- (a) 조류 배자 생식기(embryonic gonad)에서 분리된 원시생식세포(primordial germ cell, gPGC)를 세포 성장 인자 및 분화 억제 인자가 포함된 배지에서 배양하여 배자생식세포(embryonic germ cell, EG cell) 콜로니를 수득하는 단계;(b) 조류의 섬유아세포 또는 조류의 배자 섬유아세포를 기저세포층(feeder layer)으로 사용하여 상기와 동일한 배지에서 콜로니를 형성할 때까지 수득된 EG 세포를 배양하는 단계; 및(c) EG 세포를 회수하고 상기와 동일한 배지에서 계대배양을 통하여 EG 세포주를 확립하는 단계를 포함하는 조류의 배자생식(EG) 세포주의 제조방법.
- 제 1 항에 있어서,상기 배자 생식기가 배발달단계 14 내지 36인 조류 배자의 생식기인 것을 특징으로 하는 방법.
- 제 2 항에 있어서,상기 배자 생식기가 배발달단계 24 내지 30인 조류 배자의 생식기인 것을 특징으로 하는 방법.
- 제 1 항에 있어서,상기 조류가 칠면조, 닭, 메추라기, 꿩 또는 오리인 것을 특징으로 하는 방법.
- 제 1 항에 있어서,상기 (a) 단계에서 원시생식세포의 배양시 기저층으로서 원시생식기 유래 기저세포(germinal ridge stroma cells; GRSCs)층을 사용하는 방법.
- 제 1 항에 있어서,상기 성장 인자가 SCF(stem cell factor), bFGF(basic fibroblast growth factor), IL-11(interleukin-11), IGF-I(insulin-like growth factor-I) 및 이들의 혼합물로 이루어진 그룹에서 선택된 것임을 특징으로 하는 방법.
- 제 1 항에 있어서,상기 배지가 0.05 내지 500 ng/㎖의 SCF, 0.1 내지 1000 ng/㎖의 bFGF, 0.0004 내지 4 ng/㎖의 IL-11, 0.1 내지 1000 ng/㎖의 IGF-I 및 이들의 혼합물로 이루어진 그룹에서 선택된 성장 인자를 포함하는 것을 특징으로 하는 방법.
- 제 1 항에 있어서,상기 분화 억제 인자가 LIF(leukemia inhibitory factor)인 것을 특징으로 하는 방법.
- 제 8 항에 있어서,LIF가 0.1 내지 1000 units/㎖의 양으로 사용되는 것을 특징으로 하는 방법.
- 제 1 항에 있어서,상기 배지가 포유류 혈청 또는 조류 혈청을 추가로 포함하는 것을 특징으로 하는 방법.
- 제 1 항에 있어서,상기 배지가 피루브산 나트륨, 글루타민, β-머캅토에탄올 및 이들의 혼합물로 이루어진 그룹에서 선택된 보조성분을 추가로 포함하는 것을 특징으로 하는 방법.
- 제 1 항에 있어서,상기 기저세포층이 분열억제 처리되지 않은 것임을 특징으로 하는 방법.

- 제 16 항의 배자생식세포주를 수정란에 주입하는 것을 특징으로 하는 체세포 키메라 및 생식선 키메라의 제조방법.
- 제 20 항에 있어서,배자생식세포주를 상기 수정란의 배반엽 세포층 또는 혈관에 미세주입하는 것을 특징으로 하는 방법.
- 제 21 항에 있어서,배자생식세포주를 배발달단계 X인 수정란의 배반엽세포층내로 주입하는 것을 특징으로 하는 방법.
- 제 21 항에 있어서,배자생식세포주를 배발달단계 13 내지 17인 수정란의 혈관에 주입하는 것을 특징으로 하는 방법.
- 확립된 EG 세포 또는 원시생식세포내로 전기충격법(electroporation) 또는 리포좀을 이용하여 외래 유전자를 전이시키는 방법.
- 외래 유전자를 전이시킨 EG 세포를 항생제가 첨가된 배지에서 계대배양함으로써 안정적으로 외래 유전자가 전이된 세포만을 선발하는 방법.
