Abstract
Description
도2는 실시예에서 검출 대상체 (Zika virus)를 위한 리보조절인자 재조합 벡터구조를 나타낸 플라스미드 지도로, A는 27B 센서 벡터이고(ZIKV_Sensor_27B_LacZ, addgene number: Plasmid #75006) B는 32B 센서 벡터 (ZIKV_Sensor_32B_LacZ, addgene number: Plasmid #75007)를 나타낸 것이다.
도3은 본 발명의 실시예를 통한 리포터 유전자가 들어간 리보조절인자 발현을 확인한 결과로, A는 리포터 유전자 발현을 Microplate reader기의 흡광도를 통하여 정량적으로 확인한 결과이고, B는 리포터 유전자 발현을 종이결과로 육안으로 확인한 결과이다.
도4는 본 발명의 실시예에 제조한 리보조절인자를 이용한 바이러스 검출결과로, Microplate reader기의 흡광도를 통하여 정량적으로 확인한 결과이다.
도5는 본 발명의 실시예를 통한 바이러스 진단용 FET 통합 센서의 고정 항체 (Antibody)로 확인하기 위한, 리포터 유전자 항체 활성 확인용 웨스턴 블롯 (Western blot) 실험결과이다.
도6은 본 발명의 실시예를 통한 리보조절인자를 이용한 바이러스 진단용 FET 통합 센서를 이용하여 바이러스를 측정한 리포터 유전자 항체 실시간 검출 곡선이다.
도7은 본 발명의 실시예를 통한 리보조절인자 생성물 샘플링 시간에 대한 리보조절인자를 이용한 바이러스 진단용 FET 통합 센서 기반 실시간 검출 곡선 결과로, A는 리보조절인자 생성물의 샘플 시간을 화살표로 나타낸 것이고, B는 리보조절인자 생성물 샘플링 시간에 따른 바이러스 진단용 FET 통합 센서의 바이러스 검출 곡선 결과이다.

- 기판; 상기 기판에 배치되는 그래핀 채널층; 상기 그래핀 채널층의 양단에 이격되어 배치되어 소스 전극과 드레인 전극 한 쌍의 금속; 및 상기 그래핀 채널층 상에 배치되고 N-헤테로사이클릭 카벤 화합물로 이루어진 링커층; 및 상기 링커층 상에 27B 센서로 구성된 리보조절인자(Switch RNA) 및 상기 리보조절인자의 리포터 유전자(reporter gene)의 발현을 감지하는 베타-갈락토시다아제 항체가 결합된 바이오 탐침부를 포함하는 것을 특징으로 하는 지카 바이러스 진단용 FET 통합 센서를 사용하고,
1) 지카 바이러스를 검출하기 위하여 서열번호 1인 27B 센서로 구성된 리보조절인자와 리포터 유전자인 베타-갈락토시다아제 (Beta-galactosidase, LacZ)를 넣어 플라스미드 제작하는 단계;
2) 바이러스 유무 및 정량적 확인을 위해 리포터 유전자를 사용하는 단계;
3) 세포가 아닌 마이크로튜브에 세포-유리 전사 및 번역 시약과 제작한 플라스미드를 혼합하는 단계;
4) 객담, 폐포 세척액, 분변, 소변 및 혈액으로 이루어진 군에서 어느 하나 이상을 포함하는 검체로부터 RNA를 분리하는 단계; 및
5) 만들어진 혼합물을 종이 위에 떨어트려 동결건조 시킨 후 분리된 RNA를 넣어주면 리보조절인자와 반응해 리포터 유전자가 발현되어 바이러스 유무를 확인하는 단계;를 포함하는 지카 바이러스 존재 유무에 대한 정보를 제공하는 방법. - 삭제
