Abstract
Description
도 2은 본 발명의 실시예에 따라 후성 유전자 조절제가 처리된 중간엽 줄기세포와 염증성 Th17 분화조건을 처리한 건강한 성인으로부터 분리한 말초혈액 단핵세포를 공조배양한 후에 Th17 세포의 증식은 T 세포 증식 측정 방법 (mixed lymphocytes reaction)으로 확인 및 사이토카인의 함량 (B, C) 및, 면역조절 T세포인 CD25+Foxp3+의 함량 (D)을 유세포분석기로 측정한 것을 나타낸다
도 3은 본 발명의 실시예에 따라 후성 유전자 조절제가 처리된 중간엽 줄기세포와 염증성 Th17 분화조건을 처리한 건강한 성인으로부터 분리한 말초혈액 단핵세포를 공조배양한 후에 염증성 인자 IL-17, IFN-g, 및 IL-2와 항염증성 인자 IL-10 사이토카인 변화를 ELISA 방법으로 측정한 것을 나타낸다.
도 4은 본 발명의 실시예에 따라 후성 유전자 조절제가 처리된 중간엽 줄기세포와 염증성 Th0 분화조건을 처리한 류마티스 관절염 환자로부터 얻은 관절 활막내 단핵세포를 공조배양한 후에 사이토카인의 함량 (A, B) 및, 면역조절 T세포인 CD25+Foxp3+의 함량 (C)을 유세포분석기로 측정한 것을 나타낸다.

Claims (16)
- 면역조절능이 증가된 중간엽 줄기세포를 유효성분으로 포함하는, IL-17, IFN-g, IL-2, IL-10, 및 IDO으로 이루어진 군에서 선택된 1종 이상의 사이토카인 매개성 염증 질환의 예방 또는 치료용 약학 조성물로서,
상기 면역조절능이 증가된 중간엽 줄기세포는, 중간엽 줄기세포 또는 그 배양물에 메틸화 저해제 (Methylation inhibitor) 및 히스톤 탈아세틸화 저해제 (Histone deacetylase inhibitor)를 1:1 내지 1:6000 몰 비율로 처리하여 얻어진 것이며, 상기 메틸화 저해제 및 히스톤 탈아세틸화 저해제는 (a)Decitabine 및 TSA, 또는 (b)5-azacytidine 및 VPA이고,
상기 사이토카인 매개성 염증 질환은 전신 홍반성 루프스, 하시모토 갑상선염, 제1형 당뇨, 갑상선기능저하증, 갑상선기능항진증, 쇼그렌 증후군(Sjogren's syndrome), 건선, 크론병, 갑상선염, 및 궤양성 대장염으로 이루어진 군에서 선택된 1이상을 포함하는 것인, 약학 조성물. - 삭제

- 제1항에 있어서, 상기 중간엽 줄기 세포는 제대 유래 중간엽 줄기 세포, 제대혈 유래 중간엽 줄기 세포, 골수 유래 중간엽 줄기 세포, 지방 유래 중간엽 줄기 세포, 근육 유래 중간엽 줄기 세포, 신경 유래 중간엽 줄기 세포, 피부 유래 중간엽 줄기 세포, 양막 유래 중간엽 줄기 세포, 및 태반 유래 중간엽 줄기 세포로 이루어진 군에서 선택된 1종 이상인 것인, 약학 조성물.
- 제1항에 있어서, 상기 유효성분은 IL-10, 및 IDO으로 이루어진 군에서 선택된 1종 이상의 항염증 인자의 발현을 증가시키는 것인, 약학 조성물.
