Abstract
Description
도 2는 대장균 변이체 (variants)에서 나타나는 호모세린 O-숙시닐 트랜스퍼라제 아미노산 1차 서열의 상동성을 비교한 도면이다.
도 3 및 도 4는 메치오닌에 대한 피드백 조절이 해제된 변이 호모세린 O-숙시닐 트랜스퍼라제 아미노산 1차 서열의 상동성을 비교한 도면이다. 비교 대상은 야생형 호모세린 O-숙시닐 트랜스퍼라제, PCT 공개특허 (WO 2008/127240)에 개시된 피드백 조절 해제된 변이 호모세린 O-숙시닐 트랜스퍼라제 met10A, met11A, 및 PCT 공개특허 (WO 2005/108561)에 개시된 피드백 조절 해제된 변이 호모세린 O-숙시닐 트랜스퍼라제이다.
도 5는 염색체 내 acs 프로모터를 pro 프로모터로 교체하기 위하여 overlapping PCR 방법을 사용한 FRT-one step deletion 카세트 제작 도이다.
Claims (18)
- 서열번호 17의 아미노산 서열로 이루어진 폴리펩티드 또는 상기 폴리펩티드 서열과 95% 이상의 상동성을 가지는 대장균 유래의 폴리펩티드의 시작 메치오닌으로부터 111번째 아미노산인 글리신이 글루탐산으로 치환된 호모세린 O-아세틸 트랜스퍼라제 활성을 가지는 변이형 폴리펩티드.
- 제1항에 있어서, 상기 폴리펩티드는 추가로 112번째 아미노산이 쓰레오닌 또는 히스티딘으로 치환된 것인 변이형 폴리펩티드.
- 제1항 또는 제2항에 있어서, 상기 변이형 폴리펩티드는 서열번호 18 내지 20의 아미노산 서열 중 어느 하나의 아미노산 서열을 갖는 것인 변이형 폴리펩티드.
- 제1항에 있어서, 상기 폴리펩티드의 아미노산을 치환하여 메치오닌에 대한 피드백 조절이 해제된 것인 변이형 폴리펩티드.
- 제4항에 있어서, 상기 아미노산 치환은 29번째 아미노산이 프롤린으로 치환, 114번째 아미노산이 글리신으로 치환, 140번째 아미노산이 세린으로 치환, 또는 상기 3종의 아미노산 치환 중 1종 이상이 조합된 것인 변이형 폴리펩티드.
- 제5항에 있어서, 상기 폴리펩티드는 추가로 112번째 아미노산이 쓰레오닌 또는 히스티딘으로 치환된 것인 변이형 폴리펩티드.
- 제5항 또는 제6항에 있어서, 상기 변이형 폴리펩티드는 서열번호 21 내지 23의 아미노산 서열 중 어느 하나의 아미노산 서열을 갖는 것인 변이형 폴리펩티드.
- 제1항의 변이형 폴리펩티드를 코딩하는 폴리뉴클레오티드.
- 제8항에 있어서, 상기 폴리뉴클레오티드는 서열번호 24 내지 29의 염기서열 중 어느 하나의 염기서열을 갖는 것인 폴리뉴클레오티드.
- 제8항의 폴리뉴클레오티드가 작동 가능하게 연결된 재조합 벡터.
- 제8항의 폴리뉴클레오티드를 포함하는 미생물.
- 제11항에 있어서, 추가로 아세틸-CoA 신테타아제 (acetyl-CoA synthetase)의 활성이 내재적 아세틸-CoA 신테타아제 활성에 비하여 강화되거나, 또는 판토테네이트 키나아제 (pantothenate kinase) 활성이 CoA 축적에 의한 피드백 저해가 해제되도록 변형된 것인 미생물.
- 제11항에 있어서, 추가로 포스포에놀피루브산 카복실라제 암호화 유전자 (ppc), 아스파르테이트 아미노트랜스퍼라제 암호화 유전자 (aspC) 및 아스파르테이트 세미알데히드 데하이드로게나제 암호화 유전자 (asd)로 이루어진 군으로부터 선택되는 하나 이상의 유전자의 카피수가 증가되거나, 또는 상기 유전자의 프로모터가 활성이 증가된 프로모터로 치환되거나 활성이 증가되도록 변이된 미생물.
- 제10항의 재조합 벡터로 형질전환된 미생물.
- 제14항에 있어서, 상기 미생물은 에스케리키아 (Escherichia)속인 미생물.
- 제15항에 있어서, 상기 미생물은 대장균인 미생물.
- 제16항에 있어서, 상기 미생물은 수탁번호 KCCM11145P, KCCM11146P, KCCM11147P, KCCM11228P, KCCM11229P 또는 KCCM11230P인 미생물.
- 제11항 내지 제17항 중 어느 한 항의 미생물을 배양하는 단계; 및
상기 미생물 배양과정에서 생성된 O-아세틸 호모세린을 수득하는 단계를 포함하는 O-아세틸 호모세린의 생산방법.
