCR(Reverse transcription polymerase chain reaction)을 통해 필라그린(Filaggrin, FLG)의 발현 증가율을 확인하였다. 발현 증가율은 하기 식에 의해 산출되었으며 음성대조군은 시료 대신 배지를 이용하였으며 양성 대조군으로는 레티노산(Retinoic acid)을 사용하였다. 그 결과를 하기 표 5에 나타내었다. Keratinocytes (HaCaT) were adjusted to 5 × 10 5 cells/well using DMEM medium supplemented with 10% FBS, seeded in 12-well plates, and cultured for 18 hours, then treated with 200 mJ UVB and samples, and cultured for 24 hours. 시험예 4: 피부 보습 효과 평가 Test Example 4: Evaluation of skin moisturizing effect
피부의 피부전도도를 측정해주는 피부 수분 측정기를 이용하여 피부 보습에 대한 효과를 평가하였다. 피부 수분 측정은 실내온도 20~25℃, 상대습도 40~55%의 항온항습 조건 하에서 실시하였으며, 실시예 4와 대조예 1의 크림 제형을 사용하였다. 10명의 피검자 얼굴에 실시예 4와 대조예 1의 크림 제형을 각각 2회/1일 도포하여 사용 전 및 2주 후의 수분 함유량을 비교하였다. 그 결과를 표 7에 나타내었다. 피부의 거칠기 정도를 측정해주는 Dermatopblue(Breuck-Mann GmbH, Meersburg, Germany)를 이용하여 피부 결 개선 효과를 확인하였다. 10명의 피검자 얼굴을 절반으로 나누어 각각 실시예 4와 대조예 1의 크림 제형을 사용하였다. 10명의 피검자 얼굴에 실시예 4와 대조예 1의 크림 제형을 각각 2회/1일 도포하여 사용 전 및 6주 후의 피부 거칠기 개선 정도를 비교하였다. 그 결과를 표 8에 나타내었다. 피부 장벽 기능 강화 효과를 확인하기 위하여, 10명의 피검자 얼굴을 절반으로 나누어 각각 크림제형으로 제조한 대조예 1과 실시예 4를 하루 2회씩 4주간 도포하였다. 4주 후 피부 장벽 기능의 개선 정도를 경표피의 수분 손실량을 측정하는 Delfin Vapo Meter SWL4255를 통해 평가하여 결과를 표 9에 나타내었다. Claims (10)
(B) b1)상기 (A)단계에서 추출물을 제조하고 남은 오크라, 몰로키아, 귀리, 밀 및 캐롭콩 잔사를 회수하는 단계; b2) 회수된 잔사를 용매에 침지 후 유산균을 접종하여 25~30℃의 조건에서 22~26시간 동안 발효하는 단계; b3) 80~100℃에서 2~3시간 동안 추출하는 단계; b4)여과하여 제2추출물을 제조하는 단계; 및
(C) 상기 수득한 (A) 단계의 제1추출물과 (B) 단계의 제2추출물을 각각 1~2:1~2의 중량비율로 혼합하는 단계를 포함하는 오크라, 몰로키아, 귀리, 밀 및 캐롭콩 혼합추출물의 제조방법.(A) a1) a step of immersing a mixture of okra, molokhia, oats, wheat and carob beans in at least one solvent selected from the group consisting of water, lower alcohols having 1 to 4 carbon atoms, ethyl acetate, glycerin, ethylene glycol, propylene glycol and butylene glycol and extracting at a low temperature of 30 to 40°C for 2 to 3 hours; a2) a step of filtering to prepare a first extract;
(B) b1) a step of recovering the remaining okra, molokia, oats, wheat and carob bean residues after preparing the extract in the above step (A); b2) a step of immersing the recovered residues in a solvent, inoculating them with lactic acid bacteria and fermenting them at 25-30°C for 22-26 hours; b3) a step of extracting at 80-100°C for 2-3 hours; b4) a step of filtering to prepare a second extract; and
(C) A method for producing a mixed extract of okra, molokia, oats, wheat and carob beans, comprising a step of mixing the first extract of step (A) and the second extract of step (B) obtained above in a weight ratio of 1 to 2:1 to 2, respectively.
A method for producing a mixed extract of okra, molokia, oats, wheat, and carob, characterized in that in claim 9, the lactic acid bacteria is at least one selected from the group consisting of Lactobacillus plantarum, Lactobacillus lactis, Lactobacillus bulgaricus, Lactobacillus rhamnosus, Pediococcus cerevisiae, Lactobacillus breve, Lactobacillus sakei, and Lactobacillus crispatus.
