Abstract
Description
도 2는 매실 추출물(JAE)에서 FA-1 [5-하이드록시메틸-2-푸르알데히드 비스(5-포르밀푸르푸릴)아세탈]의 정제하는 과정을 나타낸 도면이다.
도 3은 FA-1 [5-하이드록시메틸-2-푸르알데히드 비스(5-포르밀푸르푸릴) 아세탈]의 구조 결정을 나타낸 도면이다. (A) FA-1의 ESI-TOFMS 데이터, (B) 1H NMR 데이터(화학적 이동, ppm; JH,H 값, Hz),(C) 13C NMR 데이터(화학적 이동, ppm) 및 (D) HMBC 상관관계이다.
도 4는 FA-1이 용량 의존적 방식으로 ALDH 및 WRN 단백질을 증가시키고 불멸화 인간 기관지 상피세포에서 담배연기-유발 세포독성(MTT 및 유세포 분석기)으로부터 보호함을 나타내는 도면이다. (A) ALDH2, ALDH3A1 및 WRN의 발현 분석한 결과로 FA-1 [DMSO(디메틸설폭사이드) 중 5, 25, 50, 75 및 150 nM] 또는 비히클(1.0% 부피의 DMSO에 해당)로 처리한 후 HBEC2 세포를 배양하였다. (B) MTT 분석결과로 HBEC2 세포를 24 시간 동안 0 및 6% CSE의 존재 또는 부재 하에 NAC(2 mM), FA-1(150 nM) 및 비히클과 함께 배양하고 생존력에 분석하였다. 데이터는 배양된 불멸화 인간 기관지 상피세포에서 평균 ±SEM(**p<0.01) 세포독성으로 표현된다. (C) 유세포 분석결과로 아넥신 V 및 프로피디움 요오드화물 (PI) 염색하고 을 세포사멸을 분석하였다.
도 5는 FA-1이 HBEC2 세포에서 담배연기-유발 산화적 손상과 DNA 손상을 약화시킴을 나타내는 도면이다. HBEC2 세포를 24 시간 동안 2% CSE의 존재 또는 부재 하에 FA-1(150 nM)과 함께 배양하고, 4-HNE 및 ATM의 인산화에 대해 면역 블롯 분석을 수행하였다.
도 6은 FA-1이 용량 의존적 방식으로 ALDH2 및 WRN 단백질을 증가시키고, 정상적인 인간 표피 각질세포에서 WRN 단백질의 담배연기-유도 하향 조절을 약화시킴을 나타낸 도면이다.
(A) ALDH2 및 WRN 면역블랏 결과이다. 다양한 농도의 FA-1 [DMSO(디메틸설폭사이드) 중 5, 25, 50, 75 및 150 nM) 또는 비히클(1.0% 부피의 DMSO에 해당)로 처리한 후 NHEK 세포를 배양하였다.
(B) 담배연기-유도 하향 조절을 약화를 면역블랏으로 분석한 결과이다. NHEK 세포를 24 시간 동안 2% CSE의 존재 또는 부재 하에 FA-1(150 nM) 및 비히클과 함께 배양하였다.
도 7은 FA-1이 정상적인 인간 표피 각질 세포에서 담배연기-유발 세포독성과 DNA 손상을 방지함을 나타내는 도면이다. (A) 0, 2, 4 및 6% CSE의 존재 또는 부재 하에 24 시간 동안 FA-1 [DMSO(디메틸설폭사이드) 중 150 nM)로 처리한 후 NHEK MTT(3-(4,5-디메틸티아졸-2-일)-2,5-디페닐 테트라졸륨 브로마이드)-계 세포 생존력을 나타낸 도면이다. 데이터는 배양된 NHEK에서 평균 ±SEM(**p<0.01) 세포독성으로 표현된다. (B) NHEK 세포를 2% CSE의 존재 또는 부재 하에 24 시간 동안 150 nM의 FA-1로 처리하였다. ATM 및 H2AX의 인산화를 위해 면역블롯 분석을 수행하였다.
도 8은 클로로포름 분획물을 재분획한 F1 내지 F4분획물의 담배연기 추출물 유도 세포독성에 대한 세포 생존율 분석결과이다.
도 9 내지 14는 분리된 활성 화합물 FA-1의 구조분석 결과이다. 구체적으로도 9는 1H NMR 스펙트럼이고, 도 10은 13C NMR 스펙트럼이고, 도 11은 COSY 스펙트럼, 도 12는 TOCSY 스펙트럼, 도 13는 HMBX 스펙트럼, 도 14는 HSQC 스펙트럼 결과이다.
Claims (5)
- 5-하이드록시메틸-2-푸르알데히드 비스(5-포르밀푸르푸릴)아세탈을 포함하는, 만성폐쇄성 폐질환 치료용 약학 조성물.
- 제1항에 있어서, 상기 5-하이드록시메틸-2-푸르알데히드 비스(5-포르밀푸르푸릴)아세탈은 매실로부터 분리된 것인 약학 조성물.
- 제1항에 있어서, 상기 5-하이드록시메틸-2-푸르알데히드 비스(5-포르밀푸르푸릴)아세탈은 ALDH2, ALDH3A1 또는 WRN의 발현을 증가시키는 것을 특징으로 하는 약학 조성물.
- 5-하이드록시메틸-2-푸르알데히드 비스(5-포르밀푸르푸릴)아세탈을 유효성분으로 포함하는, 만성폐쇄성 폐질환의 예방 또는 개선용 식품 조성물.
- 제 4항에 있어서, 상기 식품 조성물은 건강기능성 식품인, 만성폐쇄성 폐질환의 예방 또는 개선용 식품 조성물.
