Abstract
Description
도 2는 쥐의 폐 섬유아세포인 RFL-6 세포에 대한 중간엽 줄기세포의 배양액(mesenchymal stem cell-conditioned media, MSC-CM)의 증식 효과를 나타낸 것이다. 2A는 MTT assay 로 살아있는 세포 수를 나타낸 것이고, 도 2B는 BrdU uptake assay 로 세포증식을 확인한 것이다. 중간엽 줄기세포의 배양액이 RFL-6 세포에서 농도의존적으로 세포증식을 유도하는 것을 보여주고 있다.
도 3은 MSC-CM이 CSE로 손상된 폐 섬유아 세포를 회복시켜주는 효과를 나타낸 것이다. 3A는 MTT assay 결과로 살아있는 세포 수를 나타낸 것이고, B는 현미경으로 세포 형태를 관찰한 것이고, C는 유세포 분석 (flow cytometry)으로 subG1기에 있는 세포수를 나타낸 것이며, D는 TUNEL (붉은색) 및 annexin V (녹색) 염색 결과를 나타낸 것이고, E는 BrdU uptake assay 로 측정한 세포 증식 결과를 나타낸 것이다. 이상의 결과는 MSC-CM이 농도의존적으로 세포자멸사를 억제하고, CSE에 의해 억제된 세포 증식 능력을 회복시켜주는 것을 보여주고 있다.
도 4는 CSE 노출 및 MSC-CM 처리에 따른 단백질 변화를 웨스턴 블롯으로 확인한 결과이다. 이상의 결과는 MSC-CM이 세포자멸사와 관련된 단백질의 발현을 억제하고, 반대로 세포증식에 관련된 단백질의 발현 및 활성을 증가시킴을 보여주고 있다.
도 5는 MSC-CM의 CSE노출로 억제된 콜라겐 겔 수축 회복효과를 나타낸 것이다. 5B는 5A에서 보인 겔의 면적을 측정한 결과이다.
도 6은 MSC-CM의 CSE노출로 억제된 세포외기질 (extracellular matrix, ECM) 단백질 발현 회복 효과를 나타낸 것이다. 6A와 6B는 각각 세포외기질 단백의 mRNA 발현과 단백의 발현을 측정한 결과이고, 6C는 세포에서의 단백발현을 면역형광법으로 관찰한 결과이다. 6C는 콜라겐 겔 수축을 억제하는 Cox-2와 mPGES-2의 mRNA 발현 결과이다.
도 7은 CSE의 세포 증식 및 조직 재생 억제에 대한 MSC-CM의 회복 효과가 PI3-K/Akt의 활성을 통해 이루어진다는 것을 보여주는 실험 결과들이다. 7A는 MSC-CM에 의한 Akt 인산화가 PI3-K의 활성에 의한다는 것을 보여주며, 7B는 MSC-CM에 의한 세포증식 효과가 PI3-K 억제제에 의해 억제되는 것을 보여주고 있다. 7C는 MSC-CM에 의한 세포외기질 단백의 발현 증가가 역시 PI3-K 억제제에 의해 억제되는 것을 보여주고 있고, 7D는 MSC-CM에 의한 콜라겐 겔 수축능력의 증가가 PI3-K 억제제에 의해 억제됨을 보여주고 있다.
도 8은 담배연기 노출에 따른 폐기종에 대해 중간엽 줄기세포(MSC)의 배양액 (MSC-CM)의 치료효과를 나타낸 것이다. 8A는 폐의 조직학적 소견이며, 8B는 정량적 분석 결과인 MLI (median linear intercept)값을 나타낸 것이다.
도 9는 중간엽 줄기세포의 배양액 (MSC-CM)이 흡연에 의한 폐고혈압을 완화시키는 효과(9A)와, 흡연에 의해 소멸되는 혈관을 재생하는 효과(9B)를 나타낸 것이다.
Claims (10)
- 중간엽 줄기세포의 배양액을 유효성분으로 포함하는 만성폐쇄성 폐질환의 예방 또는 치료용 약학적 조성물.
- 삭제
- 제 1항에 있어서, 상기 배양액은 중간엽 줄기세포를 무혈청 배지에서 3 내지 5 대 계대 배양하여 얻어지는 것을 특징으로 하는 약학적 조성물.
- 제 3항에 있어서, 상기 배지는 DMEM 배지인 것을 특징으로 하는 약학적 조성물.
- 제 1항에 있어서, 상기 중간엽 줄기세포는 골수로부터 유래된 것을 특징으로 하는 약학적 조성물.
- 제 1항에 있어서, 상기 조성물은 주사제 또는 흡입제 형태로 제형화되는 것을 특징으로 하는 약학적 조성물.
- 삭제
- 제 1항에 있어서, 상기 만성폐쇄성 폐질환은 흡연에 의해 발생된 질환인 것을 특징으로 하는 약학적 조성물.
- 제 8항에 있어서, 상기 만성폐쇄성 폐질환은 폐기종 또는 만성기관지염인 것을 특징으로 하는 약학적 조성물.
- 제 1항에 있어서, 상기 조성물은 흡연에 의해 발생된 폐의 압력을 낮추는 것을 특징으로 하는 약학적 조성물.
