Abstract
본 명세서에는 프라그 기질 용해제가 개시된다. 상기 구강 조성물은 폴리에틸렌글리콜을 포함하는 구강 내 프라그 기질을 용해하기 위한 용제와 세균을 제거하기 위한 항균제를 포함함으로써, 끈적끈적한 밀집체를 형성하여 세균막을 치밀하게 하고 세균이 치면에서 잘 떨어져 나가지 않도록 세균의 재부착을 돕는 프라그 기질을 용해시키고 항균 성분이 프라그 내부로 침투하여 프라그를 효과적으로 제거할 수 있는 효과가 있다.
Description
본 명세서에는 프라그 기질 용해제가 개시된다.
프라그는 치아 표면에 부착되어 있는 미생물과 그 생성물로 구성된 유기성 치아 부착물로서, 구강 내 자정작용에 의해서는 제거될 수 없으며 칫솔질과 같은 물리적인 방법에 의해서만 제거된다. 따라서 칫솔질이 닿지 않는 프라그는 제거하기가 어려우며 이는 치아우식증 및 잇몸 질환의 원인이 된다.
일반적으로 사용하는 구강용 항균제는 미생물 자체에는 우수한 항균력을 보이나 프라그와 같이 미생물과 기질로 이루어진 치면세균막에서는 항균 성분이 프라그 내부로 침투가 어려워 그 효과가 미비하다. 특히 기질(plaque matrix)의 주요 성분인 세균 유래의 다당체인 난용성 글루칸 성분은 끈적끈적한 밀집체를 형성하여 세균막을 치밀하게 하고 세균이 치면에서 잘 떨어져 나가지 않도록 하여 세균의 재부착을 돕는다. 항균제는 이러한 프라그 기질을 효과적으로 제거하기 어려운 문제가 있다.

또한, 치면 세균막을 제거하지 아니한 채 장기간 있게 되면, 구강 내 타액 중의 칼슘, 마그네슘 및 인 등의 성분과 결합하여 난용성 염을 형성하여 치아와 유사한 경도를 가진 치석이 형성된다. 이렇게 형성된 치석은 계속 진행되면서 잇몸과 치아의 경계면을 파고들어 양치시 쉽게 출혈을 유발시키고, 구강 내 세균에 의한 염증을 유발시켜 치은염 및 치주질환을 발생시킨다.
본 명세서에서 "프라그 기질"(plaque matrix)은 프라그를 치아 표면 또는 잇몸에 부착, 정착시키는 역할을 수행하는 점착성 물질을 의미한다. 프라그는 세균과, 이러한 세균 또는 타액(saliva)에서 유래된 점착성 물질로 이루어지는데, 프라그 내 세균은 세균 또는 타액에서 유래된 점착성 물질의 그물망 내 존재하게 된다. 즉, 프라그 기질은 세균 또는 타액에서 유래된 글루칸 등을 포함하는 점착성 물질을 말한다.

예시적인 일 구현예에서, 상기 구강 세균은 스트렙토코커스 뮤탄스(Streptococcus mutans), 스트렙토코커스 상귀니스(Streptococcus sanguinis), 스트텝토코커스 소브리너스(Streptococcus sobrinus), 스트렙토코커스 라티(Streptococcus ratti), 스트렙토코커스 크리세티(Streptococcus criceti), 스트렙토코커스 안지노수스(Streptococcus anginosus), 스트렙토코커스 고도니(Streptococcus gordonii), 액티노바실러스 액티노마이셈템코미탄스(Actinobacillus actinomycetemcomitans), 푸소박테리움 뉴클레아툼(Fusobacterium nucleatum), 프리보텔라 인터미디어(Prevotella intermedia) 및 포필로모나스 진지발리스(Porphylomonas gingivalis)로 이루어진 군에서 선택되는 1 이상일 수 있다. 다른 측면에서, 본 명세서에 따른 프라그 기질 용해제를 포함하는 구강 조성물은 상기 구강 세균에 대해 항균 활성을 갖는 효과가 있다.

예시적인 일 구현예에서, 상기 구강 조성물은 치약, 마우스워시, 크림 또는 패치의 제형일 수 있다.
본 명세서에서 치약은 액체 또는 고체 치약을 포함하는 것으로서 그 제형에 제한이 없고, 마우스워시는 구강 청정제라고도 한다. 또한, 크림은 구강 연고를 포함하는 것이고, 패치는 치아 표면 상에 붙여서 사용하는 것을 의미한다.
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 예시하기 위한 것으로서, 본 발명의 범위가 이들 실시예에 의해 제한되는 것으로 해석되지 않는 것은 당업계에서 통상의 지식을 가진 자에게 있어서 자명할 것이다.
실시예 1~2. 마우스워시 제조
하기 조성에 따라 실시예 1 및 2의 마우스워시 구강 조성물을 제조하였다.
수용성분인 PEG, 염화세틸피리디늄, 벤조산나트륨, 사카린나트륨을 정제수에 투명해질 때까지 녹이고 향료와 폴리옥시에틸렌경화피마자유를 혼합한 용액을 수용성분을 녹인 용액에 추가한 후 혼합하여 마우스워시 구강 조성물을 제조하였다.

