- 서열번호 1의 한국산 구제역 바이러스 O/SKR/2000 유래의 재조합 2C 비구조단백질을 동물에 접종하고 그로부터 채취한 면역 세포를 암세포와 융합하여 제조함을 특징으로 하는 하이브리도마 세포주(KCTC 10137 BP).
- 상기 제 4항 기재의 하이브리도마 세포주로부터 발현됨을 특징으로 하는 2C 비구조단백질 특이 단클론항체.
- 구제역 바이러스 유래의 2C 비구조단백질 항원 및 이에 대한 단클론항체를 이용한 구제역 진단방법에 있어서,(1) 수탁번호 KCTC 10137 BP인 하이브리도마 세포주 3F-11에 의해 생성되는 2C 비구조단백질 특이 항원에 대응하는 제 5항의 단클론항체를 코팅 완충용액으로 희석하고 플레이트에 분주한 후 부착시키는 단계;(2) 상기 플레이트에 부착되지 않은 단클론항체를 세척하여 제거하는 단계;(3) 상기 (2)의 플레이트에 서열번호 1의 한국산 구제역 바이러스 O/SKR/2000 유래의 재조합 2C 비구조단백질 항원을 반응시키는 단계;(4) 상기 (3)에서 부착되지 않은 재조합 2C 비구조단백질 항원을 세척하여 제거하는 단계;(5) 상기 (4)의 플레이트에 검사하고자 하는 가검 혈청을 반응시키는 단계;(6) 상기 (5)에서 재조합 2C 비구조단백질 항원에 결합하지 않은 가검 혈청을 세척하여 제거하는 단계;(7) 효소, 방사선물질, 또는 형광물질 등이 부착되어 있고 상기 (5)의 가검 혈청 중의 구제역 바이러스에 대한 항체에 대하여 특이적으로 결합하는 콘쥬게이트를 반응시키는 단계;(8) 상기 콘쥬게이트에 결합된 물질이 효소인 경우, 효소반응을 통한 기질의 발색반응에 의한 흡광도를 측정하고; 형광물질인 경우, 형광광도를 측정하며; 방사선 물질인 경우, 방사선 방출량을 측정함으로써, 가검 혈청 중의 구제역 바이러스 항체가를 측정하는 단계를 포함하는 ELISA법으로 수행함을 특징으로 하는 구제역 진단방법.
- 제 6항에 있어서, 상기 재조합 2C 비구조단백질이 대장균 발현 재조합 2C 비구조단백질임을 특징으로 하는 ELISA에 의한 구제역 진단방법.
- 제 6항에 있어서, 상기 2C 비구조단백질에 대한 단클론항체가 제 5항 기재의 재조합 구제역 2C 비구조단백질 특이 단클론항체임을 특징으로 하는 ELISA에 의한 구제역 진단방법.