비교예 1~2. 마우스워시 제조
상기 실시예와 동일한 방법으로 마우스워시를 제조하되, 하기 조성에 따라 비교예 1 및 2의 마우스워시 구강 조성물을 제조하였다.

실험예 1. 프라그 제거능 확인
하기 방법에 따라 상기 실시예 및 비교예에서 제조한 마우스워시의 프라그 제거능을 확인하였다.
1) 스트렙토코커스(Streptococcus mutans, KCTC 3065)균을 Brain Heart infusion 배지에서 24시간 동안 35℃에서 배양하였다.
2) 치아 모사물질인 하이드록시아파타이트 플레이트를 70% 에탄올에 30분간 담구고 멸균한 후 멸균 이온수로 3회 세정하였다. 세정한 하이드록시아파타이트 플레이트를 한 개씩 24-웰 플레이트에 넝었다.
3) 각 웰에 1% 뮤신(mucin)을 함유한 멸균 이온수를 1ml씩 넣고 35℃에서 30분간 보관하여 하이드록시아파타이트 플레이트를 뮤신으로 코팅하였다.
4) 배양시킨 스트렙토코커스 뮤탄스의 배양액을 접종하여 35℃, 72시간 배양하여 바이오필름을 형성시켰다.
5) 실시예 및 비교예의 마우스워시를 각각 PBS버퍼에 20% 희석한 후 1ml를 취하여 상기 4단계에 따라 얻어진 하이드록시아파타이트 플레이트에 3분간 처리한 후 멸균 이온수로 3회 세정하였다.
6) 0.1% 크리스탈 바이올렛 용액을 처리하여 염색하고 완전히 건조시킨 후, 70% 에탄올에 넣어 염료를 추출하였다.
7) 추출된 염료 희석액의 농도를 광학밀도 OD(Optical Density, 590nm)로 측정하였다.
그 결과, 도 1에서 보는 바와 같이 비교예에 따른 마우스워시에 비해 실시예에 따른 마우스워시의 OD 값이 현저하게 낮아 바이오필름이 제거된 것을 알 수 있었다. 이에 따라, 본 명세서에 개시된 폴리에틸렌글리콜을 포함하는 구강 조성물은 프라그 기질을 용해하여 효과적으로 바이오필름을 제거하는 효과가 있음을 확인하였다.
이상, 본 발명내용의 특정한 부분을 상세히 기술하였는바, 당업계의 통상의 지식을 가진 자에게 있어서, 이러한 구체적인 기술은 단지 바람직한 실시태양일 뿐이며, 이에 의해 본 발명의 범위가 제한되는 것이 아닌 점은 명백할 것이다. 따라서 본 발명의 실질적인 범위는 첨부된 청구항들과 그것들의 등가물에 의해 정의된다고 할 것이다.
Claims (7)
- 유효성분으로서 폴리에틸렌글리콜을 포함하고,
상기 폴리에틸렌글리콜은 PEG-8 및 PEG-75 중 1 이상인, 프라그 기질(plaque matrix) 용해제.
- 제 1항에 있어서,
상기 프라그 기질은 구강 세균 유래 글루칸을 포함하는, 프라그 기질 용해제.
- 제 1항에 있어서,
상기 폴리에틸렌글리콜은 PEG-75인, 프라그 기질 용해제.
- 제 1항 내지 제 3항 중 어느 한 항의 프라그 기질 용해제; 및
항균제를 포함하는 프라그 제거용 구강 조성물.
- 제 4항에 있어서,
상기 폴리에틸렌글리콜은 조성물 전체 중량을 기준으로 0.1 내지 10 중량%로 포함되는, 프라그 제거용 구강 조성물.
- 제 4항에 있어서,
상기 항균제는 조성물 전체 중량을 기준으로 0.001 내지 3 중량%로 포함되는, 프라그 제거용 구강 조성물.
- 제 4항에 있어서,
상기 항균제는 염화세틸피리디늄, 클로르헥시딘 디글루코네이트, 염산클로르헥시딘, 염화벤젠토늄, 염화벤즈알코늄, 염화아연, 구연산아연, 유산아연, 이소프로필 메틸페놀, 살리실산메틸, 치몰, 유칼립톨, 레보멘톨로 구성된 군에서 선택되는 1 이상인, 프라그 제거용 구강 조성물.
